A three-dimensional reconstruction from electron micrographs of negatively stained cell envelopes of Halobacterium volcanii has revealed the structure of the surface glycoprotein to a resolution of 2 nm. The glycoprotein is arranged on a p6 lattice with a lattice constant of 16.8 nm. It forms 4.5 nm high, dome-shaped, morphological complexes with a narrow pore at the apex opening into a `funnel' towards the cell membrane. The polarity of the structure was derived from freeze-etching experiments and `edge' views. Six radial protrusions emanate from each morphological complex and join around the 3-fold axis to provide lateral connectivity. Using the primary structure of the surface glycoprotein of the closely related species Halobacterium halobium (Lechner and Sumper, 1987) and the cell envelope profile from a previous X-ray analysis of the same species (Blaurock et al., 1976) we have integrated our reconstruction into a model of halobacterial cell envelope.  相似文献

In his essay, Buckle analyzes some of the distinct ways of arguing about potential, and the different senses of potential on which these arguments rely. Ternary systems comprising water (1), glycine (diglycine) (2) and alkylurea (3) have been investigated using vapor pressure osmometry. Equations were obtained in terms of the molalities of the solutes for the activity coefficients of glycine and diglycine in these systems. The alkylureas used were methyl-, ethyl- and N, N'-dimethylurea. Using the activity coefficients the Gibbs free energy of transfer at infinite dilution of component 2 from water to alkylurea solutions was determined. Since the enthalpies of transfer are known, the corresponding entropies could also be obtained. Calculation of the Gibbs free energy of transfer at infinite dilution of component 2 rests on the assumption that it can be divided into two parts: the difference between the Gibbs free energy of cavity formation and that of interaction in the alkylurea solution and water, respectively. The first part was calculated by scaled particle theory using experimental density and surface tension data. The second part was taken to be due mainly to the change in dipole-dipole interactions.  相似文献

An enzyme activity, found for the first time in plants, mainly located in the 22,000g supernatant of the crude extract of sprout apices of Helianthus tuberosus L. cv OB1 tubers, is able in vitro to covalently bind polyamines to endogenous substrates of different molecular weights. The major assay parameters, such as pH, dithiothreitol, and extract concentrations were optimized. The time course of the reaction, the dependence on putrescine concentration, its competition with histamine, the capacity to bind spermidine and spermine better than putrescine, the stability of the reaction product and analysis of the latter by sodium dodecyl sulfate polyacrylamide gel electrophoresis and thin-layer chromatography suggest that putrescine is linked to endogenous substrates by means of an enzyme reaction that shows some similarities with transglutaminase activities detected in animals. However, the activities of the crude extract and of a fraction eluted from a Sephadex G25 column were not affected by CaCl2 concentrations lower or equal to 5 millimolar; 4 or 10 millimolar EGTA caused a very small reduction; higher concentrations of CaCl2 and 15 millimolar or more of EDTA were inhibitory. N,N′-dimethylcasein was not recognized as a substrate. These data indicate that this activity also displays some characteristics which are different from those of animal transglutaminases.  相似文献

Panoistic ovarioles are found in the order of fleas (Siphonaptera). Only in some species of the Hystrichopsylloidea do polytrophic meroistic ovaries occur. No stem cells and no dividing cystocytes are found in female imagines of Hystrichopsylla talpae. However, each germ cell cluster consists of 32 cells which are generated by five mitotic cycles during the pupal stage. One of the cells containing five intercellular bridges becomes the oocyte, the others serve as nurse cells. Thus, germ cell cluster formation follows the 2(n)-rule. However, no polyfusome is found and nurse cells do not form a rosette. Furthermore, nurse cells remain small and show the same ultrastructural characters as the oocytes, which became distinguishable from nurse cells only by their enhanced growth during pre-vitellogenesis. The first phase of pre-vitellogenesis is dominated by the production of an unknown cytoplasmatic component, consisting of spherical particles, clearly distinguishable from ribosomes by diameter and contrast. The next phase is characterized by a tremendous increase in the production of ribosomes. During this second phase another cytoplasmic component consisting of ball-like structures becomes prominent. During pre-vitellogenesis, germ cell nuclei undergo a pronounced structural change in which, finally, numerous extranucleolar particles predominate. Thus, H. talpae has a polytrophic meroistic ovary, but its oocyte genomes behave panoistically.  相似文献
