Resource heterogeneity across the landscape prompts animals to make behavioral tradeoffs to survive and reproduce. Behavioral thermoregulation can buffer organisms from thermal extremes but may conflict with other essential activities such as predator avoidance or foraging, and necessitate tradeoffs among resource requirements. We evaluated patterns of habitat selection relative to thermal conditions, forage availability, and concealment cover for female eastern wild turkeys (Meleagris gallopavo silvestris) with broods to assess potential tradeoffs among resource requirements. We quantified air temperature (°C), vegetation characteristics (e.g., visual obstruction), and arthropod biomass (g/m2) at locations used by broods across 5 study sites in the southeastern United States during May–July 2019–2020. We used conditional logistic regression to estimate brooding female resource selection at the second (home range) and third (within home range) orders. Specifically, we identified differences in selection between brooding and non-brooding females (second order), and factors influencing selection of sites used by brooding females during the day (when loafing and foraging) and night (roosting; third order). Brooding females selected sites with cooler temperatures (β = −0.22; 95% CI = −0.338–−0.102) and greater ground cover vegetation (β = 0.02; 95% CI = 0.013–0.033) than non-brooding females. Additionally, biomass of large prey (Orthoptera) was positively related to ambient temperature, suggesting that use of thermal refuge by brooding females may limit availability of large prey. Brooding females appeared to balance the tradeoff between thermal refuge and forage availability by altering habitat selection patterns within home ranges. Brooding females selected for herbaceous areas that provided greater biomass of large arthropods during the day, and avoided areas dominated by woody vegetation during both the day and night. We did not observe brooding females using locations where woody cover exceeded 27% of understory vegetation. Thermal refuge is an important component of brood habitat, but within thermally suitable areas brooding females can select sites with greater availability of large prey to meet energetic demands of broods. Evaluation of multiple spatial scales is key when assessing tradeoffs among resource needs and determining the potential of behavioral thermoregulation to buffer an organism's thermal environment and allow persistence in a warming climate.  相似文献

It is widely assumed that the development of memory CD8 T cells requires the escape of CTLs from programmed cell death. We show in this study that although serine protease inhibitor 6 (Spi6) is required to protect clonal bursts of CTLs from granzyme B-induced programmed cell death, it is not required for the development of memory cells. This conclusion is reached because memory cell precursors down-regulate both Spi6 and granzyme B, unlike CTLs, and they do not require Spi6 for survival. These findings suggest that memory CD8 T cells are derived from progenitors that are refractory to self-inflicted damage, rather than derived from fully differentiated CTLs.  相似文献

Inflammation is a localized, protective response to trauma or microbial invasion that destroys the injurious agent and the injured tissue. Neutrophil elastase (NE), a serine protease stored in the azurophil granules of polymorphonuclear neutrophils, digests microbes after phagocytosis. NE can also digest microbes extracellularly but is associated with tissue damage and inflammatory disease. In this study, we show that polymorphonuclear neutrophils from mice deficient in serine protease inhibitor 6, a weak intracellular NE inhibitor, had increased susceptibility to self-inflicted lysis because of increased NE activity. The resulting transient increase in local extracellular NE activity was within a narrow range that resulted in the clearance of Pseudomonas aeruginosa but did not damage the lung. Therefore, deficiency in a weak intracellular inhibitor of NE results in an acute inflammatory response that protects from P. aeruginosa but does not cause lung disease.  相似文献

C57BL/6J mice were 10(5)-fold more resistant to Chlamydia psittaci infection than DBA/2J mice by LD(100) determinations. Linkage analysis using BXD recombinant inbred strains revealed a single effector locus at a 1.5-Mbp region on chromosome 11 encoding a cluster of three p47 GTPases (Irgb10, Igtp, and Iigp2). Western blots of infected tissue showed that Irgb10 was elevated in resistant mice and one of the two possible Iigp2 protein isoforms was preferentially expressed in susceptible mice. The BXD39 strain, susceptible at Irgb10 and resistant at Iigp2, had an intermediate phenotype implicating the nonredundant role of these p47 GTPases. C57BL/6J and DBA/2J exhibited a difference in IFN-gamma-dependent chlamydial control, which was reversible by Iigp2 small interfering RNA knockdown. Microarrays of infected peritoneal lavage revealed >10-fold up-regulation of neutrophil-recruiting chemokines in susceptible mice and >100-fold increase in macrophage differentiation genes in resistant mice, indicating that the susceptibility pattern involves the stimulation of different inflammatory cell-recruiting pathways. Massive neutrophil recruitment was seen in susceptible mice by histology and flow cytometry, and neutrophil chemokine receptor (CXCR2) knockout mice on a susceptible background survived a lethal challenge, confirming that neutrophil recruitment was required for susceptibility. Congenic Igtp knockout mice also susceptible at Irgb10 and Iigp2 on a resistant background recruited neutrophils and succumbed to infection. We conclude that Irgb10 and Iigp2 act together to confer differential susceptibility against murine chlamydial infection. Data indicate that these p47 GTPases have cell-autonomous effects that result in vastly different inflammatory stimulations, leading to either recovery or death.  相似文献

Abstract: We describe a method for rapidly amplifying whole genomes via a Phi29 DNA polymerase-mediated strand displacement reaction (SDR). Genomic amplification products derived from the SDR reaction resulted in high quantities of DNA suitable for polymerase chain reaction (PCR) amplification and sequencing of mitochondrial genomes. Control region sequences of DNA derived directly from PCR amplicons of extracted DNA were identical to those derived from PCR amplification of SDR genomic DNA. Effective SDR amplification and subsequent sequencing was successful across tissues sources ranging in age from 1 year to 19 years. Strand replacement reaction genomic amplification offers a means of obtaining large quantities of DNA from small amounts of tissue.  相似文献

In Great Britain and Ireland, badgers (Meles meles) are a wildlife reservoir of Mycobacterium bovis and implicated in bovine tuberculosis transmission to domestic cattle. The route of disease transmission is unknown with direct, so‐called “nose‐to‐nose,” contact between hosts being extremely rare. Camera traps were deployed for 64,464 hr on 34 farms to quantify cattle and badger visitation rates in space and time at six farm locations. Badger presence never coincided with cattle presence at the same time, with badger and cattle detection at the same location but at different times being negatively correlated. Badgers were never recorded within farmyards during the present study. Badgers utilized cattle water troughs in fields, but detections were infrequent (equivalent to one badger observed drinking every 87 days). Cattle presence at badger‐associated locations, for example, setts and latrines, were three times more frequent than badger presence at cattle‐associated locations, for example, water troughs. Preventing cattle access to badger setts and latrines and restricting badger access to cattle water troughs may potentially reduce interspecific bTB transmission through reduced indirect contact.  相似文献

Global climate is rapidly changing, and the ability for tree species to adapt is dependent on standing genomic variation; however, the distribution and abundance of functional and adaptive variants are poorly understood in natural systems. We test key hypotheses regarding the genetics of adaptive variation in a foundation tree: genomic variation is associated with climate, and genomic variation is more likely to be associated with temperature than precipitation or aridity. To test these hypotheses, we used 9,593 independent, genomic single‐nucleotide polymorphisms (SNPs) from 270 individuals sampled from Corymbia calophylla's entire distribution in south‐western Western Australia, spanning orthogonal temperature and precipitation gradients. Environmental association analyses returned 537 unique SNPs putatively adaptive to climate. We identified SNPs associated with climatic variation (i.e., temperature [458], precipitation [75] and aridity [78]) across the landscape. Of these, 78 SNPs were nonsynonymous (NS), while 26 SNPs were found within gene regulatory regions. The NS and regulatory candidate SNPs associated with temperature explained more deviance (27.35%) than precipitation (5.93%) and aridity (4.77%), suggesting that temperature provides stronger adaptive signals than precipitation. Genes associated with adaptive variants include functions important in stress responses to temperature and precipitation. Patterns of allelic turnover of NS and regulatory SNPs show small patterns of change through climate space with the exception of an aldehyde dehydrogenase gene variant with 80% allelic turnover with temperature. Together, these findings provide evidence for the presence of adaptive variation to climate in a foundation species and provide critical information to guide adaptive management practices.  相似文献

Organisms living in habitats characterized by a marked seasonal temperature variation often have a greater thermal tolerance than those living in more stable habitats. To determine the extent to which this hypothesis applies to reef corals, we compared thermal tolerance of the early life stages of five scleractinian species from three locations spanning 17° of latitude along the east coast of Australia. Embryos were exposed to an 8 °C temperature range around the local ambient temperature at the time of spawning. Upper thermal thresholds, defined as the temperature treatment at which the proportion of abnormal embryos or median life span was significantly different to ambient controls, varied predictably among locations. At Lizard Island, the northern-most site with the least annual variation in temperature, the proportion of abnormal embryos increased and life span decreased 2 °C above ambient in the two species tested. At two southern sites, One Tree Island and Lord Howe Island, where annual temperature variation was greater, upper temperature thresholds were generally 4 °C or greater above ambient for both variables in the four species tested. The absolute upper thermal threshold temperature also varied among locations: 30 °C at Lizard Island; 28 °C at One Tree Island; 26 °C at Lord Howe Island. These results support previous work on adult corals demonstrating predictable differences in upper thermal thresholds with latitude. With projected ocean warming, these temperature thresholds will be exceeded in northern locations in the near future, adding to a growing body of evidence indicating that climate change is likely to be more detrimental to low latitude than high latitude corals.  相似文献

Marine organisms are simultaneously exposed to anthropogenic stressors with likely interactive effects, including synergisms in which the combined effects of multiple stressors are greater than the sum of individual effects. Early life stages of marine organisms are potentially vulnerable to the stressors associated with global change, but identifying general patterns across studies, species and response variables is challenging. This review represents the first meta‐analysis of multistressor studies to target early marine life stages (embryo to larvae), particularly between temperature, salinity and pH as these are the best studied. Knowledge gaps in research on multiple abiotic stressors and early life stages are also identified. The meta‐analysis yielded several key results: (1) Synergistic interactions (65% of individual tests) are more common than additive (17%) or antagonistic (17%) interactions. (2) Larvae are generally more vulnerable than embryos to thermal and pH stress. (3) Survival is more likely than sublethal responses to be affected by thermal, salinity and pH stress. (4) Interaction types vary among stressors, ontogenetic stages and biological responses, but they are more consistent among phyla. (5) Ocean acidification is a greater stressor for calcifying than noncalcifying larvae. Despite being more ecologically realistic than single‐factor studies, multifactorial studies may still oversimplify complex systems, and so meta‐analyses of the data from them must be cautiously interpreted with regard to extrapolation to field conditions. Nonetheless, our results identify taxa with early life stages that may be particularly vulnerable (e.g. molluscs, echinoderms) or robust (e.g. arthropods, cnidarians) to abiotic stress. We provide a list of recommendations for future multiple stressor studies, particularly those focussed on early marine life stages.  相似文献
