Since the effects of ketamine sedation seem to differ between subspecies of baboons, we assessed the endocrine response to an intravenous glucose tolerance test (IVGTT) in 12 hamadryas baboons. The first phase insulin secretion, basal insulin, and glucose levels, as well as the glucose clearance, were significantly lower in sedated baboons as compared to fully awake animals. Glucagon and Cortisol were significantly higher, while growth hormone was lower during ketamine sedation. Papio hamadryas appears to be a promising pre-clinical model for the study of endocrine replacement therapy in insulin-dependent diabetes. However, the data obtained must be interpreted with the knowledge that the anesthetic employed to allow for testing of the animals does have an effect on the parameters described in this report.  相似文献

The development of efficient, reproducible protocols for directed in vitro differentiation of human embryonic stem (hES) cells into insulin-producing β cells will benefit greatly from increased knowledge regarding the spatiotemporal expression profile of key instructive factors involved in human endocrine cell generation. Human fetal pancreases 7 to 21 weeks of gestational age, were collected following consent immediately after pregnancy termination and processed for immunostaining, in situ hybridization, and real-time RT-PCR expression analyses. Islet-like structures appear from approximately week 12 and, unlike the mixed architecture observed in adult islets, fetal islets are initially formed predominantly by aggregated insulin- or glucagon-expressing cells. The period studied (7–22 weeks) coincides with a decrease in the proliferation and an increase in the differentiation of the progenitor cells, the initiation of NGN3 expression, and the appearance of differentiated endocrine cells. The present study provides a detailed characterization of islet formation and expression profiles of key intrinsic and extrinsic factors during human pancreas development. This information is beneficial for the development of efficient protocols that will allow guided in vitro differentiation of hES cells into insulin-producing cells. A review is presented on the use of the technique of solute fluorescence quenching to study the structure and dynamics of proteins. A number of factors are discussed that must be considered in analyzing such data. Among these factors are the efficiency of the quenching process, the relative importance of static quenching, the heterogeneity of the emission, and the tendency of the quencher to interact with the protein.  相似文献

The 1,2-benzisothiazol-3-yl-acetic and -3-yl-butyric acids and their ethyl esters, amides and nitriles are generally active in the split pea stem test, induce an increase in both length and fresh weight of pea internodes, inhibit the development of pea roots, and, with some exceptions (1,2-benzisothiazol-3-yl-butyric amide and nitrile), induce the production of ethylene by pea segments. Moreover they stimulate cell multiplication and raise the degree of hydration of Helianthus tuberosus explants grown in vitro. These activities are often similar or sometimes higher than those of IAA. By contrast, the 1,2-benzisothiazole derivatives having a side chain with an odd number of carbon atoms (-3-yl-carboxylic and propionic acids, amides, ethyl esters and nitriles) are inactive or show a far lower activity.  相似文献
