Respiratory control, lost by rat-liver mitochondria during storage at 0°, can be restored if the mitochondria are incubated under energy-generating conditions for 2 hr at 25°C. The extent of restoration is virtually complete in 1 day old mitochondria but diminishes with aging thereafter. Endogenous auxins and cytokinins were quantitated in 24 axenic microalgal strains from the Chlorophyceae, Trebouxiophyceae, Ulvophyceae, and Charophyceae. These strains were in an exponential growth phase, being harvested on day 4. Acutodesmus acuminatus Mosonmagyaróvár Algal Culture Collection‐41 (MACC) produced the highest biomass and Chlorococcum ellipsoideum MACC‐712 the lowest biomass. The auxins, indole‐3‐acetic acid (IAA) and indole‐3‐acetamide (IAM) were present in all microalgal strains. No other auxin conjugates were detected. IAA and IAM concentrations varied greatly, ranging from 0.50 to 71.49 nmol IAA · g?1 DW and 0.18 to 99.83 nmol IAM · g?1 DW, respectively. In 19 strains, IAA occurred in higher concentrations than IAM. Nineteen cytokinins were identified in the microalgal strains. Total cytokinin concentrations varied, ranging from 0.29 nmol · g?1 DW in Klebsormidium flaccidum MACC‐692 to 21.40 nmol · g?1 DW in Stigeoclonium nanum MACC‐790. The general trend was that cis‐zeatin types were the predominant cytokinins; isopentenyladenine‐type cytokinins were present in moderate concentrations, while low levels of trans‐zeatin‐type and very low levels of dihydrozeatin‐type cytokinins were detected. Ribotides were generally the main cytokinin conjugate forms present with the cytokinin free bases and ribosides present in similar but moderate levels. The levels of O‐glucosides were low. Only one N‐glucoside was detected, being present in nine strains in very low concentrations. In 15 strains, the auxin content was 2‐ to 4‐fold higher than the cytokinin content. 相似文献

Diabetes induces oxidative stress in aged human and rat, although daily supplementation of vitamins C and E (VCE) can be beneficial to aged diabetic rats by reducing free radical production. The aim of the present study was to evaluate whether dietary VCE supplementation relieves oxidative stress in streptozotocin (STZ)-induced diabetic in aged rats. Thirty aged rats were randomly divided into three groups. The first group was used as a control. The second group was made diabetic using a single dose of intraperitoneal STZ. VCE-supplemented feed was given to aged diabetic rats constituting the third group. On the 21st day of the experiment, blood, lens and kidney samples were taken from all animals. Glutathione peroxidase (GSH-Px) activity in lens and kidney, reduced glutathione (GSH), vitamin E and β-carotene concentrations in kidney were lower in the diabetic group than in the control whereas plasma glucose, urea and creatinine, and kidney and lens peroxidation (LP) levels were higher in the diabetic group than in the control. However, kidney and lens LP levels, and plasma glucose, urea and creatinine values were decreased by VCE supplementation. Lens and kidney GSH-Px activity, kidney GSH, vitamin E and β-carotene concentrations and erythrocyte counts were increased by VCE treatment. Kidney weights, vitamin A, haemoglobin, hematocrit, leukocyte and platelets values were not changed by diabetes and/or VCE supplementation. VCE ameliorated also diabetes-induced histopathological changes in kidney. In conclusion, we observed that VCE supplementation is beneficial towards kidney and lens of aged diabetic rats by modulating oxidative and antioxidant systems. 相似文献

Duplex unwinding and ATPase activities of the DEAD-box helicase eIF4A are coupled by eIF4G and eIF4B
Eukaryotic initiation factor (eIF) 4A is a DEAD-box helicase that stimulates translation initiation by unwinding mRNA secondary structure. The accessory proteins eIF4G, eIF4B, and eIF4H enhance the duplex unwinding activity of eIF4A, but the extent to which they modulate eIF4A activity is poorly understood. Here, we use real-time fluorescence assays to determine the kinetic parameters of duplex unwinding and ATP hydrolysis by these initiation factors. To ensure efficient duplex unwinding, eIF4B and eIF4G cooperatively activate the duplex unwinding activity of eIF4A. Our data reveal that eIF4H is much less efficient at stimulating eIF4A unwinding activity than eIF4B, implying that eIF4H is not able to completely substitute for eIF4B in duplex unwinding. By monitoring unwinding and ATPase assays under identical conditions, we demonstrate that eIF4B couples the ATP hydrolysis cycle of eIF4A with strand separation, thereby minimizing nonproductive unwinding events. Using duplex substrates with altered GC contents but similar predicted thermal stabilities, we further show that the rate of formation of productive unwinding complexes is strongly influenced by the local stability per base pair, in addition to the stability of the entire duplex. This finding explains how a change in the GC content of a hairpin is able to influence translation initiation while maintaining the overall predicted thermal stability. 相似文献

Anolis sagrei, a Cuba and Bahama native lizard, is a successful invader in Florida and adjacent areas. Herein, we focus on conservatism in its climate niche axes and possible congruencies with its natural history properties. The not mutually exclusive hypotheses of the present study explaining its northern range limit are: (1) climatic conditions within species' native and invasive ranges are identical; (2) the species is pre‐adapted to novel conditions as a result of historical climate variations; and (3) only some niche axes limit the species' invasive distribution and the observed pattern is explained by an interplay between the potential niche within its native range and life‐history. Species distribution models for native and invasive distributions were built on ten bioclimatic variables. Using Schoener's niche overlap index, the degree of niche conservatism among variables was identified. Significances of hypothesis (1) were tested using null‐model approaches. Possible climatic pre‐adaptations were evaluated by comparing its actual tolerance within its invasive range with that of the Last Glacial Maximum (LGM) within its native range (hypothesis 2). Results of (1) and (2) are discussed in relation to natural history, approaching hypothesis 3. We detect varying overlaps in niche axes, indicating that natural history properties are associated with conservative niche axes. Climatic comparisons with LGM of native and current conditions of invasive range suggest that pre‐adaptations are unlikely. Biological systems with asexual reproduction have often attracted research on parasites and host immune defence, because parasites are expected to be better able to exploit genetically less diverse populations. In addition, maternally inherited parasitic microorganisms such as Wolbachia can directly alter the reproductive systems of their hosts and induce parthenogenesis. In the freshwater ostracod Eucypris virens, both sexual and asexual reproduction is known, and we speculated that parasite pressures might help to explain their co‐existence. This species complex inhabits shallow, often eutrophic temporary water bodies, conditions that should provide ample opportunities for parasite infections. We surveyed natural populations of E. virens throughout its Europe‐wide range for natural parasites, and particularly tested for the presence of intracellular Wolbachia bacteria. Surprisingly, the results indicate that very few E. virens populations support parasite infections. We also found no evidence for the presence of Wolbachia in the populations screened. In this paper we report on recently collected specimens of glass sponges belonging to Farreidae Gray, 1872, and Tretodictyidae Schulze, 1886 (Porifera: Hexactinellida: Hexactinosida). All specimens represent new geographical records for their genera: Coral Sea for Aspidoscopulia Reiswig, 2002 (Farreidae) and Psilocalyx Ijima, 1927 (Tretodictyidae); north‐west Atlantic for Sarostegia Topsent, 1904 (Farreidae). Two new species, Aspidoscopulia australia Dohrmann, Göcke & Janussen sp. nov. and Aspidoscopulia ospreya Dohrmann, Göcke & Janussen sp. nov. , are described. To investigate further the evolution of hexactinosidan sponges, we sequenced two nuclear (18S and 28S rDNA) and two mitochondrial [16S ribosomal rDNA, cytochrome oxidase subunit I (COI)] genes from these specimens, as well as from a recently described new species of Lonchiphora Ijima, 1927 (Farreidae). Besides corroborating the monophyly of Tretodictyidae, our molecular phylogenetic analyses support a clade of clavule‐bearing sponges with a farreoid dictyonal framework (i.e. Farreidae sensu stricto). In contrast, Sarostegia, which lacks these features, appears unrelated to this clade – instead our data are consistent with an earlier placement of this genus in Euretidae Zittel, 1877. We introduce formally the taxon Sceptrulophora Mehl 1992, and emend the classification of Hexactinosida to reflect this move and our new findings regarding the position of Sarostegia. A new species, Bilacunaria anatolica A. Duran (Apiaceae) is described and illustrated from southwest Turkey. The species grows on eroded marly stony slopes and in openings of Pinus brutia forest in Fethiye district (C2 Mu?la province). Bilacunaria anatolica, an endemic confined to southwest Anatolia, is related to B. microcarpa and B. scabra. It is also similar to the Mediterranean species Cachrys crassiloba and C. cristata in terms of habit, but differs in fruit characteristics. Diagnostic morphological and carpo‐anatomical characters are discussed. Notes are also presented on ecology and conservation status. In addition, pollen characteristics and mericarp surface features are examined by SEM. The geographical distribution of the new species and related species is mapped. 相似文献

A new subspecies, Jurinea cataonica Boiss. & Hausskn. subsp. mardinensis B. Do?an & A. Duran (Asteraceae), is described and illustrated from Anatolia, Turkey. The subspecies grows on abandoned fields in the Bakirkiri place (C8 Mardin Province) in southeastern Anatolia. It is morphologically similar to J. cataonica subsp. cataonica. Diagnostic morphological characters between these closely similar taxa are discussed and arranged in a key. Ecology and conservation status of the subspecies is also presented. In addition, the geographical distributions of the new subspecies and other related species are given. 相似文献
