The pharmacological activity of rac-ketoprofen and its enantiomers was investigated in vitro using different cellular models. The effect of these compounds on arachidonic acid metabolism was assessed by measuring the inhibition of prostanoid generation under the action of several agonists. Thus, we have evaluated the inhibition of (1) thromboxane B2 synthesis in rabbit platelets and human polymorphonuclear leukocytes (PMNs), (2) prostaglandin E2 synthesis in three cultured cells, namely human umbilical vein endothelial cells (HUVEC), human keratinocytes, and mouse macrophage-like P388D1 cells. The IC50 values found for (+)-(S)-ketoprofen were in the range between 0.1 nM and 0.8 μM, being slightly lower in all models than those found for rac-ketoprofen (0.4 nM–3 μM). On the other hand, (?)-(R)-ketoprofen showed inhibition of cyclooxygenase only at concentrations two or three orders of magnitude higher than those required for the (+)-(S) enantiomer. These results, obtained with cell types of relevance for inflammatory processes and with compounds of high optical purity, demonstrate that the prostanoid biosynthesis inhibition caused by the drug rac-ketoprofen is exclusively due to its dextrorotatory enantiomer. © 1993 Wiley-Liss, Inc.  相似文献

We exposed naive Siberain polecats (Mustela eversmanni) (aged 2, 3, and 4 months) to a swooping stuffed great horned owl (Buho virginianus) and a stuffed badger (Taxidae taxus) mounted on a remote control toy automobile frame. The first introduction to each was harmless, the second was accompanied by a mild aversive stimulus, the third (1 day after attack) was harmless, and the fourth (30 days after attack) was harmless. Alert behavior increased after a single attack by either predator model. Escape responses of naive polecats did not differ between ages when exposed to the badger, but 4 month old polecats reduced their escape times after a single badger attack. When exposed to the swooping owl, naive 4 month old polecats redponded more quickly than the other two age groups, and 3 and 4 month old polecats reduced escape times after a single owl attack. This indicates an innate escape response to the owl model at 4 months of age, and a short-tert ability to remember a single mild aversive encounter with the badger and owl models at 3 or 4 months of age.  相似文献

The light microscopical morphology of the dog pineal gland from the first postnatal day to maturity is described. In the first postnatal week, the pineal parenchyma shows immature cells and many mitotic figures. In this week, pigmented cells are observed for the first time, both in the pineal gland and in extrapineal nodules. Throughout the second week, the pineal parenchyma shows a cordonal pattern that disappears progressively in the following stages. From the 20-30th day onward, it is feasible to discern the cell types characteristic for the adult pineal gland. In the adult animals, the length of the pineal gland axes almost quadruplies that of the pineal gland in neonatal stages. The light microscopical features of the adult dog pineal gland are also described.  相似文献

Light- and electron-microscopic features of pigmented cells in the cat pineal gland are described. These cells were observed throughout postnatal life from the second postnatal day to the oldest cats studied (up to 13 years old). No apparent relationship was observed among the amount of pigment and the animal age or sex. Pigmented cells showed a preferential localization at the ventral surface of the pineal gland near its distal end. The pineal pigment was histochemically identified as melanin. Pineal pigment granules showed ultrastructural features similar to melanocyte melanin granules.  相似文献

The cGI PDE in particulate fractions of differentiated adipocytes (but not control 3T3-L1 fibroblasts) was cross-reactive with a polyclonal antibody raised against the bovine adipose cGI PDE. The 3T3-L1 adipocyte cGI PDE is a 135 kDa protein which is phosphorylated in 32P-labeled cells in response to beta-agonist or insulin. These results indicate that the 3T3-L1 cGI PDE is similar in structure and hormonal regulation to the analogous enzyme in the rat adipocyte.  相似文献

1. A soluble beta-glucosidase from sheep liver has been isolated and purified 114-fold by conventional enzyme fractionation procedures. The specific activity of the purified enzyme was 5910 mU/mg of protein. 2. The enzyme has a broad specificity and hydrolyzes the p-nitrophenyl derivatives of beta-D-glucose, beta-D-galactose, beta-D-fucose, beta-D-xylose and alpha-L-arabinose. The best Vmax/Km value corresponds to the beta-glucosidase activity. 3. The enzyme has a pH optimum between 4.5-5.5 for all the activities, and mol. wt 95,000. 4. A variety of chemicals was tested as possible activators or inhibitors. 5. The enzyme is strongly inhibited by aldono 1-5 lactones and DMDP. 6. The kinetic evidences suggest a substrate activation model and the existence of two active sites (a "gluco-fuco" site and a "galacto" site). 7. The activation energies were calculated from beta-glucosidase and beta-galactosidase activities.  相似文献
