1. The white-clawed crayfish Austropotamobius pallipes has a widespread distribution in Europe, but since the last century its distribution has been severely affected by a combination of factors, particularly the disease crayfish plague, introduced by foreign crayfish species. At present A. pallipes is considered as vulnerable and endangered and measures are being taken in a number of countries to conserve it.
2. The application of genetics to conservation is of practical value particularly where restocking is being considered. This study assesses levels of genetic differentiation from an analysis of mitochondrial DNA (mtDNA) variation by restriction fragment length polymorphism of four populations sampled on a European scale. Six different composite haplotypes were detected among sixty-four crayfish representing the four populations. mtDNA nucleon diversity values within species ranged from 0.63% to 12.35%. A close genetic similarity was found between British and French populations of A. pallipes .
3. Results show that mtDNA can be as a suitable diagnostic marker for measuring genetic diversity between crayfish populations and that such information is of use in planning crayfish conservation strategies. 相似文献

The present study examined the contemporary genetic composition of the Eurasian badger, Meles meles, in Ireland, Britain and Western Europe, using six nuclear microsatellite loci and a 215‐bp fragment of the mitochondrial DNA control region. Significant population structure was evident within Europe (global multilocus microsatellite FST = 0.205, P < 0.001; global mitochondrial control region ΦST = 0.399, P < 0.001). Microsatellite‐based cluster analyses detected one population in Ireland, whereas badgers from Britain could be subdivided into several populations. Excluding the island populations of Ireland and Britain, badgers from Western Europe showed further structuring, with evidence of discrete Scandinavian, Central European, and Spanish populations. Mitochondrial DNA cluster analysis grouped the Irish population with Scandinavia and Spain, whereas the majority of British haplotypes grouped with those from Central Europe. The findings of the present study suggest that British and Irish badger populations colonized from different refugial areas, or that there were different waves of colonization from the source population. There are indications for the presence of an Atlantic fringe element, which has been seen in other Irish species. Tropical forests have undergone repeated fragmentation and expansion during Pleistocene glacial and interglacial periods, respectively. The effects of this repeated forest fragmentation in driving vicariance in tropical taxa have been well studied. However, relatively little is known about how often this process results in allopatric speciation, since it may be inhibited by recurrent gene flow during repeated secondary contact, or to what extent Pleistocene‐dated speciation results from ecological specialization in the face of gene flow. Here, divergence times and gene flow between three closely‐related mosquito species of the Anopheles dirus species complex endemic to the forests of Southeast Asia, are inferred using coalescent based Bayesian analysis. An Isolation with Migration model is applied to sequences of two mitochondrial and three nuclear genes, and 11 microsatellites. The divergence of An. scanloni has occurred despite unidirectional nuclear gene flow from this species into An. dirus. The inferred asymmetric gene flow may result from the unique evolutionary adaptation of An. scanloni to limestone karst habitat, and therefore the fitness advantage of this species over An. dirus in regions of sympatry. Mitochondrial introgression has led to the complete replacement of An. dirus haplotypes with those of An. baimaii through a recent (~62 kya) selective sweep. Speciation of An. baimaii and An. dirus is inferred to have involved allopatric divergence throughout much of the Pleistocene. Secondary contact and bidirectional gene flow has occurred only within the last 100 000 years, by which time the process of allopatric speciation seems to have been largely completed. 相似文献

An important issue in consumer oriented studies is to measure how closely a specific product or perception matches the representation people have of it. We used a concept matching technique to assess the degree to which a variety of different cheeses matched subjects’ concepts of Cheddar cheese. Eighteen subjects made their judgments using their own past experiences with Cheddar cheeses. They made judgments in three separate conditions: only the odor, only the taste and texture and normal eating. This technique provided valid and reliable information on the extent to which the cheeses matched the subjects’ concept of Cheddar cheese. Agreement between classifications of cheeses based on taste and texture only and based on normal eating was high (R=0.90). Agreement between classifications based only on odor and those based on normal eating was weaker (R-0.59). Thus, the taste and texture of the cheeses were more useful than the odor for classifying cheeses as Cheddar. 相似文献

Abstract.
- 1 The response of different clones of sand-dune willow, Salix cordata, to herbivory by a specialist herbivore, Altica subplicata, was studied in three glasshouse experiments. Plants were caged and exposed to three herbivory treatments: no beetles, low number of beetles, and high number of beetles.
- 2 Plants consistently had significantly higher growth rates in the absence of herbivory than under conditions of low or high herbivory (1.5–6 times higher).
- 3 Herbivore treatment influenced mortality from drought stress; more plants from the low and high herbivory treatments (40% and 80%) died from drought stress than did control plants (0%).
- 4 Clone genotype significantly influenced growth rates and the susceptibility of plants to drought stress. However, clones showed similar growth responses to herbivory, suggesting a lack of genetic variation in tolerance or resistance to herbivory.

Correlation of Growth between Mycelium and Rhizomorphs in Sphaerostilbe repens In Sphaerostilbe repens Berkeley and Broome, cultured on a solid medium in Petri dishes, growth of mycelium and chizomorphs depends on the thickness of the culture medium underlying the thallus. On shallow layers of agar medium, aggregated units were fewer and rhizomorph growth reduced. Vegetative mycelium which starts growth earlier than rhizomorphs contributed to inhibit the development of these latter. On deep layers of substrate, rhizomorphs were numerous, elongated rapidly, and eventually inhibited undifferentiated mycelium. – There was no evidence for the involvement of any inhibitor substance diffusing into culture medium or located in the thallus. Development of aerial mycelium and rhizomorphs, could, on the contrary, be interpreted as nutritional competition between these two parts of the colony. 相似文献

SYNOPSIS. Flamingos (Aves, Phoenicopteridae) represent an ancientlineage of long-legged, microphagous, colonial wading birds.Although often perceived as tropical, flamingo distributionis more closely tied to the great deserts of the world, andto hypersaline sites, than it is to equatorial regions. Manyaspects of flamingo behavior and ecology can be studied in captivity.Experimental studies involving captive birds, when combinedwith observational studies of free-ranging birds, offer researchersopportunities to address questions that are unanswerable withfield work alone. Zoo populations of flamingos are prime candidatesfor such studies. Here, we use samples of our own work to illustrate the synergisticeffects of combining zoo and field research. Our first exampledescribes how studies of salt tolerance in captive birds areplaying a key role in assessing the impact of salt as an ecologicaldeterminant of flamingo distribution. Our second example describeshow aggression and dominance interactions affect the feedingbehavior of flamingos. We assess the implications of this researchin terms of both avicultural practices and the fundamental ecologyof the birds.
