The multfactorial nature of bone injuries in modern warfare and emergency trauma patients warrants enhancement of existing models. To develop a more appropriate model, rat tibiae (n = 195) were mechanically injured, divided into 2 groups (with or without thermal injury), and contaminated with a range of Staphylococcus aureus (Cowan 1) inocula. In some experiments, S. aureus inocula also contained Escherichia coli or foreign bodies (sand or soil). The primary outcome measure was the amount of S. aureus remaining in the tibia (tibial bacterial load) 24 h after contamination, reported as log10 cfu/g bone. S. aureus showed ID50 and ID95 values of 72 and 977 cfu, respectively. Values were lower than seen previously by using S. aureus strain SMH. S. aureus tibial bacterial loads were higher in tibiae with mechanical and thermal injury (log10 4.15 +/- 0.27 cfu/g) versus mechanical injury alone (log10 3.1 +/- 0.47 cfu/g, P = 0.028). The addition of E. coli to the S. aureus inoculum had no effect on tibial bacterial loads (S. aureus only, log10 4.24 +/- 0.92 cfu/g; S. aureus + E. coli, log10 4.1 +/- 1.0 cfu/g, P = 0.74). Sand, added as a foreign body, increased tibial bacterial load. Combined mechanical and thermal trauma of the tibia is associated with increased S. aureus tibial bacterial loads, increasing the risk of acute osteomyelitis. Understanding the interplay of mechanical and thermal injuries, bimicrobial contamination, and foreign bodies may improve our understanding of traumatic bone injuries and the pathogenesis of osteomyelitis.  相似文献

A growing body of evidence suggests that the angiotensin II fragments, Ang(1-7) and Ang(3-8), have a vasoactive role, however ACE2, the enzyme that produces Ang(1-7), or AT4R, the receptor that binds Ang (3-8), have yet been simultaneously localised in both normal and diseased human conduit blood vessels. We sought to determine the immunohistochemical distribution of ACE2 and the AT4R in human internal mammary and radial arteries from patients undergoing coronary artery bypass surgery. We found that ACE2 positive cells were abundant in both normal and diseased vessels, being present in neo-intima and in media. ACE2 positive immunoreactivity was not present in the endothelial layer of the conduit vessels, but was clearly evident in small newly formed angiogenic vessels as well as the vaso vasorum. Endothelial AT4R immunoreactivity were rarely observed in either normal and diseased arteries, but AT4R positive cells were observed adjacent to the internal elastic lamine in the internal mammary artery, in the neo-intima of radial arteries, as well as in the media of both internal mammary artery and radial artery. AT4R was abundant in vaso vasorum and within small angiogenic vessels. Both AT4R and ACE2 co-localised with smooth muscle cell alpha actin. This study identifies smooth muscle cell alpha actin positive ACE2 and AT4R in human blood vessels as well as in angiogenic vessels, indicating a possible role for these enzymes in pathological disease.  相似文献

We tested if plant species invasiveness (measured as rate of geographic spread in New Zealand) was positively associated with seedling relative growth rate (RGR) and survival, among 33 naturalized woody plant species in four families (Fabaceae, Mimosaceae, Pinaceae, Rosaceae). Eight disturbance and fertilization treatments were applied to seedlings in pots to mimic conditions the species are likely to encounter in the wild. We tested if seedlings of more invasive species have higher RGR and greater survival under these conditions. We also tested whether more invasive species had wider tolerance of the conditions to which they were subjected. There were no significant relationships between seedling RGR or survival and invasiveness for any of the treatments. Measures of tolerance (a species' relative performance across all treatments), based on both RGR and survival, also failed to explain differences in invasiveness. Our findings contrast with those from a recent study by Grotkopp et al. (2002) showing a positive correlation between seedling RGR and invasiveness in Pinus species, even though our study included 12 Pinus species. The findings of the two studies may differ because they focus on different stages in the invasion process. Our study tested whether RGR (and survival) was associated with the rate of spread of invasive species following naturalization, whereas Grotkopp et al. primarily tested whether RGR was associated with the probability of naturalization. We caution that relationships between RGR and measures of invasiveness are likely to be confounded with human introduction effort. Among pines, species with high RGR are more likely to have been widely cultivated, for purposes such as timber production, providing more opportunities for naturalization and spread. Associations between RGR, invasiveness and human introduction effort need to be partitioned out before we can reliably infer causal relationships.  相似文献

In comparison to the northern hemisphere, Sphagnum peatlands are an unusual andinfrequent component of the Australasianlandscape. Most peatlands in Australasiaare primarily composed of eitherRestionaceous or Cyperaceous peats. Sphagnum peatlands in Australia and PapuaNew Guinea/Irian Jaya (now West Papua) arelargely located in montane and alpineenvironments, but also occur down to sealevel in New Zealand and as moss patches onsome subantarctic islands. Fire is a majordeterminant of the characteristics ofpeatlands in Australasia. Peatlandmanagement in Australasia is hindered bythe need for increased understanding ofpeatland processes to enable a sustainablebalance of conservation of a small resourcewith localised utilisation. Themanagement focus in Australasia has largelybeen on ensuring ecologically sustainable Sphagnum moss harvesting, withlimited peat mining. We have found thatgeneral recovery of Sphagnum after moss harvesting canbe enhanced by harvesting larger peatlands,and by leaving one-third of the acrotelm toregenerate. The largest upland peat swampin mainland Australia, Wingecarribee Swamp,suffered a major collapse in 1998 followingpeat mining. Environmental and managementconsequences of this collapse have majorramifications for rehabilitation options. Sphagnum peatlands in Australasia arelikely to be adversely affected bydrainage, burning, grazing, trampling,global warming and peat mining.  相似文献

Cicer milkvetch (Astragalus cicer L.) is a perennial legume used as a pasture or rangeland plant for ruminants. A study was undertaken to determine whether reported variations in its ruminal digestibility may be related to the presence of an antinutritive material. In vitro fermentation of neutral detergent fiber (NDF) of cicer milkvetch by mixed rumen microflora was poorer than was the fermentation of NDF in alfalfa (Medicago sativa L.). Fermentation of cicer milkvetch NDF was improved by preextraction of the ground herbage with water for 3 h at 39 degrees C. Such water extracts selectively inhibited in vitro fermentation of pure cellulose by mixed ruminal microflora and by pure cultures of the ruminal bacteria Ruminococcus flavefaciens FD-1 and Fibrobacter succinogenes S85. Inhibition of the cellulose fermentation by mixed ruminal microflora was dependent upon the concentration of cicer milkvetch extract and was overcome upon prolonged incubation. Pure cultures exposed to the extract did not recover from inhibition, even after long incubation times, unless the inhibitory agent was removed (viz., by dilution of inhibited cultures into fresh medium). The extract did not affect the fermentation of cellobiose by R. flavefaciens but did cause some inhibition of cellobiose fermentation by F. succinogenes. Moreover, the extracts did not inhibit hydrolysis of crystalline cellulose, carboxymethyl cellulose, or p-nitrophenylcellobioside by supernatants of these pure cultures of cellulolytic bacteria or by a commercial cellulase preparation from the fungus Trichoderma reesei. The agent caused cellulose-adherent cells to detach from cellulose fibers, suggesting that the agent may act, at least in part, by disrupting the glycocalyx necessary for adherence to, and rapid digestion of, cellulose.  相似文献
