We have used diffusion and branching process methods to investigate fixation rates, probabilities of survival per generation, and times to fixation of mutant genes under different selection methods incorporating individual and family information. Diffusion approximations fit well to simulated results even for large selection coefficients. Methods that give much weight to family information, such as BLUP evaluation which is widely used in animal breeding, reduce fixation rates of mutant genes because of the reduced effective population sizes. In general, it is observed that even mutants with relatively small heterozygous effects (say 0.1 phenotypic standard deviation) are practically ‘safe’ (i.e. their probability of loss from one generation to the next is smaller than, say, 10%) after just a few generations, typically less than 10. For methods of selection with larger effective size, such as within-family selection, the mutant is ‘safe’ in the population somewhat earlier but eventual fixation takes a longer time. Finally we evaluate the amount by which the use of marker assisted selection reduces the fixation probability of newly arisen mutants.  相似文献

Aging is characterized by a gradual functional decline of tissues with age. Adult stem and progenitor cells are responsible for tissue maintenance, repair, and regeneration, but during aging, this population of cells is decreased or its activity is reduced, compromising tissue integrity and causing pathologies that increase vulnerability, and ultimately lead to death. The causes of stem cell exhaustion during aging are not clear, and whether a reduction in stem cell function is a cause or a consequence of aging remains unresolved. Here, we took advantage of a mouse model of induced adult Sox2+ stem cell depletion to address whether accelerated stem cell depletion can promote premature aging. After a short period of partial repetitive depletion of this adult stem cell population in mice, we observed increased kyphosis and hair graying, and reduced fat mass, all of them signs of premature aging. It is interesting that cellular senescence was identified in kidney after this partial repetitive Sox2+ cell depletion. To confirm these observations, we performed a prolonged protocol of partial repetitive depletion of Sox2+ cells, forcing regeneration from the remaining Sox2+ cells, thereby causing their exhaustion. Senescence specific staining and the analysis of the expression of genetic markers clearly corroborated that adult stem cell exhaustion can lead to cellular senescence induction and premature aging.  相似文献

Population fragmentation is one of the most concerning consequences of habitat fragmentation, as small and isolated populations suffer increased genetic drift and inbreeding. However, the extent to which habitat fragmentation leads to population fragmentation depends not only on the landscape structure, but also on the response of organisms to it. This behavioral component makes it difficult to detect population fragmentation even if the habitat is fragmented, unless appropriate tools are used. In this study, we used a molecular approach to evaluate if Dahl’s toad-headed turtle (Mesoclemmys dahli) population was fragmented, given that it occurs in a very restricted area within the most degraded biome of Colombia, the tropical dry forest. We developed a panel of 15 microsatellite loci in order to perform the first genetic assessment of M. dahli across its complete geographic range. We found that M. dahli has significant genetic structure with at least four subpopulations, with surprisingly moderate to high levels of genetic diversity. Despite high levels of genetic diversity, subpopulations are very small (effective population sizes?<?50) and isolated, with little to no contemporary gene flow among them. As a consequence, mating among related individuals has been occurring, and all four populations are showing high degrees of inbreeding. To counteract this threat, we recommend an urgent genetic rescue strategy accompanied by habitat restoration, and advocate for a new conservation status assessment, not based on geographic range, but on adult population size and level of fragmentation.  相似文献

Ethnobotanical information is presented on use, management, folk nomenclature and classification of the “xoconochtli” (Stenocereus stellatus) as well as on the role of this plant in subsistence of the Nahua, Mixtec and Popoloca peoples from the Tehuacán Valley and La Mixteca Baja in Central Mexico. Among all three groups, S. stellatus was used for various purposes but mainly for its edible fruits. Different variants of this species were distinguished, named and classified by indigenous people according to characteristics of the fruit; particularly size, color and flavor of the pulp, spininess and thickness of the peel. Wild plants characteristically had small red sour fruits with many spines and thick peel, while individuals selected for cultivation usually differed in one or more of these characters. Three general forms of interaction between people and this species were found: 1) gathering of useful products from the wild; 2) management of wild populations in situ which involves the sparing and enhancing of individuals with more desirable characteristics and the removal of others during clearance of the land for agriculture; and 3) cultivation, mainly in home gardens, by propagation of vegetative parts from desirable individuals. Fruit yields per individual and per population were measured and compared in wild, managed in situ and cultivated populations from the Tehuacán Valley and La Mixteca Baja. Wild and cultivated individuals from La Mixteca Baja yielded more than wild and cultivated individuals from the Tehuacán Valley. Within each region, fruit yields were similar in wild and managed in situ populations but significantly larger in cultivated populations. Forms of management of this plant species are discussed in terms of availability of products and demand for them in the local economy.  相似文献

We report the presence, in the mitochondrial DNA (mtDNA) of all of the sexual species of the salamander family Ambystomatidae, of a shared 240- bp intergenic spacer between tRNAThr and tRNAPro. We place the intergenic spacer in context by presenting the sequence of 1,746 bp of mtDNA from Ambystoma tigrinum tigrinum, describe the nucleotide composition of the intergenic spacer in all of the species of Ambystomatidae, and compare it to other coding and noncoding regions of Ambystoma and several other vertebrate mtDNAs. The nucleotide substitution rate of the intergenic spacer is approximately three times faster than the substitution rate of the control region, as shown by comparisons among six Ambystoma macrodactylum sequences and eight members of the Ambystoma tigrinum complex. We also found additional inserts within the intergenic spacers of five species that varied from 87-444 bp in length. The presence of the intergenic spacer in all sexual species of Ambystomatidae suggests that it arose at least 20 MYA and has been a stable component of the ambystomatid mtDNA ever since. As such, it represents one of the few examples of a large and persistent intergenic spacer in the mtDNA of any vertebrate clade.   相似文献

A gene, HDC1, related to the Saccharomyces cerevisiae histone deacetylase (HDAC) gene HOS2, was isolated from the filamentous fungus Cochliobolus carbonum, a pathogen of maize that makes the HDAC inhibitor HC-toxin. Engineered mutants of HDC1 had smaller and less septate conidia and exhibited an approximately 50% reduction in total HDAC activity. Mutants were strongly reduced in virulence as a result of reduced penetration efficiency. Growth of hdc1 mutants in vitro was normal on glucose, slightly decreased on sucrose, and reduced by 30 to 73% on other simple and complex carbohydrates. Extracellular depolymerase activities and expression of the corresponding genes were downregulated in hdc1 mutant strains. Except for altered conidial morphology, the phenotypes of hdc1 mutants were similar to those of C. carbonum strains mutated in ccSNF1 encoding a protein kinase necessary for expression of glucose-repressed genes. These results show that HDC1 has multiple functions in a filamentous fungus and is required for full virulence of C. carbonum on maize.  相似文献

In this study we examined interactions between two solitary endoparasitoids, the braconid Chelonus insularis and the ichneumonid Campoletis sonorensis, and a multiple-enveloped nucleopolyhedrovirus infecting Spodoptera frugiperda larvae. We examined whether ovipositing females minimize interference by discriminating amongst hosts and examined the outcome of within-host competition between parasitoid species and between the parasitoids and the virus. The egg–larval parasitoid Ch. insularis did not discriminate between virus-contaminated and uncontaminated S. frugiperda eggs; all S. frugiperda larvae that emerged from surface-contaminated eggs died of viral infection prior to parasitoid emergence. The larval parasitoid C. sonorensis also failed to discriminate between healthy and virus-infected S. frugiperda larvae or between larvae unparasitized or parasitized by Ch. insularis. Host larvae parasitized in the egg stage by Ch. insularis were suitable for the development of C. sonorensis when they were multiparasitized by C. sonorensis as first, second, third, and fourth instars, whereas emergence of Ch. insularis was dramatically reduced (by 85 to 100%) in multiparasitized hosts. Nonspecific host mortality was significantly higher in multiparasitized hosts than in singly parasitized hosts. The development time and sex ratio of C. sonorensis in multiparasitized host larvae were unaffected by the presence of Ch. insularis larval stages. Both Ch. insularis parasitized and nonparasitized larvae of the same instar (second, third, or fourth instars) had a similar quantitative response to a challenge of virus inoculum. All host larvae that ingested a lethal dose of virus were unsuitable for Ch. insularis development. In contrast, C. sonorensis did not survive in hosts that ingested a lethal virus dose immediately after parasitism, but parasitoid survival was possible with a 2-day delay between parasitism and viral infection and the percentage of parasitoid emergence increased significantly as the interval between parasitism and viral infection increased. The development time of C. sonorensis was significantly reduced in virus-infected hosts compared to conspecifics that developed in healthy hosts. C. sonorensis females that oviposited in virus-infected hosts did not transmit the virus to healthy hosts that were parasitized subsequently. Field applications of virus for biocontrol of S. frugiperda may lead to substantial mortality of immature parasitoids, although field experiments have not yet demonstrated such an effect.  相似文献
