Carbon dioxide has been found to produce various negative consequences for a number of aquatic species and is projected to rise in the future for many aquatic ecosystems. Crayfish act as indicators of water quality and function as keystone species in aquatic food webs; however, there is a paucity of research on how crayfish may respond to elevated CO2. This study quantified how shelter-seeking behavior in freshwater crayfish (e.g., family Cambaridae), a behavior critical for survival and fitness, may change following exposure to elevated pCO2. Red Swamp crayfish (Procambarus clarkii, Girard, 1852) were exposed to one of three potential levels of dissolved CO2 that could be found in freshwater basins currently or under future climate change conditions: Control (2 levels as the respective initial exposure. The treatment aquarium contained a shelter and was divided into three equal sections based on proximity to that shelter. Crayfish proximity to the shelter (defined by the tank sections) in the treatment aquarium was monitored every 5 s for a 2-min trial. Crayfish spent differing amounts of time in differing zones of the experiment and had different levels of activity, depending on their pCO2 exposure; crayfish acclimated to High pCO2 increased their time spent hiding and decreased their overall activity when compared to the Low pCO2 and Control treatments. Augmented shelter-seeking behavior may affect crayfish social hierarchies, feeding, mating, and mortality, which could generate cascading effects on the ecology of many freshwater ecosystems.  相似文献

The current study used a stationary acoustic telemetry array to monitor the depth selection of adult smallmouth bass Micropterus dolomieu in a large, oligotrophic lake in Ontario, Canada. At an annual scale, smallmouth bass demonstrated regular, seasonal changes in inhabited depths: fish occupied shallow water during the summer (2–5 m depth) and descended to deeper water (12–15 m depth) during winter under ice. Smallmouth bass remained above the thermocline in the summer, seasonal depth patterns did not vary across fish size and movements to and from seasonal depths were closely linked to the development and degradation of the thermocline. At finer time scales, smallmouth bass exhibited diel vertical migrations in summer, with fish moving to <2 m at night, and then descending to 3–5 m during the day. This pattern remained constant during the summer period examined, and varied with size such that larger fish remained deeper than smaller fish. During winter, depth did not vary across the 3-month monitoring period ( c . 14 m), but small (<2 m) changes in depth were observed periodically, suggesting limited movements were occurring. Results are further discussed in the context of climate change and reproductive success for this species.  相似文献

Secretory granules containing a hybrid protein consisting of the regulated secretory protein tissue plasminogen activator and an enhanced form of green fluorescent protein were tracked at high spatial resolution in growth cones of differentiated PC12 cells. Tracking shows that granules, unlike synaptic vesicles, generally are mobile in growth cones. Quantitative analysis of trajectories generated by granules revealed two dominant modes of motion: diffusive and directed. Diffusive motion was observed primarily in central and peripheral parts of growth cones, where most granules diffused two to four orders of magnitude more slowly than comparably sized spheres in dilute solution. Directed motion was observed primarily in proximal parts of growth cones, where a subset of granules underwent rapid, directed motion at average speeds comparable to those observed for granules in neurites. This high-resolution view of the dynamics of secretory granules in growth cones provides insight into granule organization and release at nerve terminals. In particular, the mobility of granules suggests that granules, unlike synaptic vesicles, are not tethered stably to cytoskeletal structures in nerve terminals. Moreover, the slow diffusive nature of this mobility suggests that secretory responses involving centrally distributed granules in growth cones will occur slowly, on a time scale of minutes or longer.  相似文献

Spexin is novel biomarker, which plays a potential role in glucose and lipid metabolisms. However, there was paucity of serum spexin levels in obesity and diabetes mellitus subjects. Hence the current study was aimed to find the relationship between the serum spexin levels in type 2 Diabetes mellitus (type 2 DM) with extrapolation of cardiovascular disease (CVD) risk. A cross-sectional study included 330 participants, subdivided as control (n=110), type 2 DM (n=110) and type 2 DM with CVD groups (n=110). HbA1c, insulin, lipid profile, spexin & leptin including blood pressure and body mass index were analyzed from all the participants. The serum spexin levels (ng/ml) were significantly decreased in type 2 DM (mean ± sd: 0.65 ± 0.03) and type 2 DM with CVD (0.48 ± 0.02) groups compared to the control (0.79 ± 0.03) group (p<0.001). The decreased spexin levels were observed in type 2 DM, and further more decreased in type 2 DM with CVD patients compared to controls indicating that spexin levels could be served as an early prediction of obesity-induced T2DM with CVD risk.  相似文献

Catch-and-release recreational angling has become very popular as a conservation strategy and as a fisheries management tool for a diverse array of fishes. Implicit in catch-and-release angling strategies is the assumption that fish experience low mortality and minimal sub-lethal effects. Despite the importance of this premise, research on this topic has focused on several popular North American sportfish, with negligible efforts directed towards understanding catch-and-release angling effects on alternative fish species. Here, we summarise the existing literature to develop five general trends that could be adopted for species for which no data are currently available: (1) minimise angling duration, (2) minimise air exposure, (3) avoid angling during extremes in water temperature, (4) use barbless hooks and artificial lures/flies, and (5) refrain from angling fish during the reproductive period. These generalities provide some level of protection to all species, but do have limitations. Therefore, we argue that a goal of conservation science and fisheries management should be the creation of species-specific guidelines for catch-and-release. These guidelines would take into account the inter-specific diversity of fishes and variation in fishing techniques. As recreational angling continues to grow in popularity, expanding to many developing countries, and targeting alternative species, it is important that reasonable data appropriate for specific fish and fisheries are available. The sustainable use and conservation of recreational fishery resources will depend upon the development and dissemination of effective catch-and-release angling strategies based upon sound science to stakeholders around the world.  相似文献

Trace metal (Zn, Pb, Cu, Cr and Cd) concentrations in the water column and in the liver, muscle and gill tissues of Parachanna obscura and Clarias gariepinus in Agulu Lake, Nigeria, were investigated in June 2014 and compared with WHO and FAO safe limits for water and fish. Hazard index (HI) values were estimated to assess the potential public health risk of consuming contaminated fish. Lead and cadmium exceeded WHO guideline values for drinking water. In most cases, variations in concentration of the metals in organs were liver > muscle > gill. Differences in tissue-specific concentrations between species were not significant, except for zinc in muscles and gills. Cadmium and chromium were not detected in the fish, but lead was above the FAO maximum value for consumption. Hazard index values were below 1, indicating a low risk to public health. However, trace metal contamination in Agulu Lake is increasing.  相似文献

Lead ions are particularly dangerous to the photosynthetic apparatus, but little is known about the effects of trace metals, including Pb, on regulation of chloroplast redistribution. In this study a new effect of lead on chloroplast distribution patterns and movements was demonstrated in mesophyll cells of a small-sized aquatic angiosperm Lemna trisulca L. (star duckweed). An analysis of confocal microscopy images of L. trisulca fronds treated with lead (15 μM Pb2+, 24 h) in darkness or in weak white light revealed an enhanced accumulation of chloroplasts in the profile position along the anticlinal cell walls, in comparison to untreated plants. The rearrangement of chloroplasts in their response to lead ions in darkness was similar to the avoidance response of chloroplasts in plants treated with strong white light. Transmission electron microscopy X-ray microanalysis showed that intracellular chloroplast arrangement was independent of the location of Pb deposits, suggesting that lead causes redistribution of chloroplasts, which looks like a light-induced avoidance response, but is not a real avoidance response to the metal. Furthermore, a similar redistribution of chloroplasts in L. trisulca cells in darkness was observed also under the influence of exogenously applied hydrogen peroxide (H2O2). In addition, we detected an enhanced accumulation of endogenous H2O2 after treatment of plants with lead. Interestingly, H2O2-specific scavenger catalase partly abolished the Pb-induced chloroplast response. These results suggest that H2O2 can be involved in the avoidance-like movement of chloroplasts induced by lead. Analysis of photometric measurements revealed also strong inhibition (but not complete) of blue-light-induced chloroplast movements by lead. This inhibition may result from disturbances in the actin cytoskeleton, as we observed fragmentation and disappearance of actin filaments around chloroplasts. Results of this study show that the mechanisms of the toxic effect of lead on chloroplasts can include disturbances in their movement and distribution pattern.  相似文献

Background

Using haplotype blocks as predictors rather than individual single nucleotide polymorphisms (SNPs) may improve genomic predictions, since haplotypes are in stronger linkage disequilibrium with the quantitative trait loci than are individual SNPs. It has also been hypothesized that an appropriate selection of a subset of haplotype blocks can result in similar or better predictive ability than when using the whole set of haplotype blocks. This study investigated genomic prediction using a set of haplotype blocks that contained the SNPs with large effects estimated from an individual SNP prediction model. We analyzed protein yield, fertility and mastitis of Nordic Holstein cattle, and used high-density markers (about 770k SNPs). To reach an optimum number of haplotype variables for genomic prediction, predictions were performed using subsets of haplotype blocks that contained a range of 1000 to 50 000 main SNPs.

Results

The use of haplotype blocks improved the prediction reliabilities, even when selection focused on only a group of haplotype blocks. In this case, the use of haplotype blocks that contained the 20 000 to 50 000 SNPs with the highest effect was sufficient to outperform the model that used all individual SNPs as predictors (up to 1.3 % improvement in prediction reliability for mastitis, compared to individual SNP approach), and the achieved reliabilities were similar to those using all haplotype blocks available in the genome data (from 0.6 % lower to 0.8 % higher reliability).

Conclusions

Haplotype blocks used as predictors can improve the reliability of genomic prediction compared to the individual SNP model. Furthermore, the use of a subset of haplotype blocks that contains the main SNP effects from genomic data could be a feasible approach to genomic prediction in dairy cattle, given an increase in density of genotype data available. The predictive ability of the models that use a subset of haplotype blocks was similar to that obtained using either all haplotype blocks or all individual SNPs, with the benefit of having a much lower computational demand.  相似文献

Excessive action of angiotensin II on mitochondria has been shown to play an important role in mitochondrial dysfunction, a common feature of atherogenesis and kidney injury. Angiotensin-(1–7)/Mas receptor axis constitutes a countermeasure to the detrimental effects of angiotensin II on AT1 receptors. The aim of the study was to assess the effects of angiotensin-(1–7) peptidomimetic AVE0991 on the kidney mitochondrial proteome in widely used animal model of atherosclerosis (apoE?/? mice). Proteins changed in apoE?/? mice belonged to the groups of antioxidant enzymes, apoptosis regulators, inflammatory factors and metabolic enzymes. Importantly, AVE0991 partially reversed atherosclerosis-related changes in apoE?/? mice.  相似文献
