Central administration of corticotropin releasing factor (CRF) in rats produces pituitary-adrenal activation and a variety of "anxiogenic-like" effects. The present study was designed to explore the contribution of the peripheral pituitary-adrenocortical axis in mediating these CRF responses. Intraventricularly administered CRF produced suppression of responding in the conflict test and a marked locomotor activation. Neither behavioral effect was altered by the prior administration of dexamethasone in a dose that blocked pituitary-adrenal activation to CRF. These results support the hypothesis that behavioral effects of CRF are mediated by its action at central sites and not via an action on the pituitary-adrenocortical system.  相似文献

Phorbol 12,13-dibutyrate (PDBu) increased the production of 3,4-dihydroxyphenylalanine (DOPA) in the superior cervical ganglion of the rat. This effect occurred without a detectable lag and persisted for at least 90 min of incubation. The action of PDBu was half-maximal at a concentration of approximately 0.1 microM; at high concentrations, PDBu produced about a twofold increase in DOPA accumulation. PDBu increased DOPA production in decentralized ganglia and in ganglia incubated in a Ca2+-free medium. The action of PDBu was additive with the actions of dimethylphenylpiperazinium, muscarine, and 8-Br-cyclic AMP, all of which also increase DOPA accumulation, and was not inhibited by the cholinergic antagonists hexamethonium (3 mM) and atropine (6 microM). Finally, PDBu did not increase the content of cyclic AMP in the ganglion. Thus, the action of PDBu does not appear to be mediated by the release of neurotransmitters from preganglionic nerve terminals, by the stimulation of cholinergic receptors in the ganglion, or by an increase in ganglionic cyclic AMP. PDBu also increased the incorporation of 32Pi into tyrosine hydroxylase. PDBu activates protein kinase C, which in turn may phosphorylate tyrosine hydroxylase and increase the rate of DOPA synthesis in the ganglion.  相似文献

A sequential study of electrolyte leakage from roots inoculated with Phytophthora cinnamomi demonstrated changes in leakage in response to infection. These changes may be characterised in terms of susceptibility and resistance. Field resistant species showed two types of response (i) rapid leakage 2—4 h after inoculation, such as in Eucalyptus calophylla, E. maculata and Gahnia radula, (ii) no significant increase in leakage with infection, for example in cereals and in juncus bufonius. Susceptible species, such as Xanthorrhoea australis, E. sieberi and E. marginata showed slow but continually increasing leakage after inoculation, and usually lost significantly more electrolyte than field resistant species. Both electrolyte leakage and susceptibility of two Eucalyptus species varied with root temperature. Roots of both the susceptible and field resistant Eucalyptus species grown at 14°C showed similar leakage patterns to those grown at 24°C although no lesions formed at 14°C. Both amount of leakage and lesion length increased at 28°C. E. marginata (susceptible) lost significantly greater quantities of electrolyte than E. calophylla at each temperature tested. Leakage from artificially wounded roots did not vary with either temperature or with host susceptibility. No significant changes in conductivity were recorded with saprophytic colonization of roots, or with incubation in either low molecular weight culture filtrate or β-glucan solutions. Vacuum infiltration with the culture filtrate or the β-glucan slightly increased initial electrolyte loss in comparison with that of the controls. The increased leakage from infected roots may be toxin mediated or due to enzymie degradation of host plasma membranes, and occurred within the region of the limited lesion in the case of field resistant species, compared with the larger zone of extending necrosis characteristic of susceptible species.  相似文献

A molecular probe was prepared with specificity for the human cellular homologue of transforming sequences represented within the McDonough strain of feline sarcoma virus (v-fms). By analysis of a series of mouse-human somatic cell hybrids containing variable complements of human chromosomes it was possible to assign this human oncogene, designated c-fms, to chromosome 5. Regional localization of c-fms to band q34 on chromosome 5 was accomplished by analysis of Chinese hamster-human cell hybrids containing as their only human components, terminal and interstitial deleted forms of chromosome 5. The localization of c-fms to chromosome 5 (q34) is of interest in view of reports of a specific, apparently interstitial, deletion involving approximately two thirds of the q arm of chromosome 5 in acute myelogenous leukemia cells.  相似文献

  1. At a so-called natural anaerobiosis during the first 48 hours of germination the concentration of ethanol in pea tissues increases (according to the cultivation conditions) up to 40 μmol per gram fresh weight.
  2. In a nitrogen atmosphere the content of ethanol in pea seedlings increases as well, and after a 90 hour incubation in N2 it can reach even 100 μmol ethanol per gram fresh weight. In older plants the content increases the most markedly in cotyledons, but considerable amounts were revealed also in stems and roots. Its increase in vegetative organs of plants cultivated both in light and darkness is more or less identical. Ethylalcohol can be formed by the vegetative organs themselves, as proved by the increase of this metabolite in plants deprived of reserve organs; in addition, however, it is evidently transported into them from reserve parts. Ethanol formed under anaerobiosis is catabolyzed after transferring plants to the air.
