The authors investigated the morphology and the elastic properties of living cultured rat liver macrophages (Kupffer cells) with an atomic force microscope (AFM). Continuous imaging and elasticity mapping of individual cells in physiological buffer was carried out for several hours without damaging the cells as judged by their persistent undisturbed morphology. Dynamic events such as protrusive activity were observed in time course. The importance of the cytoskeleton for the mechanical properties of the cell has been investigated by measuring the cell's elasticity as a function of position. Chemical disassembly of the actin network by applying 10μg/ml cytochalasin B decreased the cell's average elastic modulus seven-fold within less than 40 minutes. Treating the cells with 0.1μg/ml latrunculin A resulted in a two-fold decrease in the elastic modulus merely in the perinuclear region after 40 minutes, whereas other parts of the cell were not affected. 相似文献

Individuals ofArabidopsis thaliana, collected in different naturalpopulations, were grown in controlled and elevated CO2in a glasshouse.Following germination, root growth of progeny of different linesof these populations was studied in control and elevated atmosphericCO2. No significant direct effect of atmospheric CO2concentrationcould be demonstrated on root growth. An important parentaleffect was apparent, namely that root length and branching weredecreased in seeds collected from a mother plant which had beengrown in elevated CO2. This was correlated with smaller seeds,containing less nitrogen. These parental effects were geneticallyvariable. Abstract The spatial organization of individuals, or groups of individuals, within a population can provide valuable information about social organization and population dynamics. We analysed the spatial distribution of nests of the sociable weaver (Philetairus socius) on two farms in the Kalahari. Sociable weavers build large communal nests on big savannah trees, forming a pattern of trees with and without nests. We used two spatial statistics, Ripley's K and the pair correlation function, to describe characteristics of the point patterns over a range of distances. (i) At distances of 200 and 300 m, communal nests were clustered. (ii) At distances greater than 1000 m, communal nests were regularly distributed. These findings are independent of the spatial distribution of trees. Furthermore, we used Moran's I to analyse spatial autocorrelation of nest sizes. We expected negative autocorrelation because of competition between nests. But on both farms there was no significant autocorrelation of nest sizes for any distance class. The regular distribution observed at larger distances may indicate competition and/or territoriality among different nests, but the lack of spatial autocorrelation between nest sizes suggests that these interactions may happen between nest clusters rather than between single nests. This was confirmed by significant clustering of nests on small scales. We thus suggest, that colonies of P. socius consist of several nests on adjacent trees forming a cluster of subcolonies. The question why sociable weavers establish subcolonies instead of adding more chambers to the natal nest, could not simply be answered by limitation of nesting space. In diverse plant communities the relative contribution of species to community biomass may change considerably in response to elevated CO2. Along with species‐specific biomass responses, reproduction is likely to change as well with increasing CO2 and might further accelerate shifts in species composition. Here, we ask if, after 5 years of CO2 exposure, seed production and seed quality in natural nutrient‐poor calcareous grassland are affected by elevated CO2 (650 μ L L?1 vs 360 μ L L?1) and how this might affect long‐term community dynamics. The effect of elevated CO2 on the number of flowering shoots (+ 24%, P < 0.01) and seeds (+ 29%, P = 0.06) at the community level was similar to above ground biomass responses in this year, suggesting that the overall allocation to sexual reproduction remained unchanged. Compared among functional groups of species we found a 42% increase in seed number (P < 0.01) of graminoids, a 33% increase (P = 0.07) in forbs, and no significant change in legumes (? 38%, n.s.) under elevated CO2. Large responses particularly of two graminoid species and smaller responses of many forb species summed up to the significant or marginally significant increase in seed number of graminoids and forbs, respectively. In several species the increase in seed number resulted both from an increase in flowering shoots and an increase in inflorescence size. In most species, seeds tended to be heavier (+ 12%, P < 0.01), and N‐concentration of seeds was significantly reduced in eight out of 13 species. The fraction of germinating seeds did not differ between seeds produced in ambient and elevated CO2, but time to germination was significantly shortened in two species and prolonged in one species when seeds had been produced in elevated CO2. Results suggest that species specific increases in seed number and changes in seed quality will exert substantial cumulative effects on community composition in the long run. 相似文献

1. Stocking of piscivores in shallow, eutrophicated lakes to reduce cyprinid densities is a common approach in lake restorations. Young‐of‐the‐year (YOY) pike Esox lucius are frequently used to reduce cyprinid densities, but their effectiveness is equivocal. This study uses a simple model to assess the efficacy of pike stocking as a lake restoration tool. 2. The model evaluates YOY pike consumption in relation to different survival and growth patterns, timing of stocking, degree of piscivory of pike as well as YOY cyprinid growth patterns and productivity. The parameters used in the model are obtained from field surveys in eight study lakes stocked with YOY pike as well as from the literature. 3. Our model showed that all parameters studied were important for predicting the effects of pike stocking on cyprinids. In particular, body size at stocking, cyprinid production and pike survival were good predictors of biomanipulation success. 4. However, our model showed that only under very specific circumstances will stocking of YOY pike result in a reduction of YOY cyprinid densities large enough to affect water quality. 5. The findings of this study question the efficacy of pike stocking as an appropriate and reliable tool for restoration programmes in shallow lakes, and indicate that when used efforts should be made to optimize the timing of stocking in relation to YOY cyprinid production and to increase the stocking body size of the pike. 相似文献

Root-colonizing non-pathogenic bacteria can increase plant resistance to biotic and abiotic stress factors. Bacterial inoculates have been applied as biofertilizers and can increase the effectiveness of phytoremediation. Inoculating plants with non-pathogenic bacteria can provide 'bioprotection' against biotic stresses, and some root-colonizing bacteria increase tolerance against abiotic stresses such as drought, salinity and metal toxicity. Systematic identification of bacterial strains providing cross-protection against multiple stressors would be highly valuable for agricultural production in changing environmental conditions. For bacterial cross-protection to be an effective tool, a better understanding of the underlying morphological, physiological and molecular mechanisms of bacterially mediated stress tolerance, and the phenomenon of cross-protection is critical. Beneficial bacteria-mediated plant gene expression studies under non-stress conditions or during pathogenic rhizobacteria–plant interactions are plentiful, but only few molecular studies on beneficial interactions under abiotic stress situations have been reported. Thus, here we attempt an overview of current knowledge on physiological impacts and modes of action of bacterial mitigation of abiotic stress symptoms in plants. Where available, molecular data will be provided to support physiological or morphological observations. We indicate further research avenues to enable better use of cross-protection capacities of root-colonizing non-pathogenic bacteria in agricultural production systems affected by a changing climate. 相似文献

The development of insecticide resistance in pest insects is an increasing problem for agriculture, forestry and public health. Aphids are ubiquitous herbivorous insects, with approximately 4700 known species, of which less than 5% exploit the agricultural environment successfully. Of these, the peach‐potato aphid Myzus persicae Sulzer is recognized as one of the most important pests worldwide because it has acquired resistance to many insecticides. Although resistance to insecticides provides important benefits for pests in agricultural fields that are treated with insecticides, it may be associated with fitness (or other) costs in environments that are insecticide free. In the present study, the fitness and energy costs that might be experienced by M. persicae in an insecticide‐free environment when carrying at least one insecticide resistance mutation (IRM), or by having an increased production of esterases, are evaluated. The study investigates whether genotypes that have an IRM also have enhanced esterase production, whether there is any metabolic cost associated with insecticide resistance, and whether there are any fitness costs associated with insecticide resistance and metabolic expenditure. The intrinsic rate of increase, standard metabolic rate (i.e. a measure of maintenance costs) and constitutive esterase activity are determined for 30 different multilocus genotypes carrying (or not carrying) at least one of the two most frequent insecticide resistance mutations (MACE and kdr/super‐kdr) that occur in Chile. The results show that genotypes carrying at least one IRM have higher levels of total esterase activity than genotypes without an IRM, that there is no evidence of an energy cost associated with total esterase activity or IRM, and no evidence for a reproductive fitness cost associated with total esterase activity, IRM or metabolic rate. The results agree with previous studies showing linkage disequilibrium between insecticide resistance mechanisms, although they contrast with those of studies that report fitness costs associated with insecticide resistance in Myzus persicae. 相似文献

Abstract: The infaunal irregular echinoid, Tithonia oxfordiana, is described and compared to congeneric species previously described from Upper Jurassic and lowermost Cretaceous strata. This new species characterizes a monospecific echinoid assemblage, which occurs only in some places where deep‐marine middle Oxfordian deposits are exposed in south‐east France. Specimens are closely packed and clearly concentrated at the top of small carbonate chemoherms; a close connection of the echinoids with the emission of reduced chemicals, which were oxidized by chemoautotrophic bacteria, is highly probable. Based on general test shape and plate architecture, T. oxfordiana probably was a deposit feeder on chemosynthetic organic matter produced by such bacteria. In view of the fact that T. oxfordiana is the sole species of the Jurassic genus Tithonia known from Oxfordian strata, it is postulated that chemoherms possibly acted as refugia for these peculiar echinoids, which have an episodic record between the Callovian and Valanginian. 相似文献
