A schizont antigen for the indirect fluorescent antibody test was prepared from an in vitro culture suspension of lymphoid cells infected with Theileria annulata macroschizonts. Two cattle recovered from T. annulata infection showed marked rises in antibody titer to this schizont antigen, with peak titers of 1:40,960 and 1:2560. Using sera from these recovered cattle, T. annulata cell culture schizont antigen was shown to cross-react markedly with Theileria parva and Theileria lawrencei cell culture schizonts and with Theileria mutans piroplasms in the indirect fluorescent antibody test. In contrast, using high-titer antisera to T. parva, T. lawrencei, and T. mutans, serological cross-reactions with T. annulata schizonts were only detected with T. parva and T. lawrencei antisera, and in both instances these were minimal. The value of the indirect fluorescent antibody test in the differential diagnosis of Theileria species pathogenic for cattle is discussed. 相似文献

Junctions formed by skeletal muscles where they adhere to tendons, called myotendinous junctions, are sites of tight adhesion and where forces generated by the cell are placed on the substratum. In this regard, myotendinous junctions and focal contacts of fibroblasts in vitro are analogues. Talin is a protein located at focal contacts that may be involved in force transmission from actin filaments to the plasma membrane. This study investigates whether talin is also found at myotendinous junctions. Protein separations on SDS polyacrylamide gels and immunolabeling procedures show that talin is present in skeletal muscle. Immunofluorescence microscopy using anti-talin indicates that talin is found concentrated at myotendinous junctions and in lesser amounts in periodic bands over nonjunctional regions. Electron microscopic immunolabeling shows talin is a component of the digitlike processes of muscle cells that extend into tendons at myotendinous junctions. These findings indicate that there may be similarities in the molecular composition of focal contacts and myotendinous junctions in addition to functional analogies. 相似文献

Two Indian water buffalo (Bubalus bubalis), experimentally infected with Theileria lawrencei (Serengeti) by inoculation of stabilate material, died from acute theileriosis. Macroschizonts were first detected in both buffalo in the peripheral lymph node regional to the site of inoculation 8 days later. No microschizonts were seen in either animal and intraerythrocytic theilerial piroplasms were only detected in one in very low numbers. Both buffalo had marked febrile responses and died on Days 15 and 16 after exposure, when only low numbers of lymphoid cells were parasitized with macroschizonts, the vast majority of which contained few nuclei. Both buffalo showed evidence of terminal leucopenia and thrombocytopenia. On post mortem both animals showed generalised hyperplasia and edema of all lymphoid tissues, gross edema of the lungs, and hemorrhages in the heart and kidneys. T. lawrencei (Serengeti) infections of water buffalo and of cattle are compared and shown to be very similar. 相似文献

Nine known or potential components of the aggregation-attachment pheromone of the ticksAmblyomma hebraeum andA. variegatum were tested as attachment stimulants. Unfed adult male and female ticks were confined in linen bags on the ears of rabbits that had been treated with solutions of the test compounds; the numbers attached were recorded after 24 h. In both species, males attached more rapidly and more readily than females. InA. hebraeum, attachment of males was induced by eight compounds (benzaldehyde, benzyl alcohol, 2,6-dichlorophenol, heptadecane, 2-methyl propanoic acid, methyl salicylate, o-nitrophenol and salicylaldehyde) and attachment of females by six compounds (benzaldehyde, benzyl alcohol, 2,6-dichlorophenol, 2-methyl propanoic acid, o-nitrophenol and salicylaldehyde). InA. variegatum, attachment of males was induced by seven compounds (benzaldehyde, benzyl alcohol, 2,6-dichlorophenol, heptadecane, methyl salicylate, o-nitrophenol and salicyladeehyde) and attachment of females by two compounds (methyl salicylate and o-nitrophenol). In the present study, we have examined the cellular and subcellular distribution of talin in several tissues of the chicken. By immunocytochemistry, Western Blot analysis and [125I]vinculin overlay, talin was demonstrated in most of the main tissues and cell types of the body. Corresponding to the property of talin to bind to the fibronectin receptor, talin was found to be confined to the site of the plasma membrane that abuts the extracellular matrix in various types of mesenchymal and epithelial cells. In the central nervous system talin was almost exclusively confined to cells of the connective tissue, i.e., blood vessels and the connective tissue sheaths. No evidence was obtained for the association of talin with any type of intercellular junction. In nonadhering cells such as circulating platelets and leukocytes, talin displayed a diffuse distribution throughout the cytoplasm. These findings suggest a general role for talin in certain aspects of cellular adhesion to the extracellular matrix. 相似文献

The extracellular matrices of different tissues contain components which affect the migration, morphology and differentiation of many types of cells. These forms of cell behavior often involve dramatic changes in cytoskeletal organization. Extracellular matrix components are recognized by specific cell surface receptors which span the membrane and interact with the actin cytoskeleton. In cultured cells, the matrix receptors are concentrated in sites of cell attachment called focal adhesions. Information that is conveyed from the extracellular matrix to the cytoskeleton may involve matrix components, cell surface receptors, as well as the proteins at the cytoplasmic face of the focal adhesion which link the receptors to the actin cytoskeleton. 相似文献
