Short‐finned pilot whales (Globicephala macrorhynchus) have complex vocal repertoires that include calls with two time‐frequency contours known as two‐component calls. We attached digital acoustic recording tags (DTAGs) to 23 short‐finned pilot whales off Cape Hatteras, North Carolina, and assessed the similarity of two‐component calls within and among tags. Two‐component calls made up <3% of the total number of calls on 19 of the 23 tag records. For the remaining four tags, two‐component calls comprised 9%, 23%, 24%, and 57% of the total calls recorded. Measurements of six acoustic parameters for both the low and high frequency components of all two‐component calls from the five tags were compared using a generalized linear model. There were significant differences in the acoustic parameters of two‐component calls between tags, verifying that acoustic parameters were more similar for two‐component calls recorded on the same tag than for calls between tags. Spectrograms of all two‐component calls from the five tags were visually graded and independently categorized by five observers. A test of inter‐rater reliability showed substantial agreement, suggesting that each tag contained a predominant two‐component call type that was not shared across tags. 相似文献

Protein-protein interactions define many important molecular and cellular processes in prokaryotic and eukaryotic biology. In trying to delineate the contact between two proteins, the yeast two-hybrid assay has emerged as a powerful technique. Complementing the yeast two-hybrid assay are in vitro techniques (e.g. GST-fusion-protein chromatography) that can also yield information on protein-protein associations. However, unambiguous functional significance to these interactions is best supported through a finding of colocalization of two proteins inside cells. In instances where two proteins interact in vitro but have divergent localizations within cells one needs to reconsider the biological importance of the former finding. Here, we present evidence for different subcellular locations of HTLV-I Tax and the Int-6 protein. We suggest a reexploration of the functional significance between Tax and Int-6 in cellular transformation. 相似文献

The ultimate goal of the Mexican gray wolf Canis lupus baileyi captive management program is reintroduction of healthy individuals into wild habitats. To this end, zoo population managers work to provide not only for the physical well-being but also for the genetic health of these animals. However, the very limited genetic founder base, exacerbated by breeding within three distinct lineages, resulted in very high coefficients of inbreeding. Because support for measurable levels of inbreeding depression in the captive wolf population, as defined by reductions in common fitness measures such as juvenile survival or reproductive success, has been weak, we investigated the potential effects on male reproductive capacity. We analyzed semen samples from wolves from all three lineages and compared them with samples from subsequent lineage crosses and from generic gray wolves. We not only found a significant effect of inbreeding on sperm quality but we related both inbreeding and sperm quality to reproductive success. Samples from male offspring of lineage crosses, with inbreeding coefficients of zero were similar in quality to those from generic gray wolves. However, samples from a limited number of offspring from back-crosses were of extremely poor quality. Although it is reassuring that sperm quality was so much improved in male offspring of lineage crosses, the concomitant reduction in inbreeding coefficient does not eliminate the potentially deleterious alleles. Our results demonstrate that sperm quality is an important indicator of fertility and reproductive success in Mexican wolves. In addition, our data lend further support to the presence of inbreeding depression in this taxon. 相似文献

Previous studies indicate that ATP formation by the electron transport chain is impaired in sepsis. However, it is not known whether sepsis affects the mitochondrial ATP transport system. We hypothesized that sepsis inactivates the mitochondrial creatine kinase (MtCK)-high energy phosphate transport system. To examine this issue, we assessed the effects of endotoxin administration on mitochondrial membrane-bound creatine kinase, an important trans-mitochondrial ATP transport system. Diaphragms and hearts were isolated from control (n = 12) and endotoxin-treated (8 mg.kg(-1).day(-1); n = 13) rats after pentobarbital anesthesia. We isolated mitochondria using techniques that allow evaluation of the functional coupling of mitochondrial creatine kinase MtCK activity to oxidative phosphorylation. MtCK functional activity was established by 1) determining ATP/creatine-stimulated oxygen consumption and 2) assessing total creatine kinase activity in mitochondria using an enzyme-linked assay. We examined MtCK protein content using Western blots. Endotoxin markedly reduced diaphragm and cardiac MtCK activity, as determined both by ATP/creatine-stimulated oxygen consumption and by the enzyme-linked assay (e.g., ATP/creatine-stimulated mitochondrial respiration was 173.8 +/- 7.3, 60.5 +/- 9.3, 210.7 +/- 18.9, was 67.9 +/- 7.3 natoms O.min(-1).mg(-1) in diaphragm control, diaphragm septic, cardiac control, and cardiac septic samples, respectively; P < 0.001 for each tissue comparison). Endotoxin also reduced diaphragm and cardiac MtCK protein levels (e.g., protein levels declined by 39.5% in diaphragm mitochondria and by 44.2% in cardiac mitochondria; P < 0.001 and P = 0.009, respectively, comparing sepsis to control conditions). Our data indicate that endotoxin markedly impairs the MtCK-ATP transporter system; this phenomenon may have significant effects on diaphragm and cardiac function. 相似文献

Isoenzymes of sphingomyelinase have been resolved by isoelectric focusing. The two major species (I and II) in human liver have distinct isoelectric points, pH optima and Km values. mRNA extracted from polysomes of KB cells at late stages of productive infection with adenovirus type 2 was translated in a cell-free system derived from Krebs II ascites cells. Two of the polypeptides obtained in this reaction corresponded to the adenovirus core protein V and the precursor to core protein VII. Peroxynitrite may be generated in and around muscles in several pathophysiological conditions (e.g., sepsis) and may induce muscle dysfunction in these disease states. The effect of peroxynitrite on muscle force generation has not been directly assessed. The purpose of the present study was to assess the effects of peroxynitrite administration on diaphragmatic force-generating capacity in 1) intact diaphragm muscle fiber bundles (to model the effects produced by exposure of muscles to extracellular peroxynitrite) and 2) single skinned diaphragm muscle fibers (to model the effects of intracellular peroxynitrite on contractile protein function) by examining the effects of both peroxynitrite and a peroxynitrite-generating solution, 3-morpholinosydnonimine, on force vs. pCa characteristics. In intact diaphragm preparations, peroxynitrite reduced diaphragm force generation and increased muscle levels of 4-hydroxynonenal (an index of lipid peroxidation). In skinned fibers, both peroxynitrite and 3-morpholinosydnonimine reduced maximum calcium-activated force. These data indicate that peroxynitrite is capable of producing significant diaphragmatic contractile dysfunction. We speculate that peroxynitrite-mediated alterations may be responsible for much of the muscle dysfunction seen in pathophysiological conditions such as sepsis. 相似文献

Hard coral (Scleractinia and Milleporina) cover data were examined from 37 sites surveyed annually from 1996 to 2003 in the
Florida reef tract, USA. Analyses of species numbers and total cover showed that site-to-site differences were generally very
much greater than differences among times within sites. There were no significant differences among different geographical
areas within the reef tract (Upper, Middle and Lower Keys). Large-scale changes documented included a reduction in species
numbers and total cover on both deep and shallow offshore reefs between 1997 and 1999 followed by no recovery in cover, and
only scant evidence of any recovery in species numbers by 2003. These changes coincided with bleaching events in 1997 and
1998, and the passage of Hurricane Georges through the Lower Keys in 1998. The lack of recovery among offshore reefs suggests
that they were no longer resilient. Multivariate analyses revealed that some sites showed relatively little temporal variation
in community composition, essentially random in direction, while others showed relatively large year-on-year changes. There
was little evidence of any major region-wide changes affecting assemblage composition, or of any events that had impacted
all of the sampling sites in any single year. Instead, different sites exhibited differing patterns of temporal variation,
with certain sites displaying greater variation than others. Changes in community composition at some sites are interpreted
in the light of knowledge of events at those sites and the relative sensitivities of species to various stressors, such as
changes in cover of Acropora palmata and Millepora complanata at Sand Key following the bleaching events and hurricane in 1998, and declines in Montastraea annularis at Smith Shoal following a harmful algal bloom in 2002. For most sites, however, it is impossible to determine the causes
