Chemical gating of gap junction channels; roles of calcium, pH and calmodulin   总被引:11,自引:0,他引:11  
Both Ca(2+) and H(+) play a role in chemical gating of gap junction channels, but, with the possible exception of Cx46 hemichannels, neither of them is likely to induce gating by a direct interaction with connexins. Some evidence suggests that low pH(i) affects gating via an increase in [Ca(2+)](i); in turn, Ca(2+) is likely to induce gating by activation of CaM, which may act directly as a gating particle. The effective concentrations of both Ca(2+) and H(+) vary depending on cell type, type of connexin expressed and procedure employed to increase their cytosolic concentrations; however, pH(i) as high as 7.2 and [Ca(2+)](i) as low as 150 nM or lower have been reported to be effective in some cells. Some data suggest that Ca(2+) and H(+) affect gating by acting synergistically, but other data do not support synergism. Chemical gating follows the activation of a slow gate distinct from the fast V(j)-sensitive gate, and there is evidence that the chemical/slow gate is V(j)-sensitive. At the single channel level, the chemical/slow gate closes the channels slowly and completely, whereas the fast V(j) gate closes the channels rapidly and incompletely. At least three molecular models of channel gating have been proposed, but all of them are mostly based on circumstantial evidence.  相似文献

Seawater electrolysis faces fundamental chemical challenges, such as the suppression of highly detrimental halogen chemistries, which has to be ensured by selective catalyst and suitable operating conditions. In the present study, nanostructured NiFe‐layered double hydroxide and Pt nanoparticles are selected as catalysts for the anode and cathode, respectively. The seawater electrolyzer is tested successfully for 100 h at maximum current densities of 200 mA cm?2 at 1.6 V employing surrogate sea water and compared to fresh water feeds. Different membrane studies are carried out to reveal the cause of the current density drop. During long‐term dynamic tests, under simulated day‐night cycles, an unusual cell power performance recovery effect is uncovered, which is subsequently harnessed in a long‐term diurnal day‐night cycle test. The natural day‐night cycles of the electrolyzer input power can be conceived as a reversible catalyst materials recovery treatment of the device when using photovoltaic electricity sources. To understand the origin of this reversible recovery on a molecular materials level, in situ extended X‐ray absorption fine structure and X‐ray near‐edge region spectra are applied.  相似文献

Tuber tissue of Helianthus tuberosus L. (cv. OB1) contains a low amount of polyamines during dormancy but they are rapidly synthesized when tuber cells are activated in a growth medium and enter a new cell cycle. It was assumed that one of the reasons for this synthesis is that polyamines are necessary for the active conformation and correct functioning of nucleic acids. Complexes were found between spermine, spermidine and putrescine and rRNA, tRNA and an RNA fraction which contains poly(A) RNA and proteins. The amount of RNA-bound polyamines in the parenchyma cells of dormant tubers is dependent on the stage of dormancy and clearly increases (especially putrescine) when cells are activated. There are both tightly-bound and non-tightlybound polyamines. The significance of these bound polyamines is discussed in relation to their stabilizing role on nucleic acids.  相似文献

Although donkey breeding has gained new interest in the past two decades, knowledge about donkey reproduction is still scarce, particularly on jenny pregnancy. The aim of this study was to describe the ultrasonographic and endocrine profiles of the physiological pregnancy in the jenny. The study was performed on 12 pregnancies of 7 Amiata donkeys from Day 10 after ovulation to delivery. Because three pregnancies, respectively at weeks 42, 44, and 45, were considered pathologic and treated pharmacologically, data collected from 2 weeks before diagnosis to the end of pregnancy were removed from the analysis. Average length of the normal pregnancies was 353.4 ± 13.0 days (range, 339–370 days). Timing, dimensions, and development during the first phases of embryonic growth, evaluated using transrectal ultrasound, were similar to that previously described in jennies and mares: first detection of embryonic vesicle was at 11.8 ± 1.3 days of gestation and diameter was 6.5 ± 1.9 mm, loss of spherical shape occurred at 18.5 ± 1.4 days, and embryo and heart beat were first seen at 22.0 ± 1.1 and 25 ± 1.1 days, respectively. The intrauterine growth in the second half of pregnancy, evaluated using the transrectal and transabdominal approach, also showed strong positive correlations, similar to that reported for the mare. The trends of the combined thickness of the utero-placental unit and the echogenicity of the amniotic and allantoic fluids are examples. The diameters (mm) of fetal chest, eye orbit, and aorta increased throughout pregnancy and were 40.6 ± 2.9, 8.7 ± 1.5, and 3.5 ± 0.7, respectively, at week 13, and 190.9 ± 12.0, 21.4 ± 1.5, and 30.6 ± 1.8 at the last evaluation before parturition. In contrast, heart rate decreased as pregnancy progressed. Regression analyses between these parameters and day of gestation were statistically significant (P < 0.001). All fetuses consistently showed some intrauterine activity. Maternal plasma progestagens and estrone sulfate concentrations followed a pattern similar to that seen in mares, although the prepartal progestagen peak was lower in jennies. This study provides a range of ultrasonographic and endocrine values for normal pregnancy in jennies.  相似文献
