Background
We tested the hypothesis that the stretch-activated, four-transmembrane domain, two pore potassium channels (K2P), TREK-1 and TRAAK are gestationally-regulated in human myometrium and contribute to uterine relaxation during pregnancy until labor.Methodology
We determined the gene and protein expression of K2P channels in non-pregnant, pregnant term and preterm laboring myometrium. We employed both molecular biological and functional studies of K2P channels in myometrial samples taken from women undergoing cesarean delivery of a fetus.Principal Findings
TREK-1, but not TREK-2, channels are expressed in human myometrium and significantly up-regulated during pregnancy. Down-regulation of TREK-1 message was seen by Q-PCR in laboring tissues consistent with a role for TREK-1 in maintaining uterine quiescence prior to labor. The TRAAK channel was unregulated in the same women. Blockade of stretch-activated channels with a channel non-specific tarantula toxin (GsMTx-4) or the more specific TREK-1 antagonist L-methionine ethyl ester altered contractile frequency in a dose-dependent manner in pregnant myometrium. Arachidonic acid treatment lowered contractile tension an effect blocked by fluphenazine. Functional studies are consistent with a role for TREK-1 in uterine quiescence.Conclusions
We provide evidence supporting a role for TREK-1 in contributing to uterine quiescence during gestation and hypothesize that dysregulation of this mechanism may underlie certain cases of spontaneous pre-term birth. 相似文献102.

Measuring restoration outcomes is essential, but challenging and expensive, particularly on remote islands. Acoustic recording increases the scale of monitoring inexpensively; however, extracting biological information from large volumes of recordings remains challenging. Soundscape approaches, characterizing communities using acoustic indices, rapidly analyze large acoustic datasets and can be used to compare restoration sites against reference conditions. We tested this approach to measure nocturnal seabird recovery following invasive predator removal in the Aleutian Islands. We used recordings of nocturnal seabird soundscapes from six islands with varied histories of predators, from never invaded (one island) to 9–34 years post‐predator removal (four islands) and currently invaded (one island). We calculated 10 indices of acoustic intensity and complexity, and two pairwise indices of acoustic differences. Three indices reflected patterns of seabird recovery. Acoustic richness (measuring temporal entropy and amplitude) increased with time since predator removal and presence of historical predator refugia (r2 = 0.44). These factors and moonlight accounted for 30% of variation in cumulative spectral difference from the reference island. Over 10% of acoustic richness and temporal entropy was explained by Leach's Storm‐petrel (Oceanodroma leucorhoa) calls. However, indices characterized the soundscape of rat‐invaded Kiska Island like a never invaded island, likely due to high abiotic noise and few seabird calls. Soundscape indices have potential to monitor outcomes of seabird restoration quickly and cheaply, if confounding factors are considered and controlled in experimental design. We suggest soundscape indices become part of the expanding acoustic monitoring toolbox to cost‐effectively measure restoration outcomes at scale and in remote areas. 相似文献

In this study, temporal and spatial distribution of three TGF‐β isoforms and their downstream signaling pathways including pSmad2 and p38MAPK were examined during fibrotic wound repair. In normal chick corneas, TGF‐β1, ‐2, and ‐3 were weakly detected in Bowman's layer (BL). In healing corneas, TGF‐β1 was primarily deposited in the fibrin clot and the unwounded BL. TGF‐β2 was highly expressed in healing epithelial and endothelial cells, and numerous active fibroblasts/myofibroblasts. TGF‐β3 was mainly detected in the unwound region of basal epithelial cells. α‐Smooth muscle actin (α‐SMA) was initially appeared in the posterior region of repairing stroma at day 3, and was detected in the entire healing stroma by day 7. Notably, α‐SMA was absent in the central region of healing stroma by day 14, and its staining pattern was similar to those of TGF‐β2 and p38MAPK. By contrast, pSmad2 was mainly detected in the fibroblasts. In normal cornea, laminin was mainly detected in both epithelial basement membrane (BM) and Descemet's membrane (DM). By contrast to reconstitution of the BM in the wound region, the DM was not repaired although endothelial layer was regenerated, indicating that high levels of TGF‐β2 were released into the posterior region of healing stroma on day 14. High levels of α‐SMA staining, shown in cultured repair stromal cells from healing corneas on day 14 and in TGF‐β2 treated normal stromal cells, were significantly reduced by p38MAPK inhibition. Collectively, this study suggests that TGF‐β2‐mediated myofibroblast transformation is mediated, at least partly, by the p38MAPK pathway in vivo. J. Cell. Monitoring patterns in biodiversity and phenology have become increasingly important given accelerating levels of anthropogenic change. Long‐term monitoring programs have reported earlier occurrence of spring activity, reflecting species response to climate change. Although tracking shifts in spring migration represents a valuable approach to monitoring community‐level consequences of climate change, robust long‐term observations are challenging and costly. Audio recordings and metrics of bioacoustic activity could provide an effective method for monitoring changes in songbird activity and broader biotic interactions. We used 3 years of spring and fall recordings at six sites in Glacier Bay National Park, Alaska, an area experiencing rapid warming and glacial retreat, to examine the utility of bioacoustics to detect changes in songbird phenology. We calculated the Acoustic Complexity Index (ACI), an algorithm representing an index of bird community complexity. Abrupt changes in ACI values from winter to spring corresponded to spring transition, suggesting that ACI may be an effective, albeit coarse metric to detect the arrival of migrating songbirds. The first peak in ACI shifted from April 16 to April 11 from 2012 to 2014. Changes in ACI were less abrupt in the fall due to weather events, suggesting spring recordings are better suited to indicate phenology. To ensure changes in ACI values were detecting real changes in songbird activity, we explored the relationship between ACI and song of three species: varied thrush (Ixoreus naevius), Pacific wren (Troglodytes pacificus), and ruby‐crowned kinglet (Regulus calendula). ACI was positively related to counts of all species, but most markedly with song of the varied thrush, the most common species in our recordings and a known indicator of forest ecosystem health. We conclude that acoustic recordings paired with bioacoustic indices may be a useful method of monitoring shifts in songbird communities due to climate change and other sources of anthropogenic disturbance. 相似文献

Aspergillus niger produces several polygalacturonases that, with other enzymes, are involved in the degradation of pectin. One of the two previously characterized genes coding for the abundant polygalacturonases I and II (PGI and PGII) found in a commercial pectinase preparation was used as a probe to isolate five more genes by screening a genomic DNA library in phage lambda EMBL4 using conditions of moderate stringency. The products of these genes were detected in the culture medium of Aspergillus nidulans transformants on the basis of activity measurements and Western-blot analysis using a polyclonal antibody raised against PGI. These transformants were, with one exception, constructed using phage DNA. A. nidulans transformants secreted high amounts of PGI and PGII in comparison to the previously characterized A. niger transformants and a novel polygalacturonase (PGC) was produced at high levels by A. nidulans transformed with the subcloned pgaC gene. This gene was sequenced and the protein-coding region was found to be interrupted by three introns; the different intron/exon organization of the three sequenced A. niger polygalacturonase genes can be explained by the gain or loss of two single introns. The pgaC gene encodes a putative 383-amino-acid prepro-protein that is cleaved after a pair of basic amino acids and shows approximately 60% amino acid sequence similarity to the other polygalacturonases in the mature protein. The N-terminal amino acid sequences of the A. niger polygalacturonases display characteristic amino acid insertions or deletions that are also observed in polygalacturonases of phytopathogenic fungi. In the upstream regions of the A. niger polygalacturonase genes, a sequence of ten conserved nucleotides comprising a CCAAT sequence was found, which is likely to represent a binding site for a regulatory protein as it shows a high similarity to the yeast CYC1 upstream activation site recognized by the HAP2/3/4 activation complex. 相似文献

To examine the immediate phase-shifting effects of high-intensity exercise of a practical duration (1 h) on human circadian phase, five groups of healthy men 20-30 yr of age participated in studies involving no exercise or exposure to morning, afternoon, evening, or nocturnal exercise. Except during scheduled sleep/dark and exercise periods, subjects remained under modified constant routine conditions allowing a sleep period and including constant posture, knowledge of clock time, and exposure to dim light intensities averaging (+/-SD) 42 +/- 19 lx. The nocturnal onset of plasma melatonin secretion was used as a marker of circadian phase. A phase response curve was used to summarize the phase-shifting effects of exercise as a function of the timing of exercise. A significant effect of time of day on circadian phase shifts was observed (P < 0.004). Over the interval from the melatonin onset before exercise to the first onset after exercise, circadian phase was significantly advanced in the evening exercise group by 30 +/- 15 min (SE) compared with the phase delays observed in the no-exercise group (-25 +/- 14 min, P < 0.05). Phase shifts in response to evening exercise exposure were attenuated on the second day after exercise exposure and no longer significantly different from phase shifts observed in the absence of exercise. Unanticipated transient elevations of melatonin levels were observed in response to nocturnal exercise and in some evening exercise subjects. Taken together with the results from previous studies in humans and diurnal rodents, the current results suggest that 1) a longer duration of exercise exposure and/or repeated daily exposure to exercise may be necessary for reliable phase-shifting of the human circadian system and that 2) early evening exercise of high intensity may induce phase advances relevant for nonphotic entrainment of the human circadian system. 相似文献

Two tomato (Lycopersicon esculentum) mutants with dark testae displaying poor germination rate and percentage on both water and 100 microM gibberellin(4 + 7) were recovered. The mutants were allelic (black seed1-1; bks1-1 and bks1-2), inherited in Mendelian fashion as a recessive gene residing on chromosome 11. They are not allelic to bs (brown seed) -1, -2, or -4, which impair seed germination and possess dark testae. The bks/bs mutants accumulated dark pigment in the cell layers of the testa above the endothelium, which itself accumulated proanthocyanidins similar to wild type. The poor germination performance of bks mutant seeds was because of impediment of the mutant testae to radicle egress. Imbibition on gibberellin(4 + 7) did not ameliorate germination percentage or rate. The toughening of the bks testa and associated poor germination were partially overcome when seeds were not dried before germination or were dried under N(2). The seeds of the bks mutant have elevated activity of at least one enzyme responsible for the detoxification of reactive oxygen species. The bks mutant is epistatic to 12 anthocyaninless mutants of tomato. Bio- and physicochemical analysis of the bks testa determined that it accumulated a melanic substance. Inheritance of bks/bs mutations contrasts with that of the anthocyaninless mutants, which are inherited according to the genotype of the maternally derived testa. This suggests that the testa manufactures components before its demise that can maximize testa strength, whereas the endosperm/embryo produces factors that are conveyed to the testa, mitigating this process. 相似文献
