Metastasis is the final stage of tumor progression and is thought to be responsible for up to 90% of deaths associated with solid tumors. Caveolin-1 (CAV1) regulates multiple cancer-associated processes related to malignant tumor progression. In the present study, we tested the hypothesis that CAV1 modulates the metastatic ability of cells from the Ewing's sarcoma family of tumors (ESFT). First, we analyzed the expression of CAV1 by immunostaining a tissue microarray containing 43 paraffin-embedded ESFT tumors with known EWS translocations. Even though no evidence was found for a significant association between CAV1 expression and stage, size or tumor site, all metastatic samples (10 of 10) had significantly high CAV1 expression, suggesting that high CAV1 content could positively contribute to enhance ESFT metastasis. To determine the effect of CAV1 on the migratory and invasive capabilities of ESFT cells, we knocked down CAV1 expression in TC252 and A673 cells by stably transfecting a previously validated shRNA construct. In vitro, migration and invasion assays showed that for both cell lines, CAV1 knocked-down cells migrated and invaded significantly less (P ≤ 0.01) than control cells. Moreover, control A673 cells introduced into BALB/c nude mice by tail vein injection strongly colonized the lungs. In contrast, animals injected with CAV1 knocked-down cells showed either no incidence of metastasis or developed lung metastases after a significant delay (P < 0.0001). Finally, we show that the molecular mechanisms by which CAV1 carries out its key role in regulating ESFT metastasis involve matrix metalloproteinase production and activation as well as the control of the expression of SPARC, a known determinant of lung colonization.  相似文献

目的:目前认为,十二指肠胆道反流是引起胆道反复感染,进而导致胆道结石再发和胆道狭窄的原因之一。近年来应用以内镜下逆行胆胰管造影术(endoscopicretrogradecholangiopancreatography,ERCP)为基础的微创治疗胆总管结束的技术开展颇为广泛。它主要包括ERCP、Oddi括约肌切开术(endoscopicsphincterotomy,EST)、十二指肠乳头球囊扩张术(endoscopicpapillarybal—Iondilation,EPBD)、胆管结石碎石取石术、胆总管支架植入术和鼻胆管引流术六大技术。本文主要研究了采用不同术式的EST,即EST中切口和EST小切口+EPBD术,在术后早期对患者十二指肠胆道反流的影响。方法:63例胆总管结石患者,男30例,女33例,予行经内镜下逆行的胆胰管造影(ERCP)后分别采用不同术式EST,术后安放胆总管引流管。术后l周留取胆汁采用口服核素和测定胆汁中的胃蛋白酶I、II的浓度,对十二指肠胆道反流进行定量和定性的测定。结果:EST中切口术组、EST小切口+球囊扩张(EPBD)组分别与无EST组相比,年龄和性别无统计学意义(P=0.07,P=0.416)。行EST中切开和小切开+球囊扩张患者胆汁中的锝计数明显高于无EST组,且这两组不同术式的患者锝计数存在显著的统计学差异(P〈0.05)。行EST中切口者、EST小切口+球囊扩张术者胆汁中的PGII质量浓度明显低于无EST组(P〈0.05),但是EST中切口者和EST小切口+球囊扩张术后两组间胆汁中PGII的质量浓度无统计学差异。结论:行EST中切口取胆总管结石的患者在手术早期较易发生十二指肠胆道的反流。因此,建议对于胆总管结石患者尽量选择行EST小切口+球裳扩张术(EPBD)的手术方式。  相似文献

Background aimsAdipose-derived stem cells (ASCs) are a promising mesenchymal cell source for tissue engineering approaches. To obtain an adequate cell amount, in vitro expansion of the cells may be required in some cases. To monitor potential contraindications for therapeutic applications in humans, DNA strand breaks and chromosomal aberrations in ASCs during in vitro expansion were examined.MethodsAfter isolation of ASC from human lipoaspirates of seven patients, in vitro expansion over 10 passages was performed. Cells from passages 1, 2, 3, 5 and 10 were used for the alkaline single-cell microgel electrophoresis (comet) assay to detect DNA single-strand breaks and alkali labile as well as incomplete excision repair sites. Chromosomal changes were examined by means of the chromosomal aberration test.ResultsDuring in vitro expansion, ASC showed no DNA single-strand breaks in the comet assay. With the chromosomal aberration test, however, a significant increase in chromosomal aberrations were detected.ConclusionsThe study showed that although no DNA fragmentation could be determined, the safety of ASC cannot be ensured with respect to chromosome stability during in vitro expansion. Thus, reliable analyses for detecting ASC populations, which accumulate chromosomal aberrations or even undergo malignant transformation during extensive in vitro expansion, must be implemented as part of the safety evaluation of these cells for stem cell–based therapy.  相似文献

A new species of Gesneriaceae from Honghe County, Southeastern Yunnan, China, Tremacron hongheense WH. Chen & YM. Shui, is described and illustrated. The new species is similar to Tremacron forrestii Craib, but differs by its leaf blade adaxially sparsely long setose (vs. densely white appressed pubescent and sparsely rusty brown villous), corolla tube outside short white glandular (vs. nearly glabrous), corolla lobes red and thickening at apex, especially adaxial lip (vs. yellow and not thickening), stamens 16-18cm long (vs. 04-12cm long), staminode 05-14cm long (vs. 02-04cm long).  相似文献

Background

Cerebral ischaemia initiates an inflammatory response in the brain and periphery. We assessed the relationship between peak values of plasma interleukin-6 (IL-6) in the first week after ischaemic stroke, with measures of stroke severity and outcome.

Methods

Thirty-seven patients with ischaemic stroke were prospectively recruited. Plasma IL-6, and other markers of peripheral inflammation, were measured at pre-determined timepoints in the first week after stroke onset. Primary analyses were the association between peak plasma IL-6 concentration with both modified Rankin score (mRS) at 3 months and computed tomography (CT) brain infarct volume.

Results

Peak plasma IL-6 concentration correlated significantly (p < 0.001) with CT brain infarct volume (r = 0.75) and mRS at 3 months (r = 0.72). It correlated similarly with clinical outcome at 12 months or stroke severity. Strong associations were also noted between either peak plasma C-reactive protein (CRP) concentration or white blood cell (WBC) count, and all outcome measures.

Conclusions

These data provide evidence that the magnitude of the peripheral inflammatory response is related to the severity of acute ischaemic stroke, and clinical outcome.  相似文献

To link spatial patterns and ecological processes, we analysed the distribution of two shrub species (one large and dominant, the other smaller) and estimated the reproductive consequences of their distribution for the smaller species. We tested the significance of the spatial distribution pattern of the two shrubs by second-order bivariate point pattern analysis (Ripley's K function). Performance of Asparagus albus, the smaller shrub, was measured as (1) survival of transplanted seedlings in two contrasting habitats: patches of the dominant shrub (Ziziphus lotus), and open interspaces; and (2) reproductive output of plants naturally occurring in both habitats. The two species were significantly aggregated. Transplanted Asparagus albus seedlings had higher survival rates in patches than in the open. Plants produced more flowers, fruits, and showed a higher mass of seeds when living in aggregates than when isolated. The mechanisms responsible for this facilitative effect seem to be related to soil enrichment in patches. These results suggest that the spatial aggregation of species can be indicative of a positive interaction among them, directly affecting fitness of at least one of the species. Facilitation, by inducing variations in the reproductive performance may play a major role in the demography and dynamics of plant populations.  相似文献
