The effects of L-azetidine 2-carboxylic acid on growth and proline metabolism in a proline-requiring auxotroph of Escherichia coli are described. The homologue inhibited growth of the wild type and it, alone, did not substitute effectively for proline as a growth supplement for the mutant. In medium containing 0.05 mM proline, the addition of increasing amounts of homologue progressively inhibited growth of the wild type but stimulated growth of the mutant at homologue: proline ratios of 10 : 1 and 50 : 1. This suggested that the homologue exerted a “sparing effect” on proline in the mutant.The incorporation of L-[U-14C]proline and L-[3H]azetidine 2-carboxylic acid into hot trichloroacetic acid-insoluble material in the mutant was measured. Amino acid analysis of the insoluble material from cells incubated with radiolabeled proline alone revealed that proline was partially degraded and metabolized to other amino acids prior to incorporation into protein. The addition of unlabeled homologue to the incubation medium significantly reduced proline catabolism, suggesting that the homologue exerted a sparing effect on proline in this mutant. In medium containing unlabeled proline and radiolabeled L-azetidine 2-carboxylic acid, the homologuewas incorporated both intact and partially degraded prior to incorporation into protein. BackgroundRecent research suggests that schistosomiasis targets for morbidity control and elimination as a public health problem could benefit from a reanalysis. These analyses would define evidence-based targets that control programs could use to confidently assert that they had controlled or eliminated schistosomiasis as a public health problem. We estimated how low Schistosoma haematobium infection levels diagnosed by urine filtration in school-age children should be decreased so that microhematuria prevalence was at, or below, a “background” level of morbidity.MethodologyData obtained from school-age children in Burkina Faso, Mali, Niger, Tanzania, and Zambia who participated in schistosomiasis monitoring and evaluation cohorts were reanalyzed before and after initiation of preventive chemotherapy. Bayesian models estimated the infection level prevalence probabilities associated with microhematuria thresholds ≤10%, 13%, or 15%.Principal findingsAn infection prevalence of 5% could be a sensible target for urogenital schistosomiasis morbidity control in children as microhematuria prevalence was highly likely to be below 10% in all surveys. Targets of 8% and 11% infection prevalence were highly likely to result in microhematuria levels less than 13% and 15%, respectively. By contrast, measuring heavy-intensity infections only achieves these thresholds at impractically low prevalence levels.Conclusions/significanceA target of 5%, 8%, or 11% urogenital schistosomiasis infection prevalence in school-age children could be used to determine whether a geographic area has controlled or eliminated schistosomiasis as a public health problem depending on the local background threshold of microhematuria. 相似文献
Ciliates assemble basal bodies in great number at many stages of the life-cycle. In order to understand their assembly mechanisms, we screened a library of monoclonal antibodies directed against pericentriolar material. One of these antibodies, CTR210, was used previously to follow steps of this assembly process: in Paraurostyla, new basal bodies appear along a scaffold of linear structures recognized by this antibody. The very unusual behavior of this antigen deserved confirmation in other species. In the present study, we show by immunofluorescence that, in another phylogenetically very distant species, Euplotes, basal bodies are assembled in the same pathway during division. In addition, this antibody recognizes a filamentous ring located at the division furrow and linking many basal body assemblages. By cell fractionation and cytoskeletal extraction, we obtained fractions enriched in basal bodies and associated material. Such fractions still display a high complexity in protein composition. These fractions were used to characterize the main target of the antibody as a doublet of 45 kDa. These results confirm previous results in terms of functionality of the protein recognized by the antibody, but raise new questions in terms of the assignment of the recognized protein to the HSP70 family as hypothesized previously. 相似文献
Since the sixties, the National Indonesian Center for Archaeological Research, in collaboration with the Gajah Mada University, organizes field excavations on open-air sites in the Sangiran dome, one of the most famous places of palaeoanthropological discoveries in Southeast Asia. That project led to the discovery of Homo erectus more or less fragmentary fossils, and to the collection of numerous mammal fossils in open-air localities such as Tanjung, Sendang Busik, Ngrejeng Plupuh, Grogol Plupuh and Bukuran. Most of the bone assemblages from these sites come from the volcanic-sedimentary Kabuh layers (Early Middle Pleistocene) deposited in a fluvial context. The herbivores dominate the assemblages. The best-represented species are large bovids like Bubalus palaeokerabau or Bibos palaesondaicus and smaller cervids like Axis lydekkeri. The carnivore are very rare, only a Pachycrocuta brevirostris in Bukuran. The taphonomical study of the collections, involving several common and also modern methods, is among the first carried out on Javanese sites. The high degree of fragmentation is attested by the dominance of splinters among the fossils. In most sites, teeth and member extremities (autopodial) prevail. Conservation and fragmentation of fossil bones is not the same at all sites. Water action, either chemical or mechanical, is predominant, regarding the origin and the modification of the assemblages. The climato-edaphic traces like oxides and concretions are well represented on the bone surface. Different methods were applied to study transportation and to document the sites contexts in the frame of the ancient riverine paleogeography. Carnivores are almost absent in the material and traces of their action are rare, only in Bukuran and Ngrejeng. The tools are rare, only a bola in Tanjung 82 and chalcedony flakes in Tanjung 63–64. Anthropic influence is only obvious at Ngebung 2, a site in the west part of the dome, supported by fossil observation and correlated to the occurrence of lithic artefacts. This site was excavated during the 1990’s by a French-Indonesian team and it is a site of comparison for our study. Several analytical methods could be applied for the first time on Pleistocene faunal assemblages from Java, for example, the study of the fragmentation, that of fissuration and the characterization of the fluviatile transport undergone by the fossils. 相似文献
The distribution in the human uterus of the enzyme cyclooxygenase, involved in prostaglandin synthesis, was studied using the peroxidase-antiperoxidase immunohistochemical technique. Specific staining was observed only during the luteal phase of the menstrual cycle. It was found that cyclooxygenase was localised in the surface and glandular epithelium of the endometrium but could not be demonstrated in the endometrial stroma, myometrium or blood vessels. The location of specific staining for cyclooxygenase and its variation during the menstrual cycle was the same regardless of the amount of menstrual blood loss. Tissue samples obtained from early pregnant and post-menopausal patients failed to exhibit any specific staining. 相似文献
In grape must, nitrogen is available as a complex mixture of various compounds (ammonium and amino acids). Wine yeasts assimilate these multiple sources in order to suitably fulfil their anabolic requirements during alcoholic fermentation. Nevertheless, the order of uptake and the intracellular fate of these sources are likely to differ between strains and species. Using a two-pronged strategy of isotopic filiation and RNA sequencing, the metabolic network of nitrogen utilization and its regulation in Kluyveromyces marxianus were described, in comparison with those of Saccharomyces cerevisiae. The data highlighted differences in the assimilation of ammonium and arginine between the two species. The data also revealed that the metabolic fate of certain nitrogen sources differed, thereby resulting in the production of various amounts of key wine aroma compounds. This multi-year study examined temperature requirements for dormancy release in physically dormant seeds of the threatened legume Acacia awestoniana (Fabaceae) from Western Australia. Seeds were collected from a single site in three consecutive years and exposed to multiple laboratory-based ‘fire’-related temperature treatments (intensity × duration). Experiments were conducted on seeds freshly collected and after 12 months storage under dry laboratory conditions in order to separate the influence of the maternal environment from post-harvest storage conditions on thresholds for dormancy release. Initial seed viability and non-dormant seed fraction did not differ between seed cohorts but there was a clear effect of storage on seed response: fresh seeds from 2016 demonstrated greater thermal resilience than stored seeds collected in the same year. Equally, there was a strong inter-annual response to treatments from fresh seeds collected in 2016 and 2017 attributed to the influence of the maternal environment during seed development. Seeds collected in 2015 and 2016 and stored for 12 months also demonstrated significant differences in their response to treatments, with 2015 seeds responding more favourably to treatment conditions than those from 2016. Plastic responses to external stimuli provide seeds with a strong bet-hedging capacity and the potential to cope with high levels of environmental heterogeneity, especially a mosaic of fire conditions. Such data provide insight for the management, conservation and restoration of this and similar threatened plant species in fire-prone ecosystems in the face of a rapidly changing climate and expected associated changes in the fire regime.
