1. The isolation and purification of a putative apolipoprotein B-100 in the plasma of the freshwater turtle Chrysemys picta is described. 2. The protein was purified through differential ultracentrifugation and subsequent Sepharose 6B column chromatography. 3. The molecular weight of the protein determined by electrophoresis was approximately 350 kDa. 4. An antibody to chicken apolipoprotein B-100 specifically recognizes this 350 kDa protein in Western blots, suggesting its identity with apolipoprotein B-100. 5. An antibody to the putative Chrysemys apolipoprotein B-100-like protein was developed and used in an ELISA to quantitate protein levels in plasma. 6. Acute estrogen treatment increased levels of apolipoprotein B-100 (7.64 +/- 0.79 mg/ml plasma) over that of control animals (5.07 +/- 1.74 mg/ml plasma). 7. In contrast, chronic estrogen treatment reduced apolipoprotein B-100 significantly to 2.94 +/- 0.53 mg/ml plasma (P < 0.05).  相似文献

Highly specific antisera for 11-keto- and 11 beta-hydroxytestosterone have been raised in sheep. Assay systems for the simultaneous measurement of 11-ketotestosterone, 11 beta-hydroxytestosterone, testosterone, progesterone and estradiol-17 beta were validated for Ictalurus nebulosus plasma and Carassius auratus serum. In males of both species 11-ketotestosterone and testosterone were the major steroids detected. In females, testosterone and estradiol-17 beta were the predominant steroids measured. Data from samples taken at different stages of the annual cycle suggest that seasonal fluctuations in gonadal steroid secretion occur in I. nebulosus and C. auratus.  相似文献

Where aromatase and estrogen receptors are co-localized in brain and pituitary, estrogen functions as a parahormone, and estrogen levels which determine the occurrence or magnitude of a response are those in close proximity to targets. Teleost fish, a vertebrate group characterized by exceptionally high aromatase in neuroendocrine tissues, are technically advantageous animal models for studying the cellular location of aromatase, natural changes correlated with seasonal reproductive cycles, substrate-dependence of the reaction, steroid induction of enzyme activity, and possible non-genomic actions of estrogen on cultured neurons. In addition, characterization of steroid receptors reveals that the androgen receptor, like aromatase, is present in unusually high concentrations (10- to 100-fold higher than in mammalian brain). Since androgen receptors and aromatase both utilize testosterone as a ligand, their high abundance in teleost brain may be the consequence of a functional interdependence during evolution, although the primary causal factor is unknown. These studies illustrate the usefulness of unconventional species and a comparative approach for obtaining new insights into brain-steroid interactions.  相似文献

An acid-acetone extract prepared from ovaries of the skate, Raja erinacea, contained a weakly crossreacting molecule when tested in a pig relaxin radioimmunoassay. The material was isolated and purified to homogeneity by ion exchange chromatography, molecular exclusion chromatography, and HPLC. Analytical tests proved the molecule to consist of two chains and to have a molecular weight of 7,500. Sequence analyses of the A and B chains yielded the following sequence: Glu-Glu-Lys-Met-Gly-Phe-Ala-Lys-Lys-Cys-Cys-Ala-Ile-Gly-Cys-Ser-Thr-Glu- Asp-Phe-Arg-Met-Val-Cys and Arg-Pro-Asn-Trp-Glu-Glu-Arg-Ser-Arg-Leu-Cys-Gly-Arg-Asp-Leu-Ile-Arg-Ala- Phe- Ile-Tyr-Leu-Cys-Gly-Gly-Thr-Arg-Trp-Thr-Arg-Leu-Pro-Asn-Phe-Gly-Asn-Tyr- Pro-Ile-Met respectively. Skate relaxin has 0.2% of the activity of B29 pig relaxin in the symphysis pubis assay and 0.5% in the mouse uterine muscle strip contraction inhibition assay.  相似文献
