Estrogen synthesized in the brain itself by the action of cytochrome P450 aromatase (P450arom) is known to have permanent organizing effects on the developing CNS. In fish, estrogen upregulates the predominant brain isoform (P450aromB), implying that xenoestrogens (XE) could act as neurodevelopmental toxicants by altering P450aromB. To test this hypothesis, zebrafish embryos were exposed to 17beta-estradiol (E(2)), diethylstilbestrol (DES, a potent agonist), and bisphenol A (BPA, a weak agonist). RT-PCR/Southern transfer analysis showed that E(2) (0.01-10 microM) upregulated P450aromB in a dose-response manner. Contaminated groundwater plumes have formed on the Massachusetts Military Reservation (MMR), a Superfund site on Cape Cod, Massachusetts, as a result of chemical waste disposal. The plumes are of concern to the local people who rely on groundwater as a drinking water source. We used the freshwater turtle as a sentinel species to monitor the reproductive effects of exposure and, by inference, the potential for impact on human health. Our observations of male Chrysemys picta field-trapped from Moody Pond (an impacted site) and Washburn Pond (a reference site) on Cape Cod extended and supported prior observations of reproductive deficits. Morphometric comparison of precloacal length (PCL), which is a sexually dimorphic trait in the turtle, showed that Moody Pond males had a significantly longer PCL than Washburn Pond males. Moody Pond turtles showed reduced testicular weight, which was associated with significantly smaller seminiferous tubule diameter. Epididymal sperm counts were also markedly reduced in Moody Pond animals compared to Washburn Pond animals. Testicular histology and gonial proliferation, as determined by PCNA, were similar in both male populations, while the Moody Pond males had significantly higher germ cell apoptosis than the animals in Washburn Pond. These results suggest that a low-level mixture of xenobiotic contaminants impairs the reproductive functions of turtles exposed to the impacted site but not to the reference site environment.  相似文献

Decisions by uncommitted cells to differentiate down one lineage pathway or another is fundamental to developmental biology. In the immune system, lymphocyte precursors commit to T- or B-cell lineages and T-cell precursors to CD4 or CD8 independently of foreign antigen. T and B cells must also decide whether or not to respond to antigen and when a response is initiated, what sort of response to make such as the type of antibody, CD4 or CD8, and CD4 Th1 or Th2. The two basic mechanisms for these decision-making processes are selection and instruction. Selection depends on prior stochastic production of precommitted cells, which are then selected to respond by an appropriate signal; for example, CD8 and CD4 responses selected by peptide presented in association with major histocompatibility complex class I or II. In contrast, instruction occurs when an uncommitted precursor embarks upon a differentiation pathway in response to a particular set of signals; for example, Th1 and Th2 lineage commitment. In this paper, the signals that determine Th1 and Th2 differentiation are examined with a mathematical model and shown to act as a bistable switch permitting either Tbet or Gata3 to be expressed in an individual cell but not both. The model is used to show how the Tbet Gata3 network within an individual cell interacts with cytokine signals between cells and suggests how Th1 and Th2 lineage commitment can become irreversible. These considerations provide an example of how mathematical models can be used to gain a better understanding of lymphocyte differentiation in an immune response.  相似文献

Due to a cystic mode of spermatogenesis and zonal testes, the spiny dogfish shark (Squalus acanthias) and salamander (Necturus maculosus) are advantageous for studying the relationship between steroidogenesis and germ cell development. Direct analysis of steroidogenic enzymes and steroid receptors in staged tissue samples, together with light and electron microscopic observation, suggests that androgens and estrogens produced by Sertoli cells or Leydig cells immediately before or after spermiation serve as intratesticular regulators of events specific to premeiotic stages of spermatogenesis. These correlative data are consistent with limited observations in mammalian testis and provide a conceptual framework for direct testing of functional interactions between spermatogenesis and steroidogenesis in future studies.  相似文献

Aromatase activity of goldfish brain synaptosomes was not suppressed by a mouse anti-human placental aromatase cytochrome P-450 monoclonal antibody. A rabbit antiserum to human placental aromatase cytochrome P-450 did not show a significant suppression of the goldfish brain activity when compared to the placental aromatase. Histology of each of the five segments of the oviduct of the female turtle Chrysemys picta was described for successive intervals throughout their annual cycle. Uterine and glandular segments showed marked seasonal variations in the extent and content of the submucosal and epithelial glands. Submucosal glands were most prominent in preovulatory and postovulatory animals (May to June), regressing in late summer (oviposited animals) and recrudescing the following spring. These changes correlated with variations in the muscularis layer, the number of uterine epithelial blebs, oviductal vascularity, and the presence of eosinophils in cervical segment cross-sections. These cyclic seasonal changes are discussed in relationship to reported seasonal changes in gonadal steroids in this species. Hormonal control was corroborated by oviductal response to estradiol-17β injected (1 mg/kg daily for 2 weeks) into mature, reproductively inactive (winter) animals. This treatment induced increases in glandular activity, vascularity, and distribution of eosinophils comparable to those of reproductively active (summer) animals.  相似文献

In Necturus maculosus the organization of the interstitial tissue varies according to the stage of spermatogenesis. Leydig cells at various stages of differentiation and myoid cells are always present in this tissue. The Leydig cells are undifferentiated at all phases of germ cell activity and only hypertrophy following spermiation and degeneration of Sertoli cells. These Leydig cells are structurally analogous to mammalian Leydig cells. They do not form part of the lamina propria of the seminiferous lobules and hence cannot be referred to as lobule-boundary cells previously described in the urodele testis (Lofts, '74). When the Leydig cells hypertrophy, numerous unmyelinated axons appear in the interstitial tissue. These axons, often devoid of Schwann-cell cytoplasm, occur in close proximity to Leydig cells. Because the levels of both Substance P and neurotensin increased in the testis of Necturus maculosus as Leydig cells differentiated, we concluded that these neural elements may regulate Leydig-cell function locally, through the release of neuropeptides.  相似文献
