Recent studies have demonstrated that mesenchymal stem cells (MSCs) combined with CD34+ hematopoietic/stem progenitor cells (HSPCs) can function as surrogate urinary bladder cells to synergistically promote multi-faceted bladder tissue regeneration. However, the molecular pathways governing these events are unknown. The pleiotropic effects of Wnt5a and Cyr61 are known to affect aspects of hematopoiesis, angiogenesis, and muscle and nerve regeneration. Within this study, the effects of Cyr61 and Wnt5a on bladder tissue regeneration were evaluated by grafting scaffolds containing modified human bone marrow derived MSCs. These cell lines were engineered to independently over-express Wnt5a or Cyr61, or to exhibit reduced expression of Cyr61 within the context of a nude rat bladder augmentation model. At 4 weeks post-surgery, data demonstrated increased vessel number (~250 vs ~109 vessels/mm2) and bladder smooth muscle content (~42% vs ~36%) in Cyr61OX (over-expressing) vs Cyr61KD (knock-down) groups. Muscle content decreased to ~25% at 10 weeks in Cyr61KD groups. Wnt5aOX resulted in high numbers of vessels and muscle content (~206 vessels/mm2 and ~51%, respectively) at 4 weeks. Over-expressing cell constructs resulted in peripheral nerve regeneration while Cyr61KD animals were devoid of peripheral nerve regeneration at 4 weeks. At 10 weeks post-grafting, peripheral nerve regeneration was at a minimal level for both Cyr61OX and Wnt5aOX cell lines. Blood vessel and bladder functionality were evident at both time-points in all animals. Results from this study indicate that MSC-based Cyr61OX and Wnt5aOX cell lines play pivotal roles with regards to increasing the levels of functional vasculature, influencing muscle regeneration, and the regeneration of peripheral nerves in a model of bladder augmentation. Wnt5aOX constructs closely approximated the outcomes previously observed with the co-transplantation of MSCs with CD34+ HSPCs and may be specifically targeted as an alternate means to achieve functional bladder regeneration. 相似文献

The large N-linked oligosaccharides released from Schizosaccharomyces pombe
by endo-beta-N-acetylglucosaminidase H were examined to determine how the
negatively chargedpyruvylated galactoses present (Gemmill,T.R., and
Trimble,R.B., 1996, J. Biol. Chem ., 271, 25945-25949) were attached to the
oligosaccharide chains. Binding of biotinylated human serum amyloid P and
peanut agglutinin to native and depyruvylated S.pombe glycoproteins,
respectively, indicated that the pyruvylated epitope was likely to be in
the beta configuration. Examination by high- field 1H NMR of whole glycans
and a disaccharide fragment released from them on partial acid hydrolysis
showed that the pyruvylated galactose species was in fact beta1,3-linked to
a second galactose, and this occurred an average of five to six times on
nominal Gal57Man64GlcNAc N- glycans. The pyruvate-2,(4,6)Gal-beta1,3Gal
epitope is chemically similar to acetaldehyde-Galbeta1,3Gal groups found on
the glycoproteins from Paramyxovirus-infected bovine kidney cells (Prehm,
P., Scheid,A. and Choppin,P.W. ,1979, J. Biol. Chem ., 254, 9669-9677). The
1:1 stoichiometry between pyruvate and beta-linked galactose in these
S.pombe glycans indicates that either pyruvate addition to terminal
beta1,3Gal is highly efficient or that pyruvylated Gal is transferred en
bloc to alpha1,2-linked Gal residues in theN-linked chains. In
contradiction to many galactomannan-producing fungi, which add substantial
amounts of Gal in the furanose form to their glycoproteins, all detectable
Gal in the large S.pombe galactomannans is in the pyranose form, as found
in higher eukaryotes. The current work shows that the S.pombe outer chain
structure is a poly-alpha1,6Man backbone 2- O-substituted with either Gal
or the pyruvylated galactobiose and contains little alpha1,2-linked or
2-O-substituted Man. This is in contrast to the S. cerevisiae outer chain,
which is poly-alpha1,6Man substituted with alpha1,2-linked Man sidechains
(Ballou,C.E. ,1990, Methods Enzymol , 185, 440-470).

Endothelial lipase (EL) activity has been implicated in HDL catabolism, vascular inflammation, and atherogenesis, and inhibitors are therefore expected to be useful for the treatment of cardiovascular disease. Sulfonylfuran urea 1 was identified in a high-throughput screening campaign as a potent and non-selective EL inhibitor. A lead optimization effort was undertaken to improve potency and selectivity, and modifications leading to improved LPL selectivity were identified. AAF-AMC is not a specific TPP II substrate, since it is also hydrolyzed by purified proteasomes. Moreover, AAF-cmk, claimed to be a specific TPP II inhibitor, also inhibits the chymotrypsin-like activity of the proteasome. While AAF-cmk itself is mildly cytostatic to U-937 cells and induces cell cycle block in G1, its combination with PSI does not induce an increase in the cytostatic/cytotoxic effects. In this study we compared the sensitivity of different human and murine cell lines varying in the stage of transformation to dose- and time-dependent cytostatic and/or cytotoxic effects of PSI (a selective proteasome inhibitor), measured by a standard MTT assay. It was found that intensively proliferating cell lines were more sensitive to very small doses of PSI after 24 h incubation than the slow proliferating ones. Non-transformed cell lines showed no sensitivity to PSI, as there was no difference in cell viability in comparison with the control group even after 72 h incubation. 相似文献

Zinc is both a vital nutrient and an important toxicant to aquatic biota. In order to understand the interplay between nutrition and toxicity, it will be important to determine the mechanisms and the factors that regulate zinc uptake. The mechanism of apical intestinal Zn(II) uptake in freshwater rainbow trout and its potential modification by the complexing amino acid histidine was investigated using brush-border membrane vesicles (BBMVs). Following characterisation of the BBMV preparation, zinc uptake in the absence of histidine was both time- and concentration-dependent and consisted of two components. A saturable phase of uptake was described by an affinity constant of 57+/-17 microM and a transport capacity of 1867+/-296 nmol mg membrane protein(-1) min(-1). At higher zinc levels (>500 microM) a linear, diffusive component of uptake was evident. Zinc transport was also temperature-dependent, with Q10 values suggesting zinc uptake was a carrier-mediated process. Zinc uptake by vesicles in the presence of histidine was correlated to a mono-histidine species (Zn(His)+) at all Zn(II) concentrations examined. 相似文献

The present study considers the genetic structure and phylogeography of the smooth snake (Coronella austriaca) in Central Europe, as analyzed on the basis of 14 microsatellite markers and a 284‐bp fragment of cytochrome b. We found deep divergence between western and south‐eastern Poland, suggesting at least two different colonization routes for Central Europe, originating in at least two different refugia. The west/south‐east divide was reflected in the haplotype distribution and topology of phylogenetic trees as defined by mitochondrial DNA, and in population structuring seen in the admixture analysis of microsatellite data. The well supported western European clade suggests that another refugium might have existed. We also note the isolation‐by‐distance and moderate‐to‐pronounced structuring in the examined geographical demes. Our data fit the assumption of the recently suggested sex‐biased dispersal, in that we found a strong divide in the maternal line, as well as evidence for a small but existent gene flow based on biparentally inherited microsatellite markers. All studied populations were very similar in respect of allelic richness, observed and expected heterozygosities, and inbreeding coefficients. However, some genetic characteristics were different from those expected compared to a similar fine‐scale study of C. austriaca from Great Britain.
