The diet of Antarctic silverfish Pleuragramma antarcticum was evaluated by examining stomach contents of specimens collected in the Ross Sea (71°–77° S; 165°–180° E) in January to March 2008. Pleuragramma antarcticum (50–236 mm standard length, LS) and prey items were analysed for stable‐isotopic composition of carbon and nitrogen. According to index of relative importance (IRI), which incorporates frequency of occurrence, mass and number of prey items, the most important prey items were copepods (81%IRI over all specimens), predominantly Metridia gerlachei and Paraeuchaeta sp., with krill and fishes having low IRI (2·2 and 5·6%IRI overall). According to mass of prey (M) in stomachs, however, fishes (P. antarcticum and myctophids) and krill dominated overall diet (48 and 22%M, respectively), with copepods being a relatively minor constituent of overall diet by mass (9·9%M). Piscivory by P. antarcticum occurred mainly in the extreme south‐west of the region and near the continental slope. Krill identified to species level in P. antarcticum stomachs were predominantly Euphausia superba (14·1%M) with some Euphausia crystallophorias (4·8%M). Both DistLM modelling (PRIMER‐permanova+) on stomach contents (by IRI) and stepwise generalized linear modelling on stable isotopes showed that LS and location were significant predictors of P. antarcticum diet. Postlarval P. antarcticum (50–89 mm LS) consumed exclusively copepods. Juvenile P. antarcticum (90–151 mm LS) consumed predominantly krill and copepods by mass (46 and 30%M, respectively). Small adult P. antarcticum (152–178 mm LS) consumed krill, fishes and copepods (37, 36 and 15%M, respectively). Large adult P. antarcticum (179–236 mm LS) consumed predominantly fishes and krill (55 and 17%M, respectively), especially in the north (near the Ross Sea slope) and in the SW Ross Sea. Amphipods were occasionally important prey items for P. antarcticum (western Ross Sea, 39%M). General concordance between stomach contents and trophic level of P. antarcticum and prey based on δ15N was demonstrated. Pleuragramma antarcticum trophic level was estimated as 3·7 (postlarval fish) and 4·1 (fish aged 3+ years).  相似文献

Acute pulmonary inflammation is characterized by migration of polymorphonuclear neutrophils (PMNs) into the different compartments of the lung, passing an endothelial and epithelial barrier. Recent studies showed evidence that phosphodiesterase (PDE)4-inhibitors stabilized endothelial cells. PDE4B and PDE4D subtypes play a pivotal role in inflammation, whereas blocking PDE4D is suspected to cause gastrointestinal side effects. We thought to investigate the particular role of the PDE4-inhibitors roflumilast and rolipram on lung epithelium. Acute pulmonary inflammation was induced by inhalation of LPS. PDE4-inhibitors were administered i.p. or nebulized after inflammation. The impact of PDE4-inhibitors on PMN migration was evaluated in vivo and in vitro. Microvascular permeability, cytokine levels, and PDE4B and PDE4D expression were analyzed. In vivo, both PDE4-inhibitors decreased transendothelial and transepithelial migration even when administered after inflammation, whereas roflumilast showed a superior effect compared to rolipram on the epithelium. Both inhibitors decreased TNFα, IL6, and CXCL2/3. CXCL1, the strong PMN chemoattractant secreted by the epithelium, was significantly more reduced by roflumilast. In vitro assays with human epithelium also emphasized the pivotal role of roflumilast on the epithelium. Additionally, LPS-induced stress fibers, an essential requirement for a direct migration of PMNs into the alveolar space, were predominantly reduced by roflumilast. Expression of PDE4B and PDE4D were both increased in the lungs by LPS, PDE4-inhibitors decreased mainly PDE4B. The topical administration of PDE4-inhibitors was also effective in curbing down PMN migration, further highlighting the clinical potential of these compounds. In pulmonary epithelial cells, both subtypes were found coexistent around the nucleus and the cytoplasm. In these epithelial cells, LPS increased PDE4B and, to a lesser extend, PDE4D, whereas the effect of the inhibitors was prominent on the PDE4B subtype. In conclusion, we determined the pivotal role of the PDE4-inhibitor roflumilast on lung epithelium and emphasized its main effect on PDE4B in hyperinflammation.  相似文献

Both Macrourus caml and M. whitsoni are probably abundant and widespread in the Ross Sea region at depths between approximately 500 and 2100 m. We present the analysis of stomach contents (including regurgitated stomach contents) from an undifferentiated mixture of 33 specimens of these species (henceforth Macrourus spp.) in the Ross Sea region of the Southern Ocean. Samples of stomach contents were obtained from the Ross Sea slope (between about 71° and 73° S) and on the Scott Island and seamount chain. Overall, amphipods were the dominant prey found in Macrourus spp. stomachs with an index of relative importance (IRI) of 43%. About half the amphipods were Eurythenes gryllus and about half could not be identified. Based on stable isotope results, unidentified amphipods were probably herbivorous rather than carnivorous. Other important prey were copepods (especially for fish with total length less than 30 cm), krill (mainly Euphausia superba for larger fish) and fishes (Pleuragramma antarcticum and Gymnoscopelus opisthopterus). Minor prey found in stomachs included isopods, mysids, salps and polychaetes, with rocks and fragments of coral, echinoderm and shell also found. Stomach contents data provide evidence of both pelagic and benthic feeding. Carbon and nitrogen stable-isotope analysis of 161 Macrourus spp. muscle tissue samples from the Ross Sea slope, Admiralty and Scott seamounts and two parts of the Pacific-Antarctic fracture zone (near 65° S) are presented. Nitrogen isotope results (??15N = 10.6 ± 1.0??, mean ± SD) were generally consistent with stomach contents. Carbon isotope results (??13C = ?24.7 ± 0.8??) were consistent with Southern Ocean residence but higher than phytoplankton values (?29.9 ± 1.2??). We found that Macrourus spp. in the Ross Sea region have a trophic level of 3.8 ± 0.3, placing them well below the top of the food chain. Trophic level of Macrourus spp. was significantly affected by location and by size of fish (especially for fish less than 30 cm), whereas sex, fatness and bottom depth were not significant.  相似文献

ABSTRACT: BACKGROUND: Duplicated glucocorticoid receptors (GR) are present in most teleost fish. The evolutionary advantage of retaining two GRs is unclear, as no subtype specific functional traits or physiological roles have been defined. To identify factors driving the retention of duplicate GRs in teleosts, the current study examined GRs in representatives of two basal ray-finned fish taxa that emerged either side of the teleost lineage whole genome duplication event (WGD) event, the acipenseriform, Acipenser ruthenus, (pre-WGD) and the osteoglossimorph, Pantodon buchholzi, (post-WGD). RESULTS: The study identified a single GR in A. ruthenus (ArGR) and two GRs in P. buchholzi (PbGR1 and PbGR2). Phylogenetic analyses showed that ArGR formed a distinct branch separate from the teleosts GRs. The teleost GR lineage was subdivded into two sublineages, each of which contained one of the two P. buchholzi GRs. ArGR, PbGR1 and PbGR2 all possess the unique 9 amino acid insert between the zinc-fingers of the DNA-binding domain that is present in one of the teleost GR lineages (GR1), but not the other (GR2). A splice variant of PbGR2 produces an isoform that lacked these 9 amino acids (PbGR2b). Cortisol stimulated transactivation activity of ArGR, PbGR2b and PbGR1 in vitro; with PbGR2b and PbGR1, the glucocorticoid 11-deoxycortisol was a more potent agonist than cortisol. The hormone sensitivity of PbGR2b and PbGR1 differed in the transactivation assay, with PbGR2b having lower EC50 values and greater fold induction. CONCLUSIONS: The difference in transactivation activity sensitivity between duplicated GRs of P. buchholzi suggests potential functional differences between the paralogs emerged early in the teleost lineage. Given the pleiotropic nature of GR function in vertebrates, this finding is in accordance with the hypothesis that duplicated GRs were potentially retained through subfunctionalisation followed by gene sharing. A 9 amino acid insert in the DNA-binding domain emerged in basal ray-finned fish GRs. However, the presence of a PbGR2 splice variant that lacks this insert, as well as the loss of the exon encoding these amino acids in the genes encoding for other teleost GR2 suggests the selection of two receptors with different DNA-binding domain structures in teleosts.  相似文献
