Background
Volatiles emitted by herbivore-infested plants are highly attractive to parasitoids and therefore have been proposed to be part of an indirect plant defense strategy. However, this proposed function of the plant-provided signals remains controversial, and it is unclear how specific and reliable the signals are under natural conditions with simultaneous feeding by multiple herbivores. Phloem feeders in particular are assumed to interfere with plant defense responses. Background. In patients with pulmonary hypertension, it is unknown whether the treatment effect of bosentan is dependent on the duration of pulmonary vessel changes. Therefore, we studied the response to bosentan in patients with life-long pulmonary vessel changes (pulmonary arterial hypertension (PAH) due to congenital heart disease (CHD)) and in patients with subacutely induced pulmonary vessel changes (chronic thromboembolic pulmonary hypertension (CTEPH)). Methods. In this open-label study, 18 patients with PAH due to CHD and 16 patients with CTEPH were treated with bosentan for at least one year. All patients were evaluated at baseline and during follow-up by means of the six-minute walk distance (6-MWD) and laboratory tests. Results. Improvement of 6-MWD was comparable in patients with PAH due to CHD (444±112 m to 471±100 m, p=0.02), and in CTEPH (376±152 m to 423±141 m, p=0.03) after three months of treatment. After this improvement, 6-MWD stabilised in both groups. Conclusion. Although duration of pulmonary vessel changes is strikingly different in patients with PAH due to CHD and CTEPH, the effect of one year of bosentan treatment was comparable. Many rangelands around the world are degraded by severe overgrazing with resulting loss of nutrients and reduced productivity. However, grazing may also increase nutrient cycling and enhance ecosystem productivity. The aim of this study was to determine effects of grazing on availability of nitrogen (N), sources of N utilized by plants and cycling and distribution of N at a low-alpine site, Southern Norway. The study was part of a sheep grazing experiment with three density levels of sheep (no sheep, 25 km?2 and 80 km?2) since 2001. The N-content of plants was determined in June 2008, August 2008 and August 2009. Indirect effects of herbivory on sources of N and N-cycling were assessed by δ15N natural abundance and the system’s distribution of added 15NH4-N. We found little evidence for grazing induced effects on availability, sources or cycling of N based on N content of plants and δ15N natural abundance. The organic soil horizon was the largest sink for the added 15NH4-N. Proportional tracer recoveries and tracer enrichments indicate a somewhat greater N cycling at grazed than at non-grazed sites. We conclude that the experimental levels of grazing have limited impact on distribution and cycling of N and thus represent sustainable ecosystem management in terms of N dynamics in the long-term. 相似文献

Human immunodeficiency virus type-1 (HIV-1) integrase catalyzes the irreversible insertion of the viral genome into host chromosomal DNA. We have developed a mammalian expression system for the synthesis of authentic HIV-1 integrase in the absence of other viral proteins. Integrase, which bears a N-terminal phenylalanine, was found to be a short-lived protein in human embryo kidney 293T cells. The degradation of integrase could be suppressed by proteasome inhibitors. N-terminal phenylalanine is recognized as a degradation signal by a ubiquitin-proteasome proteolytic system known as the N-end rule pathway. The replacement of N-terminal phenylalanine with methionine, valine, or glycine, which are stabilizing residues in the N-end rule, resulted in metabolically stabilized integrase proteins (half-life of N-terminal Met-integrase was at least 3 h). Conversely, the substitution of N-terminal phenylalanine with other destabilizing residues retained the metabolic instability of integrase. These findings indicate that the HIV-1 integrase is a physiological substrate of the N-end rule. We discuss a possible functional similarity to the better understood turnover of the bacteriophage Mu transposase and functions of integrase instability to the maintenance and integrity of the host cell genome. 相似文献

Nitrous oxide (N2O) is a greenhouse gas that also plays the primary role in stratospheric ozone depletion. The use of nitrogen fertilizers is known as the major reason for atmospheric N2O increase. Empirical bottom‐up models therefore estimate agricultural N2O inventories using N loading as the sole predictor, disregarding the regional heterogeneities in soil inherent response to external N loading. Several environmental factors have been found to influence the response in soil N2O emission to N fertilization, but their interdependence and relative importance have not been addressed properly. Here, we show that soil pH is the chief factor explaining regional disparities in N2O emission, using a global meta‐analysis of 1,104 field measurements. The emission factor (EF) of N2O increases significantly (p < .001) with soil pH decrease. The default EF value of 1.0%, according to IPCC (Intergovernmental Panel on Climate Change) for agricultural soils, occurs at soil pH 6.76. Moreover, changes in EF with N fertilization (i.e. ΔEF) is also negatively correlated (p < .001) with soil pH. This indicates that N2O emission in acidic soils is more sensitive to changing N fertilization than that in alkaline soils. Incorporating our findings into bottom‐up models has significant consequences for regional and global N2O emission inventories and reconciling them with those from top‐down models. Moreover, our results allow region‐specific development of tailor‐made N2O mitigation measures in agriculture. 相似文献

Background and Aims
As dietary saturated fatty acids are associated with metabolic and cardiovascular disease, a potentially interesting strategy to reduce disease risk is modification of the quality of fat consumed. Vegetable oils represent an attractive target for intervention, as they largely determine the intake of dietary fats. Furthermore, besides potential health effects conferred by the type of fatty acids in a vegetable oil, other minor components (e.g. phytochemicals) may also have health benefits. Here, we investigated the potential long-term health effects of isocaloric substitution of dietary fat (i.e. partial replacement of saturated by unsaturated fats), as well as putative additional effects of phytochemicals present in unrefined (virgin) oil on development of non-alcoholic fatty liver disease (NAFLD) and associated atherosclerosis. For this, we used pumpkin seed oil, because it is high in unsaturated fatty acids and a rich source of phytochemicals.Methods
ApoE*3Leiden mice were fed a Western-type diet (CON) containing cocoa butter (15% w/w) and cholesterol (1% w/w) for 20 weeks to induce risk factors and disease endpoints. In separate groups, cocoa butter was replaced by refined (REF) or virgin (VIR) pumpkin seed oil (comparable in fatty acid composition, but different in phytochemical content).Results
Both oils improved dyslipidaemia, with decreased (V)LDL-cholesterol and triglyceride levels in comparison with CON, and additional cholesterol-lowering effects of VIR over REF. While REF did not affect plasma inflammatory markers, VIR reduced circulating serum amyloid A and soluble vascular adhesion molecule-1. NAFLD and atherosclerosis development was modestly reduced in REF, and VIR strongly decreased liver steatosis and inflammation as well as atherosclerotic lesion area and severity.Conclusions
Overall, we show that an isocaloric switch from a diet rich in saturated fat to a diet rich in unsaturated fat can attenuate NAFLD and atherosclerosis development. Phytochemical-rich virgin pumpkin seed oil exerts additional anti-inflammatory effects resulting in more pronounced health effects. 相似文献997.

Metabolites generated from fuel metabolism in pancreatic beta-cells control exocytosis of insulin, a process which fails in type 2 diabetes. To identify and quantify these metabolites, global and unbiased analysis of cellular metabolism is required. To this end, polar metabolites, extracted from the clonal 832/13 beta-cell line cultured at 2.8 and 16.7 mM glucose for 48 h, were derivatized followed by identification and quantification, using gas chromatography (GC) and mass spectrometry (MS). After culture at 16.7 mM glucose for 48 h, 832/13 beta-cells exhibited a phenotype reminiscent of glucotoxicity with decreased content and secretion of insulin. The metabolomic analysis revealed alterations in the levels of 7 metabolites derived from glycolysis, the TCA cycle and pentose phosphate shunt, and 4 amino acids. Principal component analysis of the metabolite data showed two clusters, corresponding to the cells cultured at 2.8 and 16.7 mM glucose, respectively. Concurrent changes in protein expression were analyzed by 2-D gel electrophoresis followed by LC-MS/MS. The identities of 86 spots corresponding to 75 unique proteins that were significantly different in 832/13 beta-cells cultured at 16.7 mM glucose were established. Only 5 of these were found to be metabolic enzymes that could be involved in the metabolomic alterations observed. Anticipated changes in metabolite levels in cells exposed to increased glucose were observed, while changes in enzyme levels were much less profound. This suggests that substrate availability, allosteric regulation, and/or post-translational modifications are more important determinants of metabolite levels than enzyme expression at the protein level. 相似文献

Geographic variation can lead to the evolution of different local varieties, even in widespread forage plants. We investigated the performance of common forage plants in relation to their genetic diversity and local adaptation at a continental scale using reciprocal transplants at eight field sites across Europe over a 2-year period. The overall performance of the three test species, Trifolium pratense , Dactylis glomerata , Plantago lanceolata , was generally highest for plants replanted at their home site and declined with increasing transplanting distance. The three species differed in the fitness components responsible for the increased overall performance and selection advantage at home sites. In addition to the effects of local adaptation, the majority of measured traits in all three species also showed ecotypic variation. However, no single ecotype of any species was able to outperform the locally adapted strains and do best at all sites, highlighting the importance of maintaining these plant genetic resources. 相似文献
