SYNOPSIS. The structural changes leading to the formation of motile microgametes from a single immobile intracellular gametocyte have been examiued in the electron microscope. After pigeon blood infected with Haemoproteus columbae was exposed to the air at room temperature for a few minutes axonemes appeared in the parasite's cytoplasm and the cytoplasm itself appeared less dense. The axonemes were connected with bundles of intranuclear microtubules that were perhaps spindle fibers. No conventional kinetosomes or centrioles have been observed. After the microgametocyte left the erythrocyte, it assumed the shape of a polarized slug or a dumb-bell. Half of the organism was surrounded by a single membrane and filled by part of the nucleus. The other half was surrounded by the remains of the multiple membranes of the gametocyte and contained pigment granules, mitochondria, axonemes and nuclear extensions. The axonemes and nuclear extensions were segregated at the periphery of the cell, exterior to the gametocyte's inner membrane, and were assembled in situ into microgametes. The mature microgamete appeared to peel off from the gametocyte, leaving a residual body.  相似文献

SYNOPSIS. The filiform microgamete of Haemoproteus columbae consists of an elongate double-walled nucleus paralleled by 2 axonemes embedded in a homogeneous matrix. At one end of the gamete, the axonemes are sharply flexed back on themselves, but no conventional kinetosome has been recognized. No mitochondria have been seen. Single-walled vesicles occur in the matrix, and the entire gamete is surrounded by a single membrane. The large round macrogamete has a conspicuous central nucleus with its outer membrane drawn out into anastomosing evaginations which extend to the periphery of the cell. A moderately electron dense material fills the space between the 2 nuclear membranes and the lumina of the evaginations. Nucleolar material may occur in scattered masses within the nucleus. One or 2 axonemes appear to arise endogenously next to the nuclear membrane. The cytoplasm is filled with ribosomes and perhaps glycogen granules. Typical protozoan mitochondria and vesicles containing pigment retained from the erythrocytic stage are found in the peripheral cytoplasm. Accumulations of dense-walled vesicles occur in the cytoplasm in conjunction with evaginations of the nuclear membrane. Amid these vesicles triple-ringed discs resembling the cytostomes of merozoites are frequently seen. Several distinct layers of dense material surround the micro-gamete.  相似文献

Electron micrographs are presented to show the morphology ofthe flagella in each of the two motile phases of Saprolegniaferax, and one visual light photograph of a stained cell ofthe second stage is added. The front flagellum of both phasesis a Flimmergeissel with hairs between 2µ and 3µlong arranged in two rows and each ending in a thin hair-point.The axis is covered by a wide transparent sheath made of somematerial which appears to liquefy after death. The hind flagellum,which is devoid of Flimmer, is covered by a similar sheath,though there are signs that this is flattened into a fin inthe hind flagellum of the second stage. Some very fine shorthairs 0?5µ long are interpreted as a possible internalframework to this fin. The axis of both flagella is fibrillar,but accurate numerical details have not yet been obtained. Theskins discarded from cysts have also been examined and shownto be covered with characteristic double-headed hooks, whichare strong enough to attach a cyst to passing objects.  相似文献

Abstract.  1. Most adult coprophagous beetles feed on fresh dung of mammalian herbivores, confining ingestion to small particles with measured maximum diameters from 2–5 to 130 μm, according to body size and kind of beetle. This study explores benefits and costs of selective feeding in a 'typical' dung beetle with a maximum diameter of ingested particles (MDIP) of 20 μm.
2. Examined dung types (from Danish domestic sheep, cattle and horse, and African wild buffalo, white rhino and elephant) contained 76–89% water. Costs of a 20 μm MDIP were often low, since 69–87% of the total nitrogen in bulk dung other than that of elephant and rhino (40–58%) was available to selective feeders.
3. Nitrogen concentrations were high – and C/N ratios low – in most types of bulk dung compared with the average food of terrestrial detritivores or herbivores. Exceptions were elephant and rhino dung with low nitrogen concentrations and high C/N ratios.
4. Estimated C/N ratios of 13–39 in bulk dung (sheep–elephant) were decreased by selective feeding to 7.3–12.6 in the ingested material. In assimilated food, ratios are probably only 5–7, as most assimilable nitrogen and carbon may be of microbial origin. If so, the assimilable food contains a surplus of nitrogen relative to carbon.
5. The primary advantage of selective feeding, particularly in dung with a high C/N ratio, may be to concentrate assimilable carbon in the ingested food. Effects of changing the MDIP within 20–106 μm are modest, especially in dung with a low C/N ratio.  相似文献

The accurate non-surgical diagnosis of breast lumps allows assessment of breast cancer patients for conservation or neoadjuvant primary treatment before surgical intervention. We have analysed the accuracy of clinical assessment, fine needle aspiration cytology (FNAC), and mammography in over 868 women seen in a symptomatic breast clinic. Clinical examination by an experienced breast physician, together with FNAC, detected over 99% of the cancers with a 12% false positive rate. A cytological diagnosis of definite carcinoma was obtained in 69% of women with breast cancer with no false positive result from the women with benign conditions producing a 100% positive predictive value. These results indicate that it would be acceptable to give pre-surgical systemic endocrine or chemotherapy to women with positive cytology which is therefore a prerequisite for a neoadjuvant therapy programme.  相似文献

We report a nearly complete skeleton of a new species of stem roller (Aves, Coracii) from the early Eocene Green River Formation of North America. The new species is most closely related to two species‐depauperate lineages, Coraciidae (rollers) and Brachypteraciidae (ground rollers), that form a monophyletic crown clade (Coracioidea) with an exclusively Old World extant distribution. Phylogenetic analysis utilizing a matrix of 133 morphological characters and sequence data from three genes (RAG‐1, c‐myc, and ND2) identifies the new species as a stem member of the Coracii more closely related to the crown clade than the only previously identified New World taxon, Primobucco mcgrewi. The phylogenetic placement of the new species and Primobucco mcgrewiindicates a widespread northern hemisphere distribution in the Eocene with subsequent restriction to Africa, Madagascar, Australia, and temperate to tropical parts of Europe and Asia. It provides evidence of further ecological diversity in early stem Coracii and convergence on crown morphologies. The new species contributes to mounting evidence that extant distributions for major avian subclades may be of comparatively recent origin. Coliiformes (mousebirds) are represented by just six extant species. These species, restricted to sub‐Saharan Africa, are all primarily frugivorous and are among the most sedentary of living birds. Previously described fossil Coliiformes preserving feather traces share the short, rounded wing shape of extant mousebirds. Along with osteological evidence, these observations have been proposed to support poor sustained flight capabilities across the stem mousebird lineage. We report a new species of Coliiformes from the early Eocene (51.66 ± 0.09 Ma) Fossil Butte Member of the Green River Formation, represented by one of the comparatively few fossils from these deposits preserving carbonized traces of the wing and tail feathering. Feather traces indicate an elongate, tapering wing shape similar to that of some extant aerial insectivores, and suggestive of a capacity for sustained and agile open‐air flight. Traces of the rectrices reveal the tail accounted for approximately two‐thirds of the total length of the bird, a proportion similar to that in living mousebirds. Phylogenetic analysis places the new species as a stem representative of Coliiformes, demonstrating for the first time that the two major clades of Coliiformes – Sandcoleidae and Colii – co‐occurred at Fossil Lake. Based on the recovered phylogeny, as well as the osteology and feathering of extant and fossil Coliiformes, the wing shape of the new species is interpreted as apomorphic. In addition to documenting unexpected morphological specialization within stem‐lineage Coliiformes, the new species adds yet another taxon to the emerging reconstruction of the diverse Paleogene avifauna from the tightly dated and nearly synchronous fossiliferous deposits of the Fossil Butte Member. © 2010 The Linnean Society of London, Zoological Journal of the Linnean Society, 2010, 160 , 685–706.  相似文献
