The aim was to test promotive effects of oligosaccharins on root growth and development at the root apical meristem and the cell cycle using the model systems, Arabidopsis thaliana and the tobacco (Nicotiana tabacum) BY-2 cell line. Arabidopsis was grown on medium supplemented with 0.1?mg L?1 oligoxyloglucan (OX), 10?mg L?1 Pectimorf? (P) or 0.5?mg L?1 indole butyric acid (IBA). Primary root length, number of lateral root primordia, root apical meristem (RAM) length and epidermal cell length were recorded. Three genotypes were used: wild type (WT) and transgenic lines expressing either Schizosaccharomyces pombe (Sp) cdc25 or over-expressing(oe) Arath;WEE1. All treatments promoted primary root elongation but repressed lateral root production. Only P had a clear positive effect on meristem length whereas all other genotype?×?treatment interactions showed shorter RAMs. Whilst IBA, OX and P induced an increase in cell length in Spcdc25, the same treatments caused a significant decrease in WEE1 oe . Mitotic indices were also significantly higher in roots treated with oligosaccharins suggesting a shortening of the cell cycle. This hypothesis was tested in the BY-2 cell line. Both OX and P shortened the cell cycle exclusively through a shortening of G1 whilst mitotic cell size remained constant between treatments. In conclusion, both OX and P do indeed stimulate growth and shorten the cell cycle in higher plants and at the cellular level are able to reverse large and small cell size phenotypes normally exhibited by WEE1 oe and Spcdc25 genotypes, respectively.  相似文献

Aim Climate change causes shifts in species distributions, or ‘migrations’. Despite the centrality of species distributions to biodiversity conservation, the demonstrated large migration of tropical plant species in response to climate change in the past, and the expected sensitivity of species distributions to modern climate change, no study has tested for modern species migrations in tropical plants. Here we conduct a first test of the hypothesis that increasing temperatures are causing tropical trees to migrate to cooler areas. Location Tropical Andes biodiversity hotspot, south‐eastern Peru, South America. Methods We use data from repeated (2003/04–2007/08) censuses of 14 1‐ha forest inventory plots spanning an elevational gradient from 950 to 3400 m in Manu National Park in south‐eastern Peru, to characterize changes in the elevational distributions of 38 Andean tree genera. We also analyse changes in the genus‐level composition of the inventory plots through time. Results We show that most tropical Andean tree genera shifted their mean distributions upslope over the study period and that the mean rate of migration is approximately 2.5–3.5 vertical metres upslope per year. Consistent with upward migrations we also find increasing abundances of tree genera previously distributed at lower elevations in the majority of study plots. Main conclusions These findings are in accord with the a priori hypothesis of upward shifts in species ranges due to elevated temperatures, and are potentially the first documented evidence of present‐day climate‐driven migrations in a tropical plant community. The observed mean rate of change is less than predicted from the temperature increases for the region, possibly due to the influence of changes in moisture or non‐climatic factors such as substrate, species interactions, lags in tree community response and/or dispersal limitations. Whatever the cause(s), continued slower‐than‐expected migration of tropical Andean trees would indicate a limited ability to respond to increased temperatures, which may lead to increased extinction risks with further climate change.  相似文献

The 2 howler species that occur in southern Mexico, Alouatta palliata mexicana and Alouatta pigra are endangered, mainly as a result of habitat loss and fragmentation from human activity. Little is known about the gastrointestinal parasite communities affecting their populations, and lack of baseline information for populations of howler species in continuous forest habitats, makes evaluations of gastrointestinal parasite prevalence in populations in fragmented landscapes difficult. We report the results of a one-time broad survey of gastrointestinal parasites in fecal samples of individuals from several demographically stable populations of Alouatta palliata mexicana and A. pigra existing in continuous and/or protected forests. We further report similar data for populations of both species in human-fragmented landscapes. We detected 6 parasites for each howler monkey species, but only 3 of them (Trematode I, Controrchis biliophilus, Trypanoxyuris sp.) were common to both species. While parasitic prevalence in populations of both howler species was, in general, higher in the fragmented habitat than in continuous and/or protected forests. The difference is only marginally significant in Alouatta pigra. Some parasites (Coccidia and Strongylid) only appeared in populations in fragmented landscapes. Preliminary data suggest that adult males tended to have higher parasite prevalence values than those of adult females in both howler species. Parasite prevalence is associated to average group size, but not to population density in Alouatta pigra.  相似文献

The Pleistocene dune field of Tufia, located on the east of Gran Canaria (Spain), contains different stratigraphic levels of indurated pillar-like structures that are interpreted as megarhizoliths. The megarhizoliths occur at the top of different aeolian sets and reach 31.5 cm in diameter and over 1 m in height. These scattered, free-standing, vertical, cylindrical-to-slightly conical columns usually appear as hollow cylinders, displaying elliptical cross-sections aligned with the prevailing wind. On the leeward side of some specimens the external wall shows a tail of rock matrix resembling a sort of “wind shadow”. These tails and other remains of the associated rock matrix show a texture composed of long, horizontal, parallel cylinders orientated with the wind.Internally the most complete structures show five concentrically arranged zones: Zone (a), is a central pore corresponding to the cavity originally occupied by the root; Zones (b) and (c), which include alveolar and laminated carbonate textures indicating that carbonate precipitation was mostly induced by the roots and their associated microorganisms; and Zones (d) and (e), consisting mostly of aeolian sands. In (d) the sand grains show thin micritic coatings whereas in (e) vadose aragonite cements can be seen on the grain surface suggesting a less biogenic influence in their formation. The degree of cementation and the time of the precipitation of carbonate around the roots controlled the preservation of these zones. Thus, in some cases, Zones b, c and/or d are not preserved. Cylinders are up to 30× the diameter of the root that nucleated them.The presence of the megarhizoliths at the top of the aeolianite beds indicates that aeolian sedimentation halted several times, allowing soil formation and plant colonisation during slightly more humid periods. The occurrence of megarhizoliths is further proof of the alternation of arid and slightly more humid climates in the north Atlantic during the last glacial period. It is also noted that they may be misinterpreted as animal trace fossils or tree trunk casts, resulting in incorrect ichnological or palaeoenvironmental interpretations.  相似文献

There is evidence that narrowing or collapse of the pharynx can contribute to obstructive sleep-disordered breathing (SDB) in adults and children. However, studies in children have focused on those with relatively severe SDB who generally were recruited from sleep clinics. It is unclear whether children with mild SDB who primarily have hypopneas, and not frank apnea, also have more collapsible airways. We estimated airway collapsibility in 10 control subjects (9.4 +/- 0.5 yr old; 1.9 +/- 0.2 hypopneas/h) and 7 children with mild SDB (10.6 +/- 0.5 yr old; 11.5 +/- 0.1 hypopneas/h) during stable, non-rapid eye movement sleep. None of the subjects had clinically significant enlargement of the tonsils or adenoids, nor had any undergone previous tonsillectomy or adenoidectomy. Airway collapsibility was measured by brief (2-breath duration) and sudden reductions in pharyngeal pressure by connecting the breathing mask to a negative pressure source. Negative pressure applications ranging from -1 to -20 cmH(2)O were randomly applied in each subject while respiratory airflow and mask pressure were measured. Flow-pressure curves were constructed for each subject, and the x-intercept gave the pressure at zero flow, the so-called critical pressure of the upper airway (Pcrit). Pcrit was significantly higher in children with SDB than in controls (-10.8 +/- 2.8 vs. -15.7 +/- 1.2 cmH(2)O; P < 0.05). There were no significant differences in the slopes of the pressure-flow relations or in baseline airflow resistance. These data support the concept that intrinsic pharyngeal collapsibility contributes to mild SDB in children.  相似文献

Summary Most viable alleles of homeotic genes cause partial transformations within given lineages in a topographically specific fashion. We study this phenomenon as a way to understand the normal mechanisms involved in the spatial regulation of homeotic gene expression. To this end we have investigated the distribution of Ultrabithorax (Ubx) proteins in imaginai discs mutant for hypomorphic and neomorphic alleles of Ubx and alleles of trans-acting genes. We find that the morphological discontinuities observed in the adult transformations are caused by corresponding new patterns of the Ubx proteins in the imaginai anlagen. These novel patterns of Ubx proteins are understood as a consequence of a process of reinforcement-extinction of Ubx expression. The evidence suggesting that this process results from a positive feed-back loop and cell-cell interactions is discussed.  相似文献

Introduction: Endometrial cancer (EC) is the fourth most common cancer in women in developed countries. The identification of sensitive and specific biomarkers to improve early detection of EC is crucial for an appropriate management of this disease, in which 30% of patients are diagnosed only at advanced stages, which is associated with high levels of morbidity and mortality. Despite major efforts and investments made to identify EC biomarkers, no protein has yet reached the stage of clinical application.

Areas covered: This review gathers the numerous candidate biomarkers for EC diagnosis proposed in proteomic studies published from 1978 to 2017. Additionally, we summarize limitations associated with the proteomic technologies and study designs employed in those articles. Finally, we address new perspectives in EC biomarker research, including the comprehensive knowledge of previously suggested candidate biomarkers in conjunction with novel mass spectrometry-based proteomic technologies with enhanced sensitivity and specificity not yet applied to EC studies and a directed clinical perspective in the study design.
