The contents of macroelements and microelements in ginseng roots and callus cultures was determined by atom absorption spectroscopy. Ginseng cells and tissues were shown to accumulate considerable amounts of microelements. The content of six of the eleven mineral components studied (K, Ca, Na, Mo, Mn, and Cr) in callus cultures was higher than that in roots of agricultural ginseng plants. We revealed good correlations between the contents of microelements (K, Ca, and Mg), as well as between the concentrations of macroelements (Mo, Li, Cu, and Cr), in ginseng cultures. The ability to accumulate elements varied between ginseng species, which was probably related to their genetic features. Our findings indicate that cultured ginseng cells hold much promise as a source of microelements.  相似文献

Recently, we demonstrated the presence of multiple folate-binding sites on the cell surface of Dictyostelium discoideum. These sites were divided into two major classes, with different ligand specificities (A and B). Each major class consists of several interconvertible subtypes. In the present report, the ability of 13 folate analogs to activate both adenylate and guanylate cyclase in pre- as well as postaggregative cells is examined. The patterns of correlation between binding and activation data indicate that guanylate cyclase activation is mediated by the B-sites in both developmental stages (P less than 0.001). In postaggregative cells, adenylate cyclase also seems to be activated by the B-sites (P less than 0.001). In contrast, adenylate cyclase activation in preaggregative cells was well correlated with the specificity of A-sites (P less than 0.01). Remarkably, the potencies of activation were less affected by molecular modifications than the binding affinities were, as suggested by a slope of 0.4 in a plot of K0.5 values of activation vs. binding. This observation argues against the existence of a transduction mechanism in which the response is proportional to receptor occupancy. For the B-receptor, however, the degree of receptor occupancy appears to determine the response. The existence of folic acid antagonists is demonstrated, some of which are specific for either A-sites coupled to adenylate cyclase or for B-sites coupled to guanylate cyclase.  相似文献

Experiments were made on Wistar rats with 6h tourniqueting of the hind limbs to study animal survival rate, myocardial contractile function and protective action of antioxidants and egg phospholipid liposomes during ischemic shock. It has been shown that reperfusion of the limbs leads to a high animal lethality, make lower myocardial contractile function and coronary flow of the hearts isolated from rats following a 6h reperfusion of the limbs. Well-known antioxidant butylated hydroxytoluene and a new antioxidant tetramethylpiperidine derivative bring animal lethality down and improve coronary flow and contractile function of the isolated heart. Phospholipid liposomes increase survival rate moderately but have no any effect on the heart contractile function. It has been deduced that lipid peroxidation takes part in the disturbance of heart contractile function and genesis of the death within ischemic shock.  相似文献

Heterologous expression of a constitutively active calcium-dependent protein kinase (CDPK) gene was previously shown to increase secondary metabolite production in cultured cells of Rubia cordifolia, but the critical question of how CDPK activates secondary metabolism remains to be answered. In this article, we report that the expression of the Arabidopsis CDPK gene, AtCPK1, in R. cordifolia cells caused moderate and stable elevation of intracellular reactive oxygen species (ROS) levels. In contrast, the non-active, mutated AtCPK1 gene did not cause such an effect. The active AtCPK1 also increased cell size, likely by restricting cell division. These results are consistent with the model in which constitutive expression of AtCPK1 mimics the effects of elicitors, acting on secondary metabolism via the activation of ROS production.  相似文献

Caffeoylquinic acids are found in artichokes, and they are currently considered important therapeutic or preventive agents for treating Alzheimer’s disease and diabetes. We transformed artichoke [the cultivated cardoon or Cynara cardunculus var. altilis DC (Asteraceae)] with the rolC gene, which is a known inducer of secondary metabolism. High-performance liquid chromatography with UV and high-resolution mass spectrometry (HPLC-UV-HRMS) revealed that the predominant metabolites synthesized in the transgenic calli were 1,5-dicaffeoylquinic acid, 3,4-dicaffeoylquinic acid, and chlorogenic acid. The rolC-transformed calli contained 1.5 % caffeoylquinic acids by dry weight. The overall production of these metabolites was three times higher than that of the corresponding control calli. The enhancing effect of rolC remained stable over long-term cultivation.  相似文献

The expression of the rolB gene was found to increase the pectic yield in Rubia cordifolia cells, while the rolC gene inhibited the pectin production, which correlated with its expression level. The expression of the rolA, rolB, and rolC genes led to an increase in the content of arabinogalactan (AG) in cells. The increase in the expression of the rolB and rolC genes resulted in a more significant reduction in the content of arabinose residues in pectin, which was accompanied by an increased activity of α-L-arabinofuranosidase in cells. Moreover, the amount of galactose residues in pectin increased with the enhancement of the rolB expression due to a decrease in the activity of β-galactosidase in cells. The content of galacturonic acid residues in pectin from transgenic cultures decreased in the following order: rolC > rolB > rolA. The amount of arabinose residues in AG decreased independently of the gene type. The amount of arabinose residues in AG was found to be considerably reduced when the rolB expression level was increased.  相似文献

The comparative study of adhesive, hemolytic, DNA-ase, lecithinase, antilysozymic, anticomplementary activities of mono- and associated cultures of 57 Enterobacter spp., 61 Citrobacter spp. and 55 Serratia spp. strains, isolated from patients with pyoinflammatory, intestinal and urological diseases is carried out. Different variations of cocultivated bacteria including Enterobacter and Citrobacter, Enterobacter and Serratia, Citrobacter and Serratia are used. It was shown, that cocultivated Enterobacter, Citrobacter and Serratia bacteria increased the persistent properties of mixt cultures.  相似文献

Ecologically tolerable levels (ETL) of environmental factors have been determined for freshwater ecosystems in Asiatic part of Russia and neighboring countries (basins of Ob, Yenisei, Lena, Amur, and Syr-Darya rivers). Inobservance of ecologically tolerable levels degrades ecological status of the ecosystems manifested as deviation from normal saprobic indices of phytoplankton, zooplankton, and periphyton as well as biotic indices of zoobenthos. The basins were compared by the calculated ETL values for over 40 physicochemical indices. The revealed ecologically tolerable levels were compared with the standard maximum allowable concentrations.  相似文献
