When testing for the effects of bone orientation on mode I fracture toughness, compact tension specimens are grooved with a V-notch to provide a crack guide. The effect of grooving on the expressions for the critical stress intensity factor (Kc) and the critical strain energy release rate (Gc) for mode I fracture toughness was investigated. Experiments were performed using grooved and ungrooved bovine compact tension specimens. The results indicate that the standard expression used to determine Kc for a compact tension specimen requires modification. The thickness (B) must be modified to account for the thickness between the grooves (Bn). The thickness used in the standard expression is replaced by an effective thickness written as (BBn)0.5. It was also found that the thickness between the grooves should be used in the standard formula for Gc.  相似文献

William (Bill) E. Vidaver (February 2, 1921–August 31, 2017), who did his Ph.D. with Laurence (Larry) R. Blinks at Stanford (1964) and a postdoc with C. Stacy French (1965), taught and did research at Simon Fraser University (SFU) for almost 30 years. Here he published over 80 papers in photosynthesis-related areas co-authored by his graduate students, postdocs, visiting professors and SFU colleagues. He developed a unique high-pressure cuvette for the study of oxygen exchange and studied high-pressure effects in photosynthesis. Ulrich (Uli) Schreiber, as a postdoctoral fellow from Germany, introduced measurements on chlorophyll (Chl) a fluorescence to Bill’s lab, leading to the discovery of reversible inhibition of excitation energy transfer between photosynthetic pigments and of a pivotal role of O2 in the oxidation of the electron transport chain between Photosystem II (PS II) and PS I. Bill’s and Uli’s work led to a patent of a portable chlorophyll fluorometer, the first available commercially, which was later modified to measure whole plantlets. The latter was used in pioneering measurement of the health of forest and crop plants undergoing in vitro clonal micropropagation. With several other researchers (including Doug Bruce, the late Radovan Popovic, and Sarah Swenson), he localized the quenching site of O2 and showed a dampening effect on measurements of the four-step process of O2 production by endogenous oxygen uptake. Bill is remembered as a hard-working but fun-loving person with a keen mind and strong sense of social justice.  相似文献

Two genes have been identified in Campylobacter coli VC167 which are required for the biosynthesis of post-translational modifications on flagellin proteins. The ptmA gene encodes a protein of predicted M r 28 486 which shows significant homology to a family of alcohol dehydrogenases from a variety of bacteria. The ptmB gene encodes a protein of predicted M r 26 598 with significant homology to CMP- N -acetylneuraminic acid synthetase enyzmes involved in sialic acid capsular biosynthesis in Neisseria meninigitidis and Escherichia coli K1. Site-specific mutation of either ptmA or ptmB caused loss of reactivity with antisera specific to the post-translational modifications and a change in the isoelectric focusing fingerprints relative to the parent strains. Mutation of ptmB , but not of ptmA , caused a change in apparent M r of the flagellin subunit in SDS–PAGE gels. The ptmA and ptmB genes are present in other strains of Campylobacter . In a rabbit model the ptmA mutant showed a reduced ability to elicit protection against subsequent challenge with heterologous strains of the same Lior serotype compared to the parental wild-type strain. This suggests that the surface-exposed post-translational modifications may play a significant role in the protective immune response.  相似文献

Xylella fastidiosa causes Pierce's disease (PD) on grapevines, leading to significant economic losses in grape and wine production. To further our understanding of X. fastidiosa virulence on grapevines, we examined the PD1311 gene, which encodes a putative acyl‐coenzyme A (acyl‐CoA) synthetase, and is highly conserved across Xylella species. It was determined that PD1311 is required for virulence, as the deletion mutant, ΔPD1311, was unable to cause disease on grapevines. The ΔPD1311 strain was impaired in behaviours known to be associated with PD development, including motility, aggregation and biofilm formation. ΔPD1311 also expressed enhanced sensitivity to H2O2 and polymyxin B, and showed reduced survival in grapevine sap, when compared with wild‐type X. fastidiosa Temecula 1 (TM1). Following inoculation, ΔPD1311 could not be detected in grape shoots, which may be related to its altered growth and sensitivity phenotypes. Inoculation with ΔPD1311 2 weeks prior to TM1 prevented the development of PD in a significant fraction of vines and eliminated detectable levels of TM1. In contrast, vines inoculated simultaneously with TM1 and ΔPD1311 developed disease at the same level as TM1 alone. In these vines, TM1 populations were distributed similarly to populations in TM1‐only inoculated plants. These findings suggest that, through an indirect mechanism, pretreatment of vines with ΔPD1311 suppresses pathogen population and disease.  相似文献

The chromatrope pigment of Mermis nigrescens (Phylum: Aschelminthes, Class: Nematoda) was previously thought to have a role in photoreception. In this work it is shown to be an oxyhemoglobin whose absorption spectrum in vivo and in extracts resembles that of Ascaris oxyhemoglobins in having a weak alpha-band absorptivity. The extracted hemoglobin binds O2 and CO reversibly and with an affinity higher than that of the same concentration of horse hemoglobin. The Soret band of the deoxy derivative is unusually low and broad. Absorptivities and lambdamax for absorption bands of the oxy, deoxy and CO derivatives are tabulated for comparison with other hemoglobins. Microchemical procedures were developed which revealed that the chromatrope contains an average of 5-10(-12) mol of non-dialysable protoheme. Thus the hemoglobin concentration in the approx. 0.5 nl chromatrope volume is on the order of 10 mM (heme). The O2 binding ability and high in vivo concentration of this hemoglobin make possible a role in O2 storage or facilitation of O2 diffusion.  相似文献

Conjugated linoleic acid (CLA) elevates body ash in healthy animals. The objective of the present study was to determine if single or mixed CLA isomers improve bone mass in an obese and hyperinsulinemic state. Male (n = 120) lean and obese fa/fa Zucker rats (age, 6 weeks) were randomized to 8 weeks on a control diet or to 0.4% (w/w) cis-9, trans-11 CLA (Group 1); 0.4% (w/w) trans-10, cis-12 CLA (Group 2); 0.4% (w/w) cis-9, trans-11 CLA and 0.4% (w/w) trans-10, cis-12 CLA (Group 3); 0.4% (w/w) cis-9, trans-11 CLA, 0.4% (w/w) trans-10, cis-12 CLA, and traces of other CLA isomers (Group 4); and 0.4% (w/w) cis-9, trans-11 CLA, 0.4% (w/w) trans-10, cis-12 CLA, and 0.3% (w/w) other CLA isomers (Group 5). Bone area (BA), bone mineral content (BMC), and bone mineral density (BMD) of the whole body, spine, and femur were measured at baseline (6 weeks) and at 14 weeks of age. Effects of genotype, diet, and genotype x diet interactions were assessed using factorial analysis of variance. At 6 and 14 weeks, whole-body BA and BMC were lower in lean rats compared with fa/fa rats. Similarly, at 14 weeks, fa/fa rats had a higher spine and femur BMD despite a lower femur weight. The fa/fa rats in Groups 4 and 5 had higher adjusted whole-body BMC compared with Group 3, but not with Group 1, Group 2, or the control. In lean rats, Group 3 had a greater adjusted whole-body BMC than Groups 1 and 2, but not Group 4, Group 5, or the control. Thus, commercially available CLA mixtures and single CLA isomers do not affect bone mass in a hyperinsulinemic, obese state.  相似文献

We have demonstrated a dramatic induction of manganese superoxide dismutase (Mn-SOD) mRNA levels in response to lipopolysaccharide (LPS), interleukin-1, and tumor necrosis factor in pulmonary epithelial cells. These stimuli had no effect on the corresponding mRNA levels for the copper/zinc (Cu/Zn)-SOD. Identical treatments of pulmonary fibroblast cells with LPS showed only minor changes in the Mn-SOD mRNA levels demonstrating a cell type-specific effect for this acute inflammatory mediator. Furthermore, we have shown that hyperoxia has no effect within 24 h on Mn-or Cu/Zn-SOD mRNA levels in either fibroblasts or epithelial cells. The induction of Mn-SOD mRNA levels by LPS is completely inhibited by actinomycin. Treatment of cells with cycloheximide causes an induction equal to that for LPS, whereas co-treatment with cycloheximide and LPS resulted in a "super induction." This data is strongly suggestive of an important role for the Mn-SOD in the acute inflammatory response.  相似文献

Summary The gravid females of Mermis are positively phototaxic at the time of their migration to egglaying sites in vegetation on which their grasshopper hosts feed. On a horizontal felt surface, segments of the path traced by the tail are oriented approximately towards a source of monochromatic light in the 350–540 nm region, but are not oriented at longer wavelengths and in the dark. The components of this phototaxis include locomotion by the posterior 4/5 of the body, orientational bending of the neck region while the anterior is held above the substrate, and a scanning motion (bending) of the head region (anterior 2 mm). Like other nematodes and snakes, propulsion is associated with posteriorly propagated body waves, but unlike other animals known, the waves tend to lie perpendicular to a felt surface, and unlike other nematodes, contact with the surface is on the female's ventral surface. The body waves are initiated by the motion of the anterior 1/5 (15 mm) of the body, the average orientation of which determines the path of the following 4/5.During phototaxis, the anterior tip is swung both sideways and vertically about the direction towards the light source. The tip motion is a result of a scanning motion of the head and a slower orientational bending of the neck. The base of the head appears to be actively directed towards the source by the bending of the neck. This behavior can resolve two light sources positioned 120° apart but not 90° apart. The scanning motion of the head is independent of neck orientation and appears to enhance the probability of discovering the direction of a new source. Discovery is followed by a directed turn of the base of the head towards the source which is initiated by the bending of the neck. Locomotion of the body follows the path of the anterior through the turn and phototaxis is thus initiated.  相似文献
