We evaluated the effect of low-intensity pulsed ultrasound (LIPUS) on MMP-13 and MAPKs expression in rabbit knee osteoarthritis (OA). For this purpose, 18 New Zealand white rabbits were randomly and equally divided into O + L, O – L, and SO groups. In O + L group, animals underwent right back leg ACLT operation and LIPUS radiation. In O − L group, animals underwent ACLT but no LIPUS treatment. In SO (control) group, animals underwent sham operation without LIPUS. After 6 weeks, we assessed the pathologic changes in the articular surface of femoral condyle and compared using Mankin scores. Also, expression of type-II collagen, MMP-13, ERK1/2, p38, and JNK was measured by Western blot. Compared with controls, Mankin scores were higher in O + L (P < 0.05)/O − L (P < 0.01) groups. Compared with O + L group, score was higher in O − L group (P < 0.05). Compared with controls, type-II collagen expression was less in O + L/O − L groups, with more significant decrease in O − L group (P < 0.05). Contrarily, expression of MMP-13, p-ERK1/2, and p-p38 was enhanced in O + L/O − L groups as compared with controls, with more significant increase in O − L group (P < 0.01). Compared with O + L group, expression was higher in O − L group (P < 0.05). We, therefore, concluded that LIPUS application promoted cartilage repair in OA through the downregulation of MMP-13, ERK1/2, and p38.  相似文献

Elevated triglyceride (TG) and cholesterol levels are risk factors for cardiovascular disease and are often associated with diabetes and metabolic syndrome. Recent reports suggest that fibroblast growth factor (FGF)19 and FGF21 can dramatically improve metabolic dysfunction, including hyperglycemia, hypertriglyceridemia, and hypercholesterolemia. Due to their similar receptor specificities and co-receptor requirements, FGF19 and FGF21 share many common properties and have been thought to be interchangeable in metabolic regulation. Here we directly compared how pharmacological administration of recombinant FGF19 or FGF21 proteins affect metabolism in B6.V-Lepob/J leptin-deficient mice. FGF19 and FGF21 equally improved glucose parameters; however, we observed increased serum TG and cholesterol levels after treatment with FGF19 but not with FGF21. Increases in serum TGs were also observed after a 4-day treatment with FGF19 in C57BL6/J mice on a high-fat diet. This is in contrast to many literature reports that showed significant improvements in hyperlipidemia after chronic treatment with FGF19 or FGF21 in high-fat diet models. We propose that FGF19 has lipid-raising and lipid-lowering actions mediated through different FGF receptors and target tissues, and the results described here provide a potential mechanism that may explain the inconsistency in the reported effects of FGF19 on lipid metabolism.  相似文献

作为新一代植物志iFlora的重要组成部分,DNA条形码已经成为物种鉴定中重要且有效的方法。本研究以亚热带森林的乔木树种为研究对象,开展了DNA条形码的尝试性工作。为评估DNA条形码对鉴定亚热带森林树种的有效性,收集并研究了来自哀牢山自然保护区内5l科111属中204个树种的525个乔木个体。结果显示,所选4个DNA片段(rbcL,matK,trnH-psbA和ITS)的PCR扩增成功率都超过90%;测序成功率rbcL和matK最高,分别为90.7%和90.5%,trnH-psbA次之(83.6%),ITS最低(73.5%),表明4个片段在亚热带森林乔木中都具有较好的通用性。应用BLAST与NJ Tree两种方法,对物种和属水平的鉴别成功率进行统计,发现单片段中ITS最高,分别为68.4%-81.3%和99.0%~100%,核心条码rkL和matK组合的成功率是52.8%~60.2%和86.7%~90.5%,再与补充条码trnH-psbA和ITS联合,可以成功鉴别74.7%~79.6%哀牢山自然保护区亚热带森林中的乔木物种。由于ITS片段在亚热带森林部分重要树种类群(樟科和壳斗科等)中的测序成功率较差,所以对这些植物类群采用trnH-psbA作为DNA条形码是一个更好的选择。  相似文献

The homozygous NQO1*2 polymorphism results in a null NQO1 phenotype and is a susceptibility factor for occupational benzene poisoning. NQO1 plays an important role in detoxification of benzene-derived quinones but plays a role in numerous other non-metabolic cellular functions. NQO1 is expressed in endothelial cells of bone marrow which form the vascular stem cell niche important in stem cell homing and mobilization. We therefore employed a transformed human bone marrow endothelial cell (HBMEC) line to define the effects of compromising NQO1 on endothelial function. Either inhibition or knockdown of NQO1 led to decreased expression of the adhesion molecules E-selectin, VCAM-1 and ICAM-1 and decreased functional adhesion of CD34+ progenitor cells after TNFα stimulation. Suicide inhibition or knockdown of NQO1 decreased NFκB p105 precursor and NFκB p50 subunit levels as well as leading to decreased nuclear levels of NFκB phospho-p65. An additional function of endothelial cells is tube formation and angiogenesis which was inhibited by the benzene metabolite hydroquinone suggesting that endothelial function may be affected at multiple levels after exposure of NQO1*2 polymorphic individuals to benzene. These data demonstrate that NQO1 plays an upstream role in NFκB signaling and adhesion molecule expression in HBMEC and that NQO1 has important regulatory effects in its own right in addition to being a marker for Nrf-2 activation. Metabolic susceptibility factors such as NQO1 have roles in addition to detoxification of reactive intermediates and interrogation of these novel roles can inform both mechanisms of toxicity and human risk assessment.  相似文献

以花生品种花育25号为试验材料,采用盆栽试验研究了开花期干旱和盐分胁迫对花生生长发育和荚果产量的影响,并运用高通量测序技术分析干旱、盐胁迫及旱盐双重胁迫下,花生根际土壤细菌群落结构的变化特征。结果表明: 不同胁迫处理花生根际土壤细菌群落均以变形菌门、放线菌门、Saccharibacteria、绿弯菌门、蓝藻菌门和酸杆菌门为主。干旱、盐胁迫及旱盐双重胁迫均不同程度降低了变形菌门和放线菌门的相对丰度,但显著提高了蓝藻菌门的含量,且旱盐双重胁迫较其单一胁迫引起的根际蓝藻菌门丰度变化更显著。土壤细菌功能预测分析显示,信号转导机制、防御机制及翻译后修饰、蛋白质周转和分子伴侣等相关功能在旱盐双重胁迫的细菌中活性更强,可能对花生生长及胁迫应答具有重要影响。统计学分析显示,开花期干旱、盐胁迫和旱盐双重胁迫严重影响花生生长发育,并显著降低产量。研究结果可为通过改良土壤微生物环境来提高植物胁迫耐受性提供参考。  相似文献

Background

Understanding protein complexes is important for understanding the science of cellular organization and function. Many computational methods have been developed to identify protein complexes from experimentally obtained protein-protein interaction (PPI) networks. However, interaction information obtained experimentally can be unreliable and incomplete. Reconstructing these PPI networks with PPI evidences from other sources can improve protein complex identification.

Results

We combined PPI information from 6 different sources and obtained a reconstructed PPI network for yeast through machine learning. Some popular protein complex identification methods were then applied to detect yeast protein complexes using the new PPI networks. Our evaluation indicates that protein complex identification algorithms using the reconstructed PPI network significantly outperform ones on experimentally verified PPI networks.

Conclusions

We conclude that incorporating PPI information from other sources can improve the effectiveness of protein complex identification.  相似文献

Temperature is one of the most significant environmental factors that affects germination of grass seeds. Reliable prediction of the optimal temperature for seed germination is crucial for determining the suitable regions and favorable sowing timing for turf grass cultivation. In this study, a back-propagation-artificial-neural-network-aided dual quintic equation (BP-ANN-QE) model was developed to improve the prediction of the optimal temperature for seed germination. This BP-ANN-QE model was used to determine optimal sowing times and suitable regions for three Cynodon dactylon cultivars (C. dactylon, ‘Savannah’ and ‘Princess VII’). Prediction of the optimal temperature for these seeds was based on comprehensive germination tests using 36 day/night (high/low) temperature regimes (both ranging from 5/5 to 40/40°C with 5°C increments). Seed germination data from these temperature regimes were used to construct temperature-germination correlation models for estimating germination percentage with confidence intervals. Our tests revealed that the optimal high/low temperature regimes required for all the three bermudagrass cultivars are 30/5, 30/10, 35/5, 35/10, 35/15, 35/20, 40/15 and 40/20°C; constant temperatures ranging from 5 to 40°C inhibited the germination of all three cultivars. While comparing different simulating methods, including DQEM, Bisquare ANN-QE, and BP-ANN-QE in establishing temperature based germination percentage rules, we found that the R2 values of germination prediction function could be significantly improved from about 0.6940–0.8177 (DQEM approach) to 0.9439–0.9813 (BP-ANN-QE). These results indicated that our BP-ANN-QE model has better performance than the rests of the compared models. Furthermore, data of the national temperature grids generated from monthly-average temperature for 25 years were fit into these functions and we were able to map the germination percentage of these C. dactylon cultivars in the national scale of China, and suggested the optimum sowing regions and times for them.  相似文献

通过直接在大肠杆菌碱裂解上清中加入十六烷基三甲基溴化铵(CTAB), 优化CTAB与质粒DNA量的比例、质粒DNA选择性释放溶液的选择和TritonX-114的使用, 建立了简单、易行的大规模质粒DNA纯化工艺。纯化质粒DNA的质量检测结果显示, CTAB纯化的质粒DNA无菌体RNA污染, 菌体基因组DNA、内毒素和蛋白含量分别小于 100 ng/mg、50 EU/mg和10 mg/mg质粒DNA, OD260/OD280比值介于1.75~1.85之间, 超螺旋质粒DNA的比例大于80%, 该工艺纯化的质粒DNA能达到或接近FDA规定的人用质粒DNA的各项指标, 整个过程不使用动物源性酶和苯酚、氯仿、无水乙醇等有毒或易燃、易爆试剂, 成本低廉, 工艺环保。  相似文献

水獭是水生生态系统重要的指示种和旗舰种, 由于强烈的人为干扰, 中国的水獭种群数量大幅下降, 部分区域已局部灭绝。然而目前国内对水獭的调查和研究非常有限, 本底不清的状况已经严重影响到水獭的野外保育工作。本文以东北地区的欧亚水獭指名亚种(Lutra lutra lutra)为研究对象, 基于2016-2020年的调查数据, 使用组合建模的方法评估了水獭的潜在分布区; 利用地理信息系统和系统保护规划软件分析了水獭的保护优先区并计算了各省级行政区内水獭潜在分布区和保护优先区面临的人类压力; 结合国家级自然保护区的空间布局分析了水獭的保护现状, 并以内蒙古森工集团、大兴安岭林业集团、伊春森工集团三大国有林区为例分析了重点国有林区在水獭保护中的作用。结果表明: (1)水獭潜在分布区和保护优先区面积分别为104,515.04 km2和45,448.99 km2, 其中大兴安岭的水獭保护优先区集中连片, 并与小兴安岭的保护优先区相连, 栖息地之间没有明显地理隔离, 是维持东北地区水獭种群稳定的重中之重; (2)水獭面临的人类压力大小依次为: 辽宁 > 吉林 > 黑龙江 > 内蒙古; (3)研究区内110个国家级自然保护区中有63个包含水獭潜在分布区, 覆盖面积为12,168.93 km2, 仅占水獭潜在分布区面积的11.64%, 其中32个国家级自然保护区包含水獭保护优先区, 占水獭保护优先区面积的10.88%; (4)三大国有林区涵盖了71.18%的水獭潜在分布区和79.26%的保护优先区(面积分别为74,390.89 km2和36,022.22 km2)。由此可见, 尽管水獭潜在分布区中国家级自然保护区占比较低, 但是在天然林全面禁伐的背景下, 重点国有林区可能在未来东北地区的生物多样性保护中发挥更大作用, 因此我们建议将重点国有林区中具有重要保护价值的区域逐步纳入以国家公园为主体的自然保护地体系中, 以实现生物多样性的系统性和完整性保护。最后, 本文结合研究结果和实地调研提出以下保护建议: (1)加强对河流污染物的管理; (2)控制渔民捕鱼强度; (3)开展全面的水獭专项调查并建立长期的监测体系; (4)加大对水獭的科研投入; (5)加强宣传力度, 提升公众保护意识。  相似文献
