Background

Ionized calcium (Ca) and magnesium (Mg) compete as essential messengers to regulate cell proliferation and inflammation. We hypothesized that inadequate Mg levels, perhaps relative to Ca levels (e.g. a high Ca/Mg ratio) are associated with greater prostate cancer risk.

Study Design

In this biomarker sub-study of the Nashville Men''s Health Study (NMHS), we included 494 NMHS participants, consisting of 98 high-grade (Gleason≥7) and 100 low-grade cancer cases, 133 prostate intraepithelial neoplasia (PIN) cases, and 163 controls without cancer or PIN at biopsy. Linear and logistic regression were used to determine associations between blood Ca, Mg, and the Ca/Mg ratio across controls and case groups while adjusting for potential confounding factors.

Results

Serum Mg levels were significantly lower, while the Ca/Mg ratio was significantly higher, among high-grade cases vs. controls (p = 0.04, p = 0.01, respectively). Elevated Mg was significantly associated with a lower risk of high-grade prostate cancer (OR = 0.26 (0.09, 0.85)). An elevated Ca/Mg ratio was also associated with an increased risk of high-grade prostate cancer (OR = 2.81 (1.24, 6.36) adjusted for serum Ca and Mg). In contrast, blood Ca levels were not significantly associated with prostate cancer or PIN.Mg, Ca, or Ca/Mg levels were not associated with low-grade cancer, PIN, PSA levels, prostate volume, or BPH treatment.

Conclusion

Low blood Mg levels and a high Ca/Mg ratio were significantly associated with high-grade prostate cancer. These findings suggest Mg affects prostate cancer risk perhaps through interacting with Ca.  相似文献

The Aspergillus fumigatus chsE (AfchsE) gene was isolated from an A. fumigatus DNA library on the basis of hybridization to a segment of Saccharomyces cerevisiae CHS3 (ScCHS3). The amino acid sequence derived from AfchsE is 28% identical with ScCHS3 and 80% identical with the product of Aspergillus nidulans chsD (AnchsD). A mutant strain constructed by disruption of AfchsE has reduced levels of mycelial chitin, periodic swellings along the length of hyphae, and a block in conidiation that can be partially restored by growth in osmotic stabilizer. This phenotype is different from that reported for an AnchsD mutant, in which germinating conidia and hyphal tips undergo lysis and the colonial growth rate is significantly reduced. Despite the defects associated with the AfchsE- strain, its virulence was not significantly reduced when compared with the wild-type parental strain in a mouse model of pulmonary aspergillosis.  相似文献

Renal-coloboma syndrome is a recently described autosomal dominant syndrome of abnormal optic nerve and renal development. Two families have been reported with renal-coloboma syndrome and mutations of the PAX2 gene. The PAX2 gene, which encodes a DNA-binding protein, is expressed in the developing ear, CNS, eye, and urogenital tract. Ocular and/or renal abnormalities have been consistently noted in the five reports of patients with renal-coloboma syndrome, to date, but PAX2 expression patterns suggest that auditory and CNS abnormalities may be additional features of renal-coloboma syndrome. To determine whether additional clinical features are associated with PAX2 mutations, we have used PCR-SSCP to identify PAX2 gene mutations in patients. We report here four patients with mutations in exon 2, one of whom has severe ocular and renal disease, microcephaly, and retardation, and another who has ocular and renal disease with high-frequency hearing loss. Unexpectedly, extreme variability in clinical presentation was observed between a mother, her son, and an unrelated patient, all of whom had the same PAX2 mutation as previously described in two siblings with renal-coloboma syndrome. These results suggest that a sequence of seven Gs in PAX2 exon 2 may be particularly prone to mutation.  相似文献

The suction pipet method of intracellular dialysis and voltage clamp of cells has proven extremely useful in analysing the electrical properties of cells too small for the application of conventional microelectrode techniques and in larger cells for studying the effects of alterations in the internal ionic composition. Using neurons of the snail Lymnaea stagnalis, we have analysed several problems involved in the latter application of this technique and present several solutions to them. One major problem centers around the degree of control over the ionic composition of the cytoplasm achieved by altering the pipet solution. Using ion-sensitive microelectrodes during internal dialysis, we found that the efficiency of exchange between pipet and cytoplasm was much poorer for highly buffered ions such as H+ and Ca2+, than for K+, for example. Special precautions are described that can help this situation. The second problem involves the study of the effects of low internal pH on ion-channel properties. We summarize evidence for a specific voltage-dependent hydrogen ion channel, current through which becomes prominent at low internal pH. We analyse how the presence of this heretofore unrecognized current can seriously confuse the results of experiments designed to study the effects of low internal pH on other voltage-dependent currents.  相似文献

An adult, captive European spotted fallow deer (Cervus dama) was submitted for necropsy due to sudden death. Gross lesions consisted of serosanguinous fluid in the thoracic cavity with multiple, often confluent, nodules covering visceral and parietal pleura. Microscopic examination revealed tubular structures lined by cuboidal cells covering a delicate fibrous stroma. Gross and microscopic morphology was consistent with a mesothelioma.  相似文献

We argue that the existence of species as distinct and relatively homogeneous groupings of individuals is a consequence of the nonlinear dynamics inherent in sexual reproduction. This approach provides an answer to two interrelated problems which Darwin posed and tried to solve. Why are there missing links (i.e. gaps) between species in habitat space, and why are there missing links between species in time as evidenced in the fossil record? A crucial difference between outcrossing sexual organisms (i.e. organisms in which mating is between different individuals) and obligate selfers or parthenogens lies in the dynamic of the underlying replication process. Replication is a linear function of density for obligate selfers or parthenogens but nonlinear for outcrossing sexuals. The non-linearity stems from the simple fact that with outcrossing, two individuals must come together to mate. We argue that this fact leads to density dependent fitness (per capita rate of increase) with an intrinsic disadvantage of low population density. This cost of rarity results in a distribution of distinct species. By establishing the causal connections in evolution between outcrossing sex and the very existence of species as distinct collections of organisms, our account lends theoretical support to a unitary concept of species with interbreeding as the fundamental defining property.  相似文献
