Thyroid autoregulation has been linked to an organified iodocompound. Since several iodolipids are produced by the gland their possible role in thyroid autoregulation was examined. The following pure synthetic compounds were prepared: 1) 14-iodo-15-hydroxy-5,8,11-eicosatrienoic acid (I-OH-A); 2) its omega lactone (IL-ω);3)5-hydroxy-6-iodo-8,11,14-eicosatrienoic acid delta lactone (IL-δ). Their action on iodine metabolism was studied. Iodine uptake was measured in calf thyroid slices. At 10-4M I-OH-A caused a 64% decrease in the T/M ratio while IL-ω inhibited it by 36% and IL-δ was without effect. At 10−5M the inhibition was 44% for I-OH-A and 19% for IL-ω, while T3 was without action. A possible isotopic dilution effect was excluded, and no change in iodine efflux was observed. The inhibition by I-OH-A of iodine uptake was observed after only 15 min preincubation. This compound alse decreased 125I accumulation in rats.In calf thyroid slice, I-OH-A at 10−4M, inhibited PB125I formation by 80%, IL-ω 62% and IL-ω by 37% and arachidonic acid were without action. I-OH-A also caused a dose-dependent inhibition of TSH-stimulated iodide organification.The present results demonstrate, for the first time, that iodinated derivatives of arachidonic acid inhibition thyroid function and mimic the effect of iodine on thyroid autoregulation.  相似文献

Rhodanese has been utilized as a model enzyme for the study of protein structure-function relationships. The enzyme has recently been cloned and the recombinant enzyme is now available for investigation. However, prior to use in structure-function studies, the recombinant enzyme must be shown to have the same structure and activity as the bovine liver enzyme used in the previous studies. An immunological study of the conformations of these enzyme conformers is described. Three antibodies (two monoclonal and one polyclonal, site-directed antibody) were shown to detect distinct and nonoverlapping epitopes. The epitopes of the monoclonal antirhodanese antibodies (R207 and MAB11) were mapped to the same CNBr digest fragment of the amino terminal domain of rhodanese, and the epitope of the site-directed antibody prepared against the interdomain tether sequence of rhodanese (PAT-T1) was mapped to that region of rhodanese (residues 142–156). The rhodanese conformers were studied by monitoring the accessibility of the epitopes recognized by each antibody in each conformer using an indirect ELISA. None of the antibodies could detect its epitope on the purified liver enzyme. Two of the antibodies (R207 and PAT-T1) could also not detect their epitopes on the recombinant enzyme. However, MAB11 did detect a conformational difference between the natural and recombinant rhodanese conformers, indicating the conformational difference is localized in the first 73 amino acids of rhodanese. This difference presumably reflects the difference in the histories of the two enzymes and may be due to differences in enzyme folding, differences in the purification procedures, and differences in storage conditions—all of which could influence the final conformation of the enzyme.  相似文献

Neurotrophic factors are essential for neuronal survival and function. Recent data have demonstrated that the product of the tyrosine kinase trk proto-oncogene binds NGF and is a component of the high affinity NGF receptor. Analysis of the trkB gene product, gp145trkB, in NIH 3T3 cells indicates that this tyrosine kinase receptor is rapidly phosphorylated on tyrosine residues upon exposure to the NGF-related neurotrophic factors BDNF and NT-3. Furthermore, gp145trkB specifically binds BDNF and NT-3 in NIH 3T3 cells and in hippocampal cells, but does not bind NGF. Thus, the trk family of receptors are likely to be important signal transducers of NGF-related trophic signals in the formation and maintenance of neuronal circuits.  相似文献

Taxonomic and genetic diversities of microbial communities disturbed by chemical pollutants were lower than in undisturbed reference communities. The dominant populations within the disturbed communities had enhanced physiological tolerances and substrate utilization capabilities, indicating that generalized physiological versatility is an adaptive characteristic of populations that successfully compete within disturbed communities.  相似文献

Modifiers of position-effect-variegation in Drosophila encode proteins that are thought to modify chromatin, rendering it heritably changed in its expressibility. In an attempt to identify similar modifier genes in other species we have utilized a known sequence homology, termed chromo box, between a suppressor of position-effect-variegation, Heterochromatin protein 1 (HP1), and a repressor of homeotic genes, Polycomb (Pc). A PCR generated probe encompassing the HP1 chromo box was used to clone full-length murine cDNAs that contain conserved chromo box motifs. Sequence comparisons, in situ hybridization experiments, and RNA Northern blot analysis suggest that the murine and human sequences presented in this report are homologues of the Drosophila HP1 gene. Chromo box sequences can also be detected in other animal species, and in plants, predicting a strongly conserved structural role for the peptide encoded by this sequence. We propose that epigenetic (yet heritable) changes in gene expressibility, characteristic of chromosomal imprinting phenomena, can largely be explained by the action of such modifier genes. The evolutionary conservation of the chromo box motif now enables the isolation and study of putative modifier genes in those animal and plant species where chromosomal imprinting has been described.  相似文献

Summary Single-stranded cDNA was prepared from RNA obtained from a patient with type 1 Gaucher disease. The cDNA was amplified in vitro and analyzed by sequencing. Three base-pair changes were identified which included a G to C transversion at nucleotide 3119 of the active gene (Asp140His), an A to C transversion at nucleotide 3170 (Lys157Gln) and a G to A change at nucleotide 5309 (Glu326Lys). To study the mode of inheritance of the three different base-pair changes, genomic DNA was prepared from blood or skin fibroblasts of several family members. Genomic glucocerebrosidase DNA sequences were amplified and subjected to hybridization with allele-specific oligonucleotides (ASOs). The hybridization profiles demonstrated that two of the basepair changes originated from the mother and were transmitted to her two affected sons and to a grandchild, while the third base-pair change, originating from the father, was transmitted to his two affected sons, a carrier daughter and a second grandchild. Tests of other patients with Gaucher disease failed to disclose the presence of the three base-changes. This is a unique family with three base-pair changes tightly linked to Gaucher disease.  相似文献

Benthic grab samples, taken through core holes in sea-ice 1983, returned eight species of marine macrophytes which had associated fauna on at least one sample. A total of 24 animal taxa were found on these eight macrophytes. Phyllophora antarctica had a significantly greater biomass of attached fauna per unit of macrophyte biomass than any of the other seven species. It also had the greatest variety of epiphytic fauna (21 taxa). Phyllophora antarctica was the only macrophyte collected from comparatively protected Ellis Fjord, apart from filamentous algae, including Cladophora subsimplex and Geminocarpus austrogeorgiae, which were found in the most landward basin. It therefore played an important role in the ecology of the fjord. Palmaria decipiens was commonly collected at more exposed coastal sites and was dominant in Long Fjord, characterized here as intermediate in terms of water movement. Sixteen animal taxa were collected from this macrophyte with the greatest numbers being found in July and August. Only four animal taxa were collected from Iridaea cordata. The other five macrophyte species from which epiphytic animals were collected (Porphyra endiviifolium, Plocamium cartilagineum, Desmarestia menziesii, Himantothallus grandifolius and Cladophora subsimplex had animals associated with them on less than half the occasions when they were found.  相似文献

The distributions of trace elements in Shield, Ace and Burton Lakes of the Vestfold Hills were investigated. Three aspects are discussed as follows: (1) the vertical distribution of 18 trace elements in the three lakes, (2) the behaviour of trace elements in the lakes, especially that of manganese in Shield Lake, and (3) the origin of trace elements in antarctic saline lakes.High concentrations of trace elements were found in these coastal saline lakes, when compared to open ocean water.We suggest that the peak of total extractable manganese, found at 20 m in Shield Lake, was related to the oxic/anoxic water interface brought about by microbiological activity. Solid phase manganese at the upper oxic layer may have precipitated and then reached the anoxic boundary to be there reduced to manganese ion. This dissolved manganese may then have diffused upwards to be reoxidized to a solid form. This cycle, repeated many times, may have produced the Mn profile.The alkali, alkaline earth elements and Cl were probably derived from relict seawater. Other elements were present in similar concentration ratios to those of South Polar aerosols. Residence time calculations indicate that fallout of aerosol particles, themselves derived from various sources, is capable of accounting for the measured concentrations of some trace elements in Shield Lake. This source of trace elements may be significant for other antarctic saline lakes.  相似文献

The compositions of carotenoids, chlorophylls and lipids at four depths in Ace Lake have been determined as a means of studying the vertical zonation of species in the lake and for comparison with the lipids found in the bottom sediments. The four major species of phytoplankton found in the lake were identified by electron microscopy. The most abundant phytoplankter was Pyramimonas gelidicola McFadden (Chlorophyta, Prasinophyceae) which occurred in greatest numbers at 10 m, the base of the oxylimnion. The pigments and lipids at this depth were mainly derived from this alga. At 11 m (the top of the anoxylimnion) only traces of lipids and pigments attributable to P. gelidicola were found, indicating only limited settling of algal cells through to the anoxylimnion, at least in summer. The pigments at 11 m were dominated by bacteriochlorophylls c derived from green photosynthetic bacteria Chlorobium spp. These pigments were also abundant at 23 m suggesting the presence of intact bacterial cells which had settled out from higher in the water column. Major non-polar lipid classes in the sediments included sterols, alcohols, hydrocarbons and an unusual suite of very long-chain unsaturated ketones and esters which have not previously been reported from antarctic environments. Several novel compounds, not found previously in either sediments or organisms, are reported. These include tri- and tetra-unsaturated straight-chain C39 methyl ketones and C40 ethyl ketones and the methyl ester of a tetra-unsaturated straight-chain C36 fatty acid. The distributions of lipids in the sediment were markedly different from those in the water column indicating extensive bacterial degradation and recycling of labile lipids.  相似文献
