In vitro metabolism or testosterone by breast microcysts

Breast microcysts are considered to be a normal findings in the adult female breast without any increased risk of developing carcinomatous change. Breast cysts fluid contains steroid but not studies have been reported on the ability of breast microcysts to metabolise steroid hormones. It was, therefore, the aim of this study to identify the metabolites formed on incubation of radiolabelled testosterone with microcysts. In all instances dihydrotestosterone and androstenedione were formed. Oestrogens were not identified. Tis study, therefore, provides evidence for th presence of 5-alpha-reductase and 17-oxidoreductase enzyme systems in breast microcysts.     

Analysis of a common inheritable idiotype in IgA-deficient sera using monoclonal antibodies

In these studies we describe the production of three mAb raised to an idiotype on an IgG anticasein antibody isolated from the serum of one IgA-deficient blood donor. These are IgM kappa and block the binding of casein Ag to anticasein antibody. Sera of unrelated IgA-deficient donors were tested for the presence of the idiotype; 15 of 56 IgA-deficient sera (25%) contain the anticasein idiotype, whereas 1 of 45 normal sera was positive. Anticasein antibodies as a whole were predominantly of the IgG1 and IgG3 subclass; idiotype-positive anticaseins are predominantly of the IgG1 subclass. For IgA-deficient donors, the relative amount of idiotype-positive anticasein antibody was correlated with the level of anticasein present in the serum. Studies were done to investigate the potential inheritance of the idiotype in families; in three of four families the idiotype was inherited in an apparent autosomal dominant pattern. Our data show that a common cross-reactive idiotype can be detected in the sera of IgA-deficient individuals and their family members. This suggests that V region markers may be conserved in this humoral immunodeficiency disease.     

Lethal Temperatures in Ammocoetes of Four Species of Lampreys

Abstract An investigation has been made of the resistance time and upper lethal temperature of ammocoetes of four species of lampreys provided with a substrate into which they could readily burrow. In general, ammocoetes burrowed after transfer from the acclimation to the experimental temperature baths and later came out of the substrate only in lethal temperatures. A relationship was observed between the resistance time and the time taken to emerge, with the resistance time increasing exponentially with decreasing experimental temperature. In Ichthyomyzon fossor, landlocked Petromyzon marinus, Lampetra (Lethenteron) Lamottenii and in Lampetra (Lampetra) planeri from two different times of the year, the incipient lethal levels over a two week experimental period for larvae acclimated to 15° G were respectively 30.5, 30, 29.5, 28.5 and 28° C. Values for P. marinus acclimated to 5 and 25° C were respectively 29.5 and 31° C, whereas in L. planeri they were 28 and 29° C in April/May and 27 and 29° C in July/August. Extrapolation of the results for the three acclimation temperatures yielded ultimate incipient lethal levels of 31.4° G in P. marinus and 29.2 and 29.4° C for L. planeri examined in the spring and summer respectively.     

Clomiphene citrate alters vaginal surface morphology in cycling rats.

Cycling virgin female rats were treated with clomiphene citrate (CC) during dioestrus of the reproductive cycle. Animals were sacrificed 2 days after the initial injection and their vaginal tissue was examined by scanning electron microscopy. Animals treated with one dose of CC had an epithelium consistent with a prolonged dioestrus. Treatment with CC for 2 days induced changes in the epithelium that had no resemblance to any normal hormonally controlled event in the vagina. It was found that CC had effects consistent with progesterone alone as well as effects unique to this superovulatory drug.     

Properties of the H-4-II-E tumor cell system. I. Growth and cell proliferation kinetics of an experimental hepatoma.

Growth and cell proliferation kinetics of hepatoma H-4-II-E and its tissue culture derivative have been studied to establish the characteristics of an in vivo--in vitro solid tumor model. The H-4-II-E line, originating from the Reuber H-35 hepatoma, can be maintained and studied either in cell culture or as a transplantable solid tumor in ACI male rats. In addition it allows for the in vitro assay of cell survival following treatment of animal tumors in situ. In vivo, hepatoma H-4-II-E is rapidly growing tumor with a mean doubling time of 49-2 hr. The cell cyle time is 39-1 hr with a cell loss factor of 0-32. Retrospective examination of tumor specimens obtained during the establishment of the H-4-II-E tumor system demonstrates that both structural as well as cell population changes have occurred. The biological characteristics of the primary tumor (H-35) and an early intermediate stage (H-35tc2) are compared with H-4-II-E and the histopathological, growth and cell kinetic changes are discussed.     

Existence of an ejaculatory-like reaction in ewes ovariectomized and treated with androgens in adulthood

The complete behavioral repertoire of male sexual activity can be observed during daily androgen treatment (testosterone propionate, 15 mg/day) of normal ewes ovariectomized as adults. This includes the ejaculatory pattern (deep thrust accompanied by a rapid backwards movement of the head) which is followed by a dramatic decrease in the frequency of sexual interactions, similar to the male's postejaculatory reduction of activity. However, the sexual performances of the genetic females remain lower than those of normal males in ejaculation latency, postejaculation latency, and mount/ejaculation ratio.
