The obligate intracellular bacterium Coxiella burnetii causes the zoonotic disease Q‐fever. Coxiella pathogenesis depends on a functional type IV secretion system (T4SS). The T4SS effector AnkG inhibits pathogen‐induced host cell apoptosis, which is believed to be important for the establishment of a persistent infection. However, the mode of action of AnkG is not fully understood. We have previously demonstrated that binding of AnkG to p32 is crucial for migration of AnkG into the nucleus and that nuclear localization of AnkG is essential for its anti‐apoptotic activity. Here, we compared the activity of AnkG from the C. burnetii strains Nine Mile and Dugway. Although there is only a single amino acid exchange at residue 11, we observed a difference in anti‐apoptotic activity and nuclear migration. Mutation of amino acid 11 to glutamic acid, threonine or valine results in AnkG mutants that had lost the anti‐apoptotic activity and the ability to migrate into the nucleus. We identified Importin‐α1 to bind to AnkG, but not to the mutants and concluded that binding of AnkG to p32 and Importin‐α1 is essential for migration into the nucleus. Also during Coxiella infection binding of AnkG to p32 and Importin‐α1 is crucial for nuclear localization of AnkG.  相似文献

The aim of the study was to determine tolerance of plant growth promoting rhizobacteria (PGPR) in different concentrations of Cu, Cr, Co, Cd, Ni, Mn, and Pb and to evaluate the PGPR-modulated bioavailability of different heavy metals in the rhizosphere soil and wheat tissues, grown in saline sodic soil. Bacillus cereus and Pseudomonas moraviensis were isolated from Cenchrus ciliaris L. growing in the Khewra salt range. Seven-day-old cultures of PGPR were applied on wheat as single inoculum, co-inoculation and carrier-based biofertilizer (using maize straw and sugarcane husk as carrier). At 100 ppm of Cr and Cu, the survival rates of rhizobacteria were decreased by 40%. Single inoculation of PGPR decreased 50% of Co, Ni, Cr and Mn concentrations in the rhizosphere soil. Co-inoculation of PGPR and biofertilizer treatment further augmented the decreases by 15% in Co, Ni, Cr and Mn over single inoculation except Pb and Co where decreases were 40% and 77%, respectively. The maximum decrease in biological concentration factor (BCF) was observed for Cd, Co, Cr, and Mn. P. moraviensis inoculation decreases the biological accumulation coefficient (BAC) as well as translocation factor (TF) for Cd, Cr, Cu Mn, and Ni. The PGPR inoculation minimized the deleterious effects of heavy metals, and the addition of carriers further assisted the PGPR.  相似文献

The ultrastructure of the corpuscles of Stannius (CS) ofHeteropneustes fossilis reveals a homogenous cellular composition characterized by only one cell type, with large secretory granules and abundant ribosomal endoplasmic reticulum. These cells are comparable to the type 1 cell described in the CS of other teleosts; type 2 cells, whose presence is ubiquitous in the CS of freshwater species are absent inH. fossilis. Our data on the CS ofH. fossilis demonstrate that not all freshwater species possess type 2 cells in their CS and these are not essential for life in freshwater.  相似文献

The current work was performed to know the impact of cadmium (Cd) toxicity on two different genotypes of chickpea (Cicer arietinum L.) namely Pusa-BG1053 and Pusa-BG372. Cadmium was applied in the form of cadmium chloride (CdCl2), in varying levels, 0, 25, 50, 75, and 100 mg Cd kg-1 soil. Plant growth as well as physiological attributes were decreased with increasing concentration of Cd. Both genotypes showed the maximum and significant reduction at the maximum dose of Cd (100 mg Cd kg-1 soil). Results of this study proved that the genotype Pusa-BG1053 was more tolerant and showed a lower decline in growth, photosynthetic and biochemical attributes than Pusa-BG372. This later genotype showed the maximum reduction and was sensitive to Cd stress. A better activity of antioxidants protected Pusa-BG1053 from Cd toxicity; on the other hand, the activity of antioxidants was much lower in Pusa-BG372. Scanning electron microscopic studies showed differences in both genotypes. In Pusa-BG1053, stomatal quantity was higher and stomata were slightly close to the characteristic guard cells. In Pusa-BG372 stomata were lower, slightly open and with highly affected guard cells. Root cell mortality due to the harsh effects of Cd appeared to be more evident in Pusa-BG372 than Pusa-BG1053, which was visible under a confocal microscope. As a result of this study, Pusa-BG1053 was a more tolerant genotype, and exhibited a minimum reduction in terms of all studied parameters than Pusa-BG372, which was a sensitive genotype to Cd toxicity.

Lectin‐like oxidized low‐density lipoprotein receptor‐1 (LOX‐1) is a canonical receptor for oxidized LDL (oxLDL) among the known modified LDL particles. Topographical variance on LOX‐1 expression in different cell types and its influence on the atherogenic potential of the particular cell type is the main focus of this review. Characteristic features of LOX‐1 on the atherogenic potential of aortic endothelial cells, macrophages, platelets, and vascular smooth muscle cells have been discussed. Nonspecificity of ligands, besides oxLDL, is also the highlight of this review to show the chameleon characteristics in the functional activity of the receptor protein. Induction of LOX‐1 has been reported in diseases like atherosclerosis, diabetes, and hypertension, as well as in the inflammatory response of immune reactions. The expression of LOX‐1 is upregulated by the vicious cycle of stimulatory response from proatherogenic molecules.  相似文献

Since COVID‐19 took a strong hold around the globe causing considerable morbidity and mortality, a lot of effort was dedicated to manufacturing effective vaccines against SARS‐CoV‐2. Many questions have since been raised surrounding the safety of the vaccines, and a lot of media attention to certain side effects. This caused a state of vaccine hesitancy that may prove problematic in the global effort to control the virus. This review was undertaken with the aim of putting together all the reported cardiovascular and haematological events post COVID‐19 vaccination in published literature and to suggest possible mechanisms to explain these rare phenomena.  相似文献

The synthesis of novel N-heterocyclic carbene complexes derived from a tripeptide ligand (L), containing non-natural amino acid, thiazolylalanine is described here. The peptide ligand was reacted with suitable precursors to generate gold and mercury carbene complexes. The plausible structures of both complexes were predicted by spectroscopic data and DFT calculations. The binding energy data was also analyzed to predict their stability. The gold carbene complex (1A), showed activity against MCF7 breast cancer cell line due to mitochondrial triggered caspase-3 mediated programmed cell death. Its internalization inside cells could be observed due to autofluorescence. This study affords a methodology for successful generation of peptide carbene complexes for their therapeutic potential.  相似文献

Background

Hyperhomocysteinemia (>15 µmol/L) is highly prevalent in South Asian populations including Pakistan. In order to investigate the genetic determinants of this condition, we studied 6 polymorphisms in genes of 3 enzymes - methylenetetrahydrofolate reductase (MTHFR; C677T; A1298C), methionine synthase (MS; A2756G), cystathionine-β-synthase (CBS; T833C/844ins68, G919A) involved in homocysteine metabolism and investigated their interactions with nutritional and environmental factors in a Pakistani population.

Methodology/Principal Findings

In a cross-sectional survey, 872 healthy adults (355 males and 517 females; age 18–60 years) were recruited from a low-income urban population in Karachi. Fasting venous blood was obtained and assessed for plasma/serum homocysteine; folate, vitamin B12, pyridoxal phosphate and blood lead. DNA was isolated and genotyping was performed by PCR-RFLP (restriction-fragment-length- polymorphism) based assays. The average changes in homocysteine levels for MTHFR 677CT and TT genotypes were positive [β(SE β), 2.01(0.63) and 16.19(1.8) µmol/L, respectively]. Contrary to MTHFR C677T polymorphism, the average changes in plasma homocysteine levels for MS 2756AG and GG variants were negative [β(SE β), −0.56(0.58) and −0.83(0.99) µmol/L, respectively]. The average change occurring for CBS 844ins68 heterozygous genotype (ancestral/insertion) was −1.88(0.81) µmol/L. The combined effect of MTHFR C677T, MS A2756G and CBS 844ins68 genotypes for plasma homocysteine levels was additive (p value <0.001). Odds of having hyperhomocysteinemia with MTHFR 677TT genotype was 10-fold compared to MTHFR 677CC genotype [OR (95%CI); 10.17(3.6–28.67)]. Protective effect towards hyperhomocysteinemia was observed with heterozygous (ancestral/insertion) genotype of CBS 844ins68 compared to homozygous ancestral type [OR (95% CI); 0.58 (0.34–0.99)]. Individuals with MTHFR 677CT or TT genotypes were at a greater risk of hyperhomocysteinemia in folate and vitamin B12 deficiencies and high blood lead (p value <0.05) level.

Conclusions

Gene polymorphism (especially MTHFR C677T transition), folate and vitamin B12 deficiencies, male gender and high blood lead level appear to be contributing towards the development of hyperhomocysteinemia in a Pakistani population.  相似文献
