In landings from a flight phase the mass centre of an athlete experiences rapid decelerations. This study investigated the extent to which co-contraction is beneficial or necessary in drop landings, using both experimental data and computer simulations. High speed video and force recordings were made of an elite martial artist performing drop landings onto a force plate from heights of 1.2, 1.5 and 1.8 m. Matching simulations of these landings were produced using a planar 8-segment torque-driven subject-specific computer simulation model. It was found that there was substantial co-activation of joint flexor and extensor torques at touchdown in all three landings. Optimisations were carried out to determine whether landings could be effected without any co-contraction at touchdown. The model was not capable of landing from higher than 1.05 m with no initial flexor or extensor activations. Due to the force–velocity properties of muscle, co-contraction with net zero joint torque at touchdown leads to increased extensor torque and decreased flexor torque as joint flexion velocity increases. The same considerations apply in any activity where rapid changes in net joint torque are required, as for example in jumps from a running approach.  相似文献

The Neotropics contains half of remaining rainforests and Earth's largest reservoir of amphibian biodiversity. However, determinants of Neotropical biodiversity (i.e., vicariance, dispersals, extinctions, and radiations) earlier than the Quaternary are largely unstudied. Using a novel method of ancestral area reconstruction and relaxed Bayesian clock analyses, we reconstructed the biogeography of the poison frog clade (Dendrobatidae). We rejected an Amazonian center-of-origin in favor of a complex connectivity model expanding over the Neotropics. We inferred 14 dispersals into and 18 out of Amazonia to adjacent regions; the Andes were the major source of dispersals into Amazonia. We found three episodes of lineage dispersal with two interleaved periods of vicariant events between South and Central America. During the late Miocene, Amazonian, and Central American-Chocoan lineages significantly increased their diversity compared to the Andean and Guianan-Venezuelan-Brazilian Shield counterparts. Significant percentage of dendrobatid diversity in Amazonia and Chocó resulted from repeated immigrations, with radiations at <10.0 million years ago (MYA), rather than in situ diversification. In contrast, the Andes, Venezuelan Highlands, and Guiana Shield have undergone extended in situ diversification at near constant rate since the Oligocene. The effects of Miocene paleogeographic events on Neotropical diversification dynamics provided the framework under which Quaternary patterns of endemism evolved.  相似文献

Gene silencing via promoter CpG island hypermethylation offers tumor cells growth advantages. This epigenetic event is pharmacologically reversible, and uncovering a unique set of methylation-silenced genes in tumor cells can bring a new avenue to cancer treatment. However, high-throughput tools capable of surveying the methylation status of multiple gene promoters are needed for this discovery process. Herein we describe an oligonucleotide-based microarray technique that is both versatile and sensitive in revealing hypermethylation in defined regions of the genome. DNA samples are bisulfite-treated and PCR-amplified to distinguish CpG dinucleotides that are methylated from those that are not. Fluorescently labeled PCR products are hybridized to arrayed oligonucleotides that can discriminate between methylated and unmethylated alleles in regions of interest. Using this technique, two clinical subtypes of non-Hodgkin's lymphomas, mantle cell lymphoma, and grades I/II follicular lymphoma, were further separated based on the differential methylation profiles of several gene promoters. Work is underway in our laboratory to extend the interrogation power of this microarray system in multiple candidate genes. This novel tool, therefore, holds promise to monitor the outcome of various epigenetic therapies on cancer patients.  相似文献

The oxygen-radical absorbance capacity (ORAC) assay has become a standard method to quantify the antioxidative properties of phytonutrients in fruit and vegetable extracts. However, it is usually not possible to determine directly the contribution of specific phytonutrients to the total ORAC value. Separation of the components in the plant extracts by HPLC followed by ORAC analyses of the column fractions might permit the determination of free radical-scavenging profiles. The accurate determination of ORAC values may require 1 to 2 h/sample. Considering the number of samples that would be generated by a single HPLC separation, a device was constructed which permits up to 45 simultaneous ORAC analyses. Varying degrees of automation were included in the design. Furthermore, since the assay has a Q(10) for peroxyl radical-scavenging of about 3, elevation of the assay temperature from the standard 37 to 47 degrees C significantly reduced the assay times. Relatively simple modifications would allow the apparatus to be used in a variety of time-dependent fluorescence and absorbance assays.  相似文献

Growth and attachment rates of Thermothrix thiopara on calcite and pyrite were quantitated in a thiosulfate‐limited chemostat and in the thermal spring where the organism is found in nature. Surface growth rates were quantitated by using the surface colonization and exponential growth equations. These two models were compared as means of determining surface growth rates. In the chemostat, T. thiopara cells colonizing calcite and pyrite surfaces grew at approximately one‐third the rate of suspended cells. However, T. thiopara attached to pyrite faster than to calcite. In the thermal spring, growth and attachment rates were equal on calcite and pyrite. It was concluded that the exponential growth equation overestimates in‐situ surface growth rates and that T. thiopara grows more slowly when colonizing mineral surfaces than when growing in suspension. Lower growth rates on surfaces may be due to a reduced cell surface area for nutrient uptake or an increased specific maintenance rate.  相似文献
