Nonhistone protein BA has been shown to decrease in amount in the chromatin of growth- stimulated normal rat liver (Yeoman et al. 1975. Cancer Res. 35:1249-1255) and in mitogen-stimulated normal human lymphocytes (Yeoman et al. 1976. Exp. Cell Res. 100:47- 55.). Subsequently, protein BA was purified and was shown to prefer to bind to double- stranded A-T-rich DNAs (Catino et al. 1978. Biochemistry. 17:983-987.). Immunization of rabbits with highly purified protein BA has resulted in the production of a specific antibody. A specific immunoreactivity for chromosomal protein BA has been demonstrated by immunoelectrophoresis and double antibody immunoprecipitation analysis with rabbit anti-BA immunoglobulin and IgG fractions. Light microscope examination of normal rat liver crysections by the indirect immunofluorescence procedure has demonstrated a cytoplasmic as well as a nuclear localization for protein BA with a pronounced perinucleolar fluorescence. Immunoelectron microscopy employing the peroxidase antiperoxidase method of antigen localization has confirmed the immunofluorescence data and has show a heterochromatin localization for protein BA. The relationship of the localization of protein BA to gene control in quiescent cells or to configurations of heterochromatin as well as the marked reduction in the amounts of protein BA which occur in stimulated growth states remains to be defined.  相似文献

ADP-glucose pyrophosphorylase (AGP) represents a key regulatory step in polysaccharide synthesis in organisms ranging from bacteria to plants. Higher plant AGPs are complex in nature and are heterotetramers consisting of two similar but distinct subunits. How the subunits are assembled into enzymatically active polymers is not yet understood. Here, we address this issue by using naturally occurring null mutants of the Shrunken2 (Sh2) and Brittle2 (Bt2) loci of maize as well as the yeast two-hybrid expression system. In the absence of the maize endosperm large AGP subunit (SH2), the BT2 subunit remains as a monomer in the developing endosperm. In contrast, the SH2 protein, in the absence of BT2, is found in a complex of 100 kD. A direct interaction between SH2 and BT2 was proven when they were both expressed in yeast. Several motifs are essential for SH2:BT2 interaction because truncations removing the N or C terminus of either subunit eliminate SH2:BT2 interactions. Analysis of subunit interaction mutants (sim) also identified motifs essential for protein interactions.  相似文献

Nine microsatellite loci were isolated from the insect Dolichogenidea homoeosomae (Hymenoptera: Braconidae), an important parasitoid of the sunflower moth Homosoeosoma electellum (Lepidoptera: Pyralidae), and assayed for polymorphism. All nine loci were polymorphic within the five populations tested, with two to 14 alleles per locus. Expected and observed heterozygosities ranged from 0.39 to 0.90 and 0.25 to 0.72 respectively. These are the first microsatellite primers developed for D. homeosomae and will be useful for studies of population dynamics and connectivity.  相似文献

Pearl millet (Pennisetum americanum (L.) Leeke) has a juvenilephase after which the time to panicle initiation is reducedby short daylengths. To understand more fully the mechanismunderlying temperature ? daylength interactions on panicle initiationand differentiation, plants were grown (a) at a range of constanttemperatures under a short daylength from sowing until afterpanicle differentiation and (b) at one temperature until 20d after emergence and then at a range of temperatures duringa 10 d exposure to short daylength. Temperature prior to panicle initiation determined the numberof leaves initiated on the main stem and the size of the apicaldome at the start of panicle initiation. The number of leaves,in turn, influenced the duration of the phase from panicle initiationto anthesis: this phase required a constant thermal time whenexpressed as day degrees per leaf. At anthesis, panicle lengthwas positively correlated with the number of leaves on the mainstem (and temperature) prior to panicle initiation. Changingthe temperature only during exposure to inductive daylengthsaffected the rate of growth of the apical dome so that panicledifferentiation began within 10 d at high temperature (30?C)whereas differentiation did not commence in 10 dat 21?C. Paniclesdeveloped normally if differentiation had commenced under inductivedaylengths whereas panicles were abnormal when plants were returnedto long daylengths after panicle initiation but before visibledifferentiation. Relative extension rates of the panicle during differentiationwere correlated positively with temperature. The results areconsistent with the hypothesis that panicle initiation dependson the apex attaining a critical size and that temperature,by determining the number of leaves initiated on the main stem,affects the size of the apical dome and thus the onset of panicleinitiation, the duration of paniclc differentiation and thenumber of spikelets differentiated. Vector competence for La Crosse virus (LACV) was compared for four species in the Aedes (Protomacleaya) triseriatus group: Ae. triseriatus (Say), Ae. hendersoni Cockerell, Ae. zoosophus Dyar and Knab and Ae. brelandi Zavortink (Diptera: Culicidae). Rates of replication and dissemination of virus in the mosquito hosts were compared and rates of oral transmission of virus to suckling mice were determined. Barriers to virus dissemination which limited the ability of each species to transmit virus were identified. Ae. zoosophus displayed the highest vector competence for LACV. Both infection and transmission rates were high: 99% and 85% respectively; no significant barriers to LACV were found. Disseminated infection of Ae. triseriatus with LACV was controlled primarily be a midgut escape barrier. When virus was introduced directly into the haemocoel, transmission rates were significantly increased (37% v. 79%). Ae. hendersoni showed high susceptibility to LACV infection but a very low rate of oral transmission (7%). Ae. brelandi was also highly susceptible to infection by LACV and transmitted virus at an intermediate rate (27%). Modulation of vector competence in both Ae. hendersoni and Ae. brelandi resulted from a salivary gland escape barrier. As these four species of mosquitoes comprise a closely related monophyletic series, their differences of vector competence for LACV provide an excellent model for studying the genetic basis of the barriers involved.  相似文献

Summary

We describe the spawning behaviour and some aspects of spawning periodicity in a diverse group of marine invertebrates, principally echinoderms, but including sponges, anthozoans, molluscs, and polychaetes. Our observations were made both opportunistically and on a systematic basis between 1978 and 1992 on the central and northern Great Barrier Reef. Spawning was predictable in some of the species observed, for example Bohadschia argus, Euapta godeffroyi, and Stichopus variegatus (Holothuroidea), which exhibited regular lunar and diel periodicity. Others, such as Holothuria coluber, Actinopyga lecanora, and Bohadschia graffei (Holothuroidea), Acanthaster planei (Asteroidea), Hyotissa hyotis and Arca spp. (Bivalvia) exhibited no clear lunar or diel periodicity in spawning behaviour. Mass heterospecific spawnings which involved several species, often from different phyla, were commonly observed. The species participating were usually those with unpredictable spawning patterns and while the species involved were diverse there were also occasions when spawning involved species from the same genera. Fertilization rates were measured in situ for the predictable spawner Bohadschia argus and were found to vary between 0–96% depending on the circumstances of the spawnings. Fertilization rates for the unpredictable spawners showed similar variability; Holothuria coluber and Actinopyga lecanora ranged from 9–83%.  相似文献

SUMMARY. The life-cycles of green and white morphs of the freshwater sponge Spongilla lacustris were examined in the light of past evidence that zoochlorellae may augment their sponge host's nutrition. Field collections from a lotic population of S. lacustris were supplemented by laboratory experiments on gemmule hatching and gemmule size. Both white and green S. lacustris produced sperm for a 6-week period in 1976 starting in the middle of May. Out of thirty white and thirty green sponges examined during this period, twenty white and ten green sponges contained sperm. Sperm production in both morphs was limited primarily to the basal 3.18mm of sponge tissue, and the density of sperm packets in the two morphs was the same. Out of 180 white and green sponges examined in 1976, only four eggs, no embryos, and no larvae were observed. White sponges gemmulated a week or two earlier, and produced smaller gemmules which were more uniform in size than those of green sponges. White and green gemmules hatched synchronously in the spring. In 1977 one female and numerous male specimens of S. lacustris , and numerous females but no males of another sponge, Eunapius fragilis , were found. The life-cycles are discussed in the light of other recent studies on freshwater sponges.  相似文献
