Objective: To examine obesity prevalence and weight control practices among veterans who use Department of Veterans Affairs (VA) medical facilities (VA users). Research Methods and Procedures: Data from the 2000 Behavioral Risk Factor Surveillance System, a telephone survey of 184, 450 adults, were analyzed. Outcome measures included BMI, weight control practices (the intent to manage weight, and diet and physical activity patterns), and receipt of professional weight control advice. Results: Of VA users, 44% were overweight and 25% were obese. After controlling for demographic factors, VA users were somewhat less likely to be overweight (odds ratio, 0.86; 95% confidence interval, 0.74 to 1.00) but equally likely to be obese (odds ratio, 1.08; 95% confidence interval, 0.92 to 1.27), compared with non‐VA users. Among obese VA users, 75% reported trying to lose weight, and another 17% reported trying to maintain weight. Of these, only 40% decreased both calorie and fat intake. Only 27% of obese VA users who reported increasing exercise to lose weight followed recommendations for regular and sustained physical activity. Of obese VA users, 59% were inactive or irregularly active. Only 51% of obese VA users received professional advice to lose weight. Obese VA users were more likely than obese non‐VA users to report trying to lose weight, modifying diet to lose weight by decreasing both calories and fat intake, and receiving professional weight control advice. Discussion: Interventions for weight management programs in VA facilities need to take into account the high prevalence of overweight/obesity among VA users and should emphasize effective weight control practices.  相似文献

The homeostatic function of endothelial cells (EC) is critical for a number of physiological processes including vascular integrity, immunity, and wound healing. Indeed, vascular abnormalities resulting from EC dysfunction contribute to the development and spread of malignancies. The alternative SDF-1/CXCL12 receptor CXCR7 is frequently and specifically highly expressed in tumor-associated vessels. In this study, we investigate whether CXCR7 contributes to vascular dysfunction by specifically examining the effect of CXCR7 expression on EC barrier function and motility. We demonstrate that CXCR7 expression in EC results in redistribution of CD31/PECAM-1 and loss of contact inhibition. Moreover, CXCR7+ EC are deficient in barrier formation. We show that CXCR7-mediated motility has no influence on angiogenesis but contributes to another motile process, the invasion of CXCR7+ EC into ligand-rich niches. These results identify CXCR7 as a novel manipulator of EC barrier function via alteration of PECAM-1 homophilic junctions. As such, aberrant expression of CXCR7 in the vasculature has the potential to disrupt vascular homeostasis and could contribute to vascular dysfunction in cancer systems.  相似文献

The 3D encapsulation of cells within hydrogels represents an increasingly important and popular technique for culturing cells and towards the development of constructs for tissue engineering. This environment better mimics what cells observe in vivo, compared to standard tissue culture, due to the tissue-like properties and 3D environment. Synthetic polymeric hydrogels are water-swollen networks that can be designed to be stable or to degrade through hydrolysis or proteolysis as new tissue is deposited by encapsulated cells. A wide variety of polymers have been explored for these applications, such as poly(ethylene glycol) and hyaluronic acid. Most commonly, the polymer is functionalized with reactive groups such as methacrylates or acrylates capable of undergoing crosslinking through various mechanisms. In the past decade, much progress has been made in engineering these microenvironments - e.g., via the physical or pendant covalent incorporation of biochemical cues - to improve viability and direct cellular phenotype, including the differentiation of encapsulated stem cells (Burdick et al.).The following methods for the 3D encapsulation of cells have been optimized in our and other laboratories to maximize cytocompatibility and minimize the number of hydrogel processing steps. In the following protocols (see Figure 1 for an illustration of the procedure), it is assumed that functionalized polymers capable of undergoing crosslinking are already in hand; excellent reviews of polymer chemistry as applied to the field of tissue engineering may be found elsewhere (Burdick et al.) and these methods are compatible with a range of polymer types. Further, the Michael-type addition (see Lutolf et al.) and light-initiated free radical (see Elisseeff et al.) mechanisms focused on here constitute only a small portion of the reported crosslinking techniques. Mixed mode crosslinking, in which a portion of reactive groups is first consumed by addition crosslinking and followed by a radical mechanism, is another commonly used and powerful paradigm for directing the phenotype of encapsulated cells (Khetan et al., Salinas et al.).  相似文献

A device was developed to monitor the vaginal temperature of cattle in a research setting. This device decreases labor involved with monitoring body temperature compared with manual temperature readings, allows for continuous monitoring of vaginal temperature at 1 min intervals, and also allows for temperature measurements without the presence of a human handler or without restraint, which can agitate cattle. The device consists of a blank controlled internal drug release (CIDR) device (designed by Pfizer Animal Health as an indwelling vaginal probe) that holds an indwelling vaginal temperature probe logger. The fabrication of the vaginal probe costs approximately US $325 per unit. Similar rectal and vaginal temperature responses to lipopolysaccharide challenge were observed when vaginal and rectal temperatures were measured simultaneously in the same heifer (P>0.05). Additionally, rectal and vaginal temperatures were highly correlated (r=0.97; P<0.0001). Similar to the rectal temperature monitoring device, the vaginal device allows for the measurement of vaginal temperature without the potential biases associated with the stress response produced as a reaction to the handling by and (or) presence of humans. The vaginal temperature recording device will provide researchers with an additional inexpensive tool to study physiological responses in female cattle.  相似文献

Disrupted-in Schizophrenia 1 (DISC1), a susceptibility gene for major mental disorders, encodes a scaffold protein that has a multifaceted impact on neuronal development. How DISC1 regulates different aspects of neuronal development is not well understood. Here, we show that Fasciculation and Elongation Protein Zeta-1 (FEZ1) interacts with DISC1 to synergistically regulate dendritic growth of newborn neurons in the adult mouse hippocampus, and that this pathway complements a parallel DISC1-NDEL1 interaction that regulates cell positioning and morphogenesis of newborn neurons. Furthermore, genetic association analysis of two independent cohorts of schizophrenia patients and healthy controls reveals an epistatic interaction between FEZ1 and DISC1, but not between FEZ1 and NDEL1, for risk of schizophrenia. Our findings support a model in which DISC1 regulates distinct aspects of neuronal development through its interaction with different intracellular partners and such epistasis may contribute to increased risk for schizophrenia.  相似文献

In recent years, salt marsh restoration projects have focused upon restoring hydrology through culvert enlargement to return functional values lost due to reduced tidal flow. To evaluate culvert effects on upstream nekton assemblages, fyke nets were set upstream of tidally restricted creeks, creeks recently restored with larger culverts, and paired reference creeks in New Hampshire and Maine, U.S.A. Subtidal habitats created or enlarged by scour were found immediately upstream of undersized culverts. All marshes supported similar assemblages and densities of fish, suggesting that marshes upstream of moderately restrictive culverts provide suitable habitat to support fish communities. However, densities of Crangon septemspinosa (sand shrimp) were significantly reduced upstream of culverts. A mark–recapture study was conducted in tidally restricted, restored, and reference marsh creeks to evaluate culvert effects on the movement of Fundulus heteroclitus (mummichog), the numerically dominant fish species in New England salt marshes. Recapture data indicated that small culvert size and consequently increased water velocity significantly decreased fish passage rates. We infer that upstream subtidal habitats and greater water velocities due to undersized culverts decreased nekton movements between upstream and downstream areas, resulting in segregated nekton populations. Restoration of salt marsh hydrology by the installation of adequately sized culverts will support increased fish access to marsh habitats and nekton‐mediated export of marsh‐derived production to coastal waters.  相似文献

Reconstructing the phylogeographic patterns of widely distributedand common freshwater mussel species (Bivalvia: Unionidae) mayprovide insight into unionid evolution and speciation. The Wabashpigtoe, Fusconaia flava, is currently recognized as a single,polytypic species that is widely distributed and common throughoutthe Mississippi River drainage and parts of the Canadian Interior,Great Lakes and Gulf Coast drainages. Sequence analysis of themitochondrial COI gene revealed two divergent (3.43%) clades.Clade A consisted of specimens located throughout the upperand lower Mississippi River drainage and in the Red River (Canada)and Lake Erie drainages and all F. cerina specimens. All haplotypeswithin clade A differed by three (0.55%) or fewer nucleotidesubstitutions from the most widely distributed and abundanthaplotype, F1. Clade B, consisting of specimens located in thefar western portion of the species' range, may comprise an undescribedspecies. There was no evidence of genetic differentiation amongF. flava inhabiting headwater and intermediate-sized river localitiesof the Muskingum River system and large river localities ofthe nearby Ohio River. The divergence among F. flava haplotypescomprising clade A (0.18–1.10%) was similar to the divergencebetween the F. cerina haplotypes and the F. flava haplotypescomprising clade A (0–1.10%). This study illustrates theimportance of accessing genetic diversity across the distributionof a polytypic species. Additional analyses based on a combinationof morphology and genetics are needed to determine the taxonomicstatus of clade B and to strengthen our understanding of therelationship between F. flava and F. cerina.
