ESS models for the evolution of seed size variation assume that seedlings compete with each other for the occupancy of 'safe sites' or vegetation gaps. If mortality rates are high and/or frequency-independent, ESS models reduce to the classical model of Smith and Fretwell which predicts that a single, optimum seed size should occur. We tested whether seedlings compete with one another by following the survival of seedlings colonizing experimental gaps in a grazed grassland community. In small gaps (3 cm diameter) the proximity of established plants slightly, though significantly reduced seedling survival, but density-dependent mortality also occurred among seedlings in these gaps. In larger gaps (6 cm, 9 cm diameter) survival was significantly positively frequency-dependent. These results strongly support the validity of ESS models.  相似文献

Skeletal homeostasis is sensitive to perturbations in Wnt signaling. Beyond its role in the bone, Wnt is a major target for pharmaceutical inhibition in a wide range of diseases, most notably cancers. Numerous clinical trials for Wnt-based candidates are currently underway, and Wnt inhibitors will likely soon be approved for clinical use. Given the bone-suppressive effects accompanying Wnt inhibition, there is a need to expose alternate pathways/molecules that can be targeted to counter the deleterious effects of Wnt inhibition on bone properties. Activation of the Pi3k/Akt pathway via Pten deletion is one possible osteoanabolic pathway to exploit. We investigated whether the osteopenic effects of β-catenin deletion from bone cells could be rescued by Pten deletion in the same cells. Mice carrying floxed alleles for Pten and β-catenin were bred to Dmp1-Cre mice to delete Pten alone, β-catenin alone, or both genes from the late-stage osteoblast/osteocyte population. The mice were assessed for bone mass, density, strength, and formation parameters to evaluate the potential rescue effect of Pten deletion in Wnt-impaired mice. Pten deletion resulted in high bone mass and β-catenin deletion resulted in low bone mass. Compound mutants had bone properties similar to β-catenin mutant mice, or surprisingly in some assays, were further compromised beyond β-catenin mutants. Pten inhibition, or one of its downstream nodes, is unlikely to protect against the bone-wasting effects of Wnt/βcat inhibition. Other avenues for preserving bone mass in the presence of Wnt inhibition should be explored to alleviate the skeletal side effects of Wnt inhibitor-based therapies.  相似文献

BACKGROUND: Violence in the workplace is an ill-defined and underreported concern for health care workers. The objectives of this study were to examine perceived levels of violence in the emergency department, to obtain health care workers'' definitions of violence, to determine the effect of violence on health care workers and to determine coping mechanisms and potential preventive strategies. METHODS: A retrospective written survey of all 163 emergency department employees working in 1996 at an urban inner-city tertiary care centre in Vancouver. The survey elicited demographic information, personal definition of violence, severity of violence, degree of stress as a result of violence and estimate of the number of encounters with violence in the workplace in 1996. The authors examined the effects of violence on job performance and job satisfaction, and reviewed coping and potential preventive strategies. RESULTS: Of the 163 staff, 106 (65%) completed the survey. A total of 68% (70/103) reported an increased frequency of violence over time, and 60% (64/106) reported an increased severity. Most of the respondents felt that violence included witnessing verbal abuse (76%) and witnessing physical threats or assaults (86%). Sixty respondents (57%) were physically assaulted in 1996. Overall, 51 respondents (48%) reported impaired job performance for the rest of the shift or the rest of the week after an incident of violence. Seventy-seven respondents (73%) were afraid of patients as a result of violence, almost half (49%) hid their identities from patients, and 78 (74%) had reduced job satisfaction. Over one-fourth of the respondents (27/101) took days off because of violence. Of the 18 respondents no longer working in the emergency department, 12 (67%) reported that they had left the job at least partly owing to violence. Twenty-four-hour security and a workshop on violence prevention strategies were felt to be the most useful potential interventions. Physical exercise, sleep and the company of family and friends were the most frequent coping strategies. INTERPRETATION: Violence in the emergency department is frequent and has a substantial effect on staff well-being and job satisfaction.  相似文献

A Lactobacillus sp. was consistently isolated from dead and moribund postspawning brood stock rainbow trout. Pathology was characterized by massive chronic inflammation throughout the abdominal cavity and formation of pseudomembranes. The bacteria were most abundant in spent testes. Synchronous cultures of the fission yeast Schizosaccharomyces pombe 972 h−1 are most sensitive to killing by 15 min, 49 °C pulses during a stage stretching from nuclear division through short G1 and S phases to a point early in G2. In this work the cell cycle position of the S phase has been altered by growing the cells in the presence of 2-phenylethanol. The heat sensitivity of these cells was greater at all stages of the cell cycle compared with the cells grown without 2-phenylethanol. However, the position of the most heat sensitive stage in the cell cycle was unaltered. This heat sensitive stage did not include S phase in the cells grown with 2-phenylethanol.  相似文献

The chromatographic behavior of the rabbit uterine progesterone receptor interaction on several different hydrophobic matrices was characterized. Receptor, prepared in 0.6 M NaCl, exhibited a progressive retardation of elution, followed by retention, on a series of alkyl agarose columns as the length of the alkyl chain [(CH2)nH-] increased (n = 0-10), reflecting the presence of hydrophobic regions on the protein. Adsorption did not occur directly at the steroid binding site of the molecule and did not require activation to the DNA-binding form. Elution could be achieved by a decrease in the ionic strength of the buffer or the addition of glycerol, resulting in partial purification of receptor. Receptor bound tightly to phenyl agarose, although elution of the receptor under mild conditions (decreasing salt gradient, increasing glycerol gradient) resulted in poor yield and only modest purification. Passage of the non-activated progesterone receptor over Reactive Blue Sepharose effectively removed albumin, presumably by a hydrophobic interaction, although receptor was not retained. In the activated form, approximately 25% of receptor was bound to Reactive Blue Sepharose, reflecting an interaction of the Cibacron Blue dye with the polynucleotide binding site of the receptor. Hydrophobic chromatography may be an important adjunct to methods for purification of the progesterone receptor.  相似文献

Invasive alien vertebrates (IAVs) pose a significant threat to island biodiversity worldwide, and their removal is an important nature conservation management goal. As methods advance, eradications from larger islands and of multiple species simultaneously are increasingly undertaken. Effective targeting to maximise conservation gain is important given limited resources. We build on existing prioritisation methods and use the islands of the UK and Crown Dependencies (UK) as an example of how vertebrate eradications might be prioritised and invasive-free status maintained through biosecurity. For each of the 9688 UK islands, we assessed ecological importance for native vertebrates and the anticipated impacts of the IAVs present to estimate the benefit of restoration based on the feasibility and sustainability of IAV eradications in relation to island size, human population and risk of unassisted reinvasion by swimming. As reinvasion poses a threat to the long-term benefits of eradication, we incorporated species-specific swimming distances and explored the effects of varying reinvasion probability from risk-averse to higher-risk strategies. The 25 islands that would benefit most from eradications were in Scotland, Northern Ireland and the Channel Islands. Our prioritisation method should be seen as an initial guide to identify islands that might benefit from intervention when faced with a large list of potential sites. Feasibility studies taking account of factors such as interspecific interactions, anthropogenic reinvasion, views of residents or ‘social feasibility’ and cost need to be undertaken before planning any eradication. We prioritised biosecurity for rat-free islands to highlight where comprehensive measures might be most beneficial.  相似文献
