Acid extracts of the intermediate pituitary of the holostean fish, Amia calva, were fractionated by gel filtration chromatography and analyzed with radioimmunoassays specific for N-acetylated beta-endorphin and C-terminally amidated alpha-MSH. In these extracts beta-endorphin-related immunoreactive material and alpha-MSH-related immunoreactive material were present in roughly equimolar amounts. The immunoreactive beta-endorphin-sized material was tested for opiate receptor binding activity using a beta-endorphin radioreceptor assay. The results of these studies were negative. The immunoreactive beta-endorphin-sized material was further analyzed by cation exchange chromatography at pH 2.5. Two major and three minor peaks of immunoreactive material were isolated. Peak 5 exhibited a net charge of +7 at pH 2.5 and represented 53% of the total immunoreactivity recovered. Peak 2 with a net charge of +3 at this pH represented 38% of the total immunoreactivity recovered. The minor forms, Peaks 1, 3 and 4, exhibited net charges of +2, +4 and +6, respectively. The apparent molecular weights of Peaks 2 and 5 were determined on a Sephadex G-50 column. Peak 2 had an apparent molecular weight of 2.7 Kd and Peak 5 had an apparent molecular weight of 3.5 Kd. Reverse phase HPLC analysis of Peak 5 indicates that this form of Amia beta-endorphin had chromatographic properties similar to salmon beta-endorphin II. These results would suggest that N-terminal acetylation and C-terminal proteolytic cleavage are important post-translational modifications of the forms of Amia beta-endorphin.  相似文献

Dravet syndrome (DS) is one of the most pharmacoresistant and devastating forms of childhood epilepsy syndromes. Distinct de novo mutations in the SCN1A gene are responsible for over 80% of DS cases. While DS is largely resistant to treatment with existing anti-epileptic drugs, promising results have been obtained in clinical trials with human patients treated with the serotonin agonist fenfluramine as an add-on therapeutic. We developed a zebrafish model of DS using morpholino antisense oligomers (MOs) targeting scn1Lab, the zebrafish ortholog of SCN1A. Zebrafish larvae with an antisense knockdown of scn1Lab (scn1Lab morphants) were characterized by automated behavioral tracking and high-resolution video imaging, in addition to measuring brain activity through local field potential recordings. Our findings reveal that scn1Lab morphants display hyperactivity, convulsive seizure-like behavior, loss of posture, repetitive jerking and a myoclonic seizure-like pattern. The occurrence of spontaneous seizures was confirmed by local field potential recordings of the forebrain, measuring epileptiform discharges. Furthermore, we show that these larvae are remarkably sensitive to hyperthermia, similar to what has been described for mouse models of DS, as well as for human DS patients. Pharmacological evaluation revealed that sodium valproate and fenfluramine significantly reduce epileptiform discharges in scn1Lab morphants. Our findings for this zebrafish model of DS are in accordance with clinical data for human DS patients. To our knowledge, this is the first study demonstrating effective seizure inhibition of fenfluramine in an animal model of Dravet syndrome. Moreover, these results provide a basis for identifying novel analogs with improved activity and significantly milder or no side effects.  相似文献

Antimicrobial photodynamic inhibition (aPI) utilizes radical stress generated from the excitation of a photosensitizer (PS) with light to destroy pathogens. Its use against Trichophyton rubrum, a dermatophytic fungus with increasing incidence and resistance, has not been well characterized. Our aim was to evaluate the mechanism of action of aPI against T. rubrum using curcumin as the PS in both free and nanoparticle (curc-np) form. Nanocarriers stabilize curcumin and allow for enhanced solubility and PS delivery. Curcumin aPI, at optimal conditions of 10 μg/mL of PS with 10 J/cm2 of blue light (417 ± 5 nm), completely inhibited fungal growth (p<0.0001) via induction of reactive oxygen (ROS) and nitrogen species (RNS), which was associated with fungal death by apoptosis. Interestingly, only scavengers of RNS impeded aPI efficacy, suggesting that curcumin acts potently via a nitrosative pathway. The curc-np induced greater NO expression and enhanced apoptosis of fungal cells, highlighting curc-np aPI as a potential treatment for T. rubrum skin infections.  相似文献

The AtGRP5 gene from Arabidopsis thaliana encodes a glycine-rich protein which has a major activity in protoderm-derived cells and is expressed in cells that undergo the first anatomical modifications leading to somatic embryo development. It has been previously demonstrated that its minimum promoter is 316 bp long including the 5′UTR and presents three putative TATA-boxes sequences and several regions that are homologous to previous characterized cis-acting elements. In order to better characterize the AtGRP5 expression and to identify the promoter regions involved in its preferential epidermal expression, in situ hybridization and 5′ promoter deletions were employed. In situ hybridization and GUS expression assays indicate that, besides being present during somatic embryogenesis, AtGRP5 is also expressed during the zygotic embryo development. The sequential 5′ deletions indicate that multiple negative and positive regulatory elements are present in the AtGRP5 promoter and operate in order to confer its distinct expression pattern. A 44-bp region was shown to be essential for the epidermal expression of this gene in leaves, stems, flowers and fruits, and is also responsible for high activity of the AtGRP5 promoter in zygotic embryos. An element responsible for the phloem expression was also identified in a 35-bp region.  相似文献

A signature feature of tetrapod pro-opiomelanocortin (POMC) is the presence of three melantropin (MSH) coding regions (α-MSH, β-MSH, γ-MSH). The MSH duplication events occurred early during the radiation of the jawed vertebrates well over 400 million years ago. However, in at least one order of modern bony fish (subdivision Teleostei; order Salmoniformes; i.e. salmon and trout) the γ-MSH sequence has been deleted from POMC. To determine whether the γ-MSH deletion has occurred in other teleost orders, a POMC cDNA was cloned from the pituitary of the neoteleost Oreochromis mossambicus (order Perciformes). In O. mossambicus POMC, the deletion is more extensive and includes the γ-MSH sequence and most of the joining peptide region. Because the salmoniform and perciform teleosts do not share a direct common ancestor, the γ-MSH deletion event must have occurred early in the evolution of the neoteleost fishes. The post-translational processing of O. mossambicus POMC occurs despite the fact that the proteolytic recognition sequence, (R/K)-Xn-(R/K) where n can be 0, 2, 4, or 6, a common feature in mammalian neuropeptide and polypeptide hormone precursors, is not present at several cleavage sites in O. mossambicus POMC. These observations would indicate that either the prohormone convertases in teleost fish use distinct recognition sequences or vertebrate prohormone convertases are capable of recognizing a greater number of primary sequence motifs around proteolytic cleavage sites.  相似文献

In this study, the geographic and host plant-associated genetic structure of Tomoplagia spp. (Diptera: Tephritidae) was investigated. The study was developed in the rupestrian montane grasslands of the Espinhaço Mountain Range, Brazil, where these flies’ larvae develop within flower heads of Asteraceae. The three species of Tomoplagia investigated have different host spans within the subtribe Vernoniinae: one is strictly monophagous (Tomoplagia grandis Prado, Norrbom & Lewinsohn) and two are oligophagous [Tomoplagia incompleta (Williston) and Tomoplagia bicolor Prado, Norrbom & Lewinsohn]. The genetic variability and differentiation throughout their geographic distribution were investigated at the local scale (sympatric, between different host species in the same locality) and the regional scale (allopatric, on the same or different host species in different localities). Allele richness was greater for populations of the two oligophagous species. Moreover, population genetic structure was low in all species both on the local and regional scales, and the attribution of individuals of the three species to different genetic groups was not strongly associated with either locality or host plant. These results indicate the absence of simple geographic and host-associated genetic structure in these specialized herbivores, suggesting that females of T. bicolor and T. incompleta have low host philopatry, using various host species indiscriminately within their distribution range. Therefore, the use of host plants by fruit flies of the genus Tomoplagia is a plastic character that can shift temporally and spatially among phylogenetically related and chemically similar plants. Contrary to expectations, given the specialized and intimate nature of their host association, the studied Tomoplagia species showed no clear host-associated genetic differentiation; instead, genetic variation presents a mosaic pattern across hosts and localities.  相似文献
