We have analysed the influence of the initial pH of the medium and the quantity of aeration provided during the batch fermentation of solutions of d-xylose by the yeast Hansenula polymorpha (34438 ATCC). The initial pH was altered between 3.5 and 6.5 whilst aeration varied between 0.0 and 0.3 vvm. The temperature was kept at 30 °C during all the experiments. Hansenula polymorpha is known to produce high quantities of xylitol and low quantities of ethanol. The most favourable conditions for the growth of xylitol turned out to be: an initial pH of between 4.5 and 5.5 and the aeration provided by the stirring vortex alone. Thus, at an initial pH of 5.5, the maximum specific production rate (μm) was 0.41 h−1, the overall biomass yield (Y x/s G) was 0.12 g g−1, the specific d-xylose-consumption rate (q s ) was 0.075 g g−1 h−1 (for t = 75 h), the specific xylitol-production rate (q Xy ) was 0.31 g g−1 h−1 (for t = 30 h) and the overall yields of ethanol (Y E/s G) and xylitol (Y Xy/s G) were 0.017 and 0.61 g g−1 respectively. Both q s and q Xy decreased during the course of the experiments once the exponential growth phase had finished. Recruitment rates, natal philopatry and the onset of breeding activity are documented for the first time for the order Caprimulgiformes. Of 171 Red‐necked Nightjars Caprimulgus ruficollis ringed as fledglings in southwestern Spain between 2008 and 2011, 31 (18%) were later recovered. Females tended to disperse slightly further (680 m) than males (570 m), and no individual from the study site or nearby locations was ever recovered outside the area where it hatched, suggesting high natal philopatry. Most males (94%) and females (73%) recruited into the breeding population in their first year of life, and only a few individuals were not recovered until their second (10%) or third (7%) year of life. In contrast to most long‐lived birds, nearly all (95.5%) Red‐necked Nightjars started to reproduce in their first potential breeding season.  相似文献

Although variation in the dorsal plumage colour of male European Pied Flycatchers Ficedula hypoleuca has received a great deal of attention, females of the species have been usually considered to be nearly uniformly monochromatic brown. Using reflectance spectrophotometry, we explore the age‐dependent variation of plumage colour in females. We disentangle the within‐ and between‐individual effects of this pattern and show a within‐individual darkening of the mantle colour with age, whereas differences between individuals in structural colour expression may underlie the trend for a more reflective white in the females' breast plumage with advancing age. The darkening of the dorsal plumage as females age reflects the most common pattern of age‐related variation in males in most European populations of the species.  相似文献

The Beijing lineage of Mycobacterium tuberculosis is causing concern due to its global distribution and its involvement in severe outbreaks. Studies focused on this lineage are mainly restricted to geographical settings where its prevalence is high, whereas those in other areas are scarce. In this study, we analyze Beijing isolates in the Mediterranean area, where this lineage is not prevalent and is mainly associated with immigrant cases.  相似文献

The community structure and physiological characteristics of three microbial mat communities in Byers Peninsula (Livingston Island, South Shetland Islands, Antarctica) were compared. One of the mats was located at the edge of a stream and was dominated by diatoms (with a thin basal layer of oscillatorian cyanobacteria), whereas the other two mats, located over moist soil and the bottom of a pond, respectively, were dominated by cyanobacteria throughout their vertical profiles. The predominant xanthophyll was fucoxanthin in the stream mat and myxoxanthophyll in the cyanobacteria-dominated mats. The sheath pigment scytonemin was absent in the stream mat but present in the soil and pond mats. The stream mat showed significantly lower delta13C and higher delta15N values than the other two mats. Consistent with the delta15N values, N2 fixation was negligible in the stream mat. The soil mat was the physiologically most active community. It showed rates of photosynthesis three times higher than in the other mats, and had the highest rates of ammonium uptake, nitrate uptake and N2 fixation. These observations underscore the taxonomic and physiological diversity of microbial mat communities in the maritime Antarctic region.  相似文献

The NASA Artificial Gravity Bed Rest Pilot Study was the first cross-institutional study to investigate the effectiveness of intermittent artificial gravity (AG) as a multi-system countermeasure to bed rest deconditioning in human subjects. Daily treatments by 60 min exposures to short radius centrifugation (SRC) were used to load the longitudinal body axis of eight male human subjects during 21 days of head down tilt bed rest as a means of protecting the bone, muscle, and cardiovascular systems from deconditioning. Data from these treatment subjects were compared with those from seven male human control subjects who were not exposed to SRC loading. This paper reports on implementation issues and lessons learned during the conduct of this complex study.  相似文献

The extracellular concentration of glutamate is highly regulated due to its excitotoxic nature. Failure of glutamate uptake or reversed activation of its transporters contributes to neurodegeneration related to some pathological conditions. We have compared the neurotoxicity of the substrate glutamate uptake inhibitor, l-trans-pyrrolidine-2,4-dicarboxylate (PDC), which promotes glutamate release by heteroexchange, with that of DL-threo-beta-benzyloxyaspartate (DL-TBOA), a non-substrate inhibitor, in cerebellar granule cell cultures. PDC substantially increases the extracellular concentration of glutamate during 30 min exposure and causes neuronal death at high concentrations, while DL-TBOA neurotoxicity is only observed after long-term exposure (8–24 h). During mitochondrial inhibition by 3-nitropropionic acid (3-NP), PDC-induced neuronal death is facilitated, but not that of DL-TBOA. In cultures containing a higher population of astrocytes DL-TBOA-induced increase in glutamate levels is more pronounced, but neuronal death is only triggered in the presence of 3-NP. Results suggest that cerebellar granule neurons are more vulnerable to acute transport-mediated glutamate release than to uptake blockade, which correlates with the extracellular excitatory amino acids levels.  相似文献

The bioinformatics revolution of the last decade has been instrumental in the development of empirical potentials to quantitatively estimate protein interactions for modeling and design. Although computationally efficient, these potentials hide most of the relevant thermodynamics in 5-to-40 parameters that are fitted against a large experimental database. Here, we revisit this longstanding problem and show that a careful consideration of the change in hydrophobicity, electrostatics, and configurational entropy between the folded and unfolded state of aliphatic point mutations predicts 20-30% less false positives and yields more accurate predictions than any published empirical energy function. This significant improvement is achieved with essentially no free parameters, validating past theoretical and experimental efforts to understand the thermodynamics of protein folding. Our first principle analysis strongly suggests that both the solute-solute van der Waals interactions in the folded state and the electrostatics free energy change of exposed aliphatic mutations are almost completely compensated by similar interactions operating in the unfolded ensemble. Not surprisingly, the problem of properly accounting for the solvent contribution to the free energy of polar and charged group mutations, as well as of mutations that disrupt the protein backbone remains open.  相似文献
