Diabetes in pregnancy constitutes an unfavorable environment for embryonic and fetal development, where the child has a higher risk of perinatal morbidity and mortality, with high incidence of congenital malformations and predisposition to long-term metabolic diseases that increase with a hypercaloric diet. To analyze whether hyperglycemia differentially affects proliferation, apoptosis, and mRNA expression in cells from children of normoglycemic pregnancies (NGPs) and diabetes mellitus pregnancies (DMPs), we used umbilical cord Wharton jelly cells as a research model. Proliferation assays were performed to analyze growth and determine the doubling time, and the rate of apoptosis was determined by flow cytometry-annexin-V assays. AMPK, BNIP3, HIF1α, and p53 mRNA gene expression was assessed by semi-quantitative RT-PCR. We found that hyperglycemia decreased proliferation in a statistically significant manner in NGP cells treated with 40?mM D-glucose and in DMP cells treated with 30 and 40?mM D-glucose. Apoptosis increased in hyperglycemic conditions in NGP and DMP cells. mRNA expression of BNIP3 and p53 was significantly increased in cells from DMPs but not in cells from NGPs. We found evidence that maternal irregular metabolic conditions, like diabetes with hyperglycemia in culture, affect biological properties of fetal cells. The genus Oreochromis is among the most popular of the tilapiine cichlid tribe for aquaculture. However, their temperature and hypoxia tolerance, if tested at all, is usually tested at temperatures of 20–25°C, rather than at the considerably higher temperatures of 30–35°C typical of tropical aquaculture. We hypothesized that both larvae and adults of the heat and hypoxia-adapted Tabasco-line of the Nile tilapia Oreochromis niloticus would be relatively hypoxia-tolerant. Oxygen consumption rate (), Q10 and aquatic surface respiration (ASR) was measured using closed respirometry at 2 (c. 0.2 g), 30 (c. 2–5 g), 105 c. (10–15 g) and 240 (c. 250 g) days of development, at 25°C, 30°C and 35°C. at 30°C was inversely related to body mass: c. 90 μM O2 g−1/h in larvae down to c. 1 μM O2 g−1/h in young adults. Q10 for was typical for fish over the range 25–35°C of 1.5–2.0. ASR was exhibited by 50% of the fish at pO2 of 15–50 mmHg in a temperature-dependent fashion. However, the largest adults showed notable ASR only when pO2 fell to below 10 mmHg. Remarkably, pcrit for was 12–17 mmHg at 25–30°C and still only 20–25 mmHg across development at 35°C. These values are among the lowest measured for teleost fishes. Noteworthy is that all fish maintain equilibrium, ventilated their gills and showed routine locomotor action for 10–20 min after ceased at near anoxia and when then returned to oxygenated waters, all fish survived, further indicating a remarkable hypoxic tolerance. Remarkably, data assembled for from >30 studies showed a > x2000 difference, which we attribute to calculation or conversion errors. We used mitochondrial DNA to assess the genetic structure of endangered lake chubsucker Erimyzon sucetta across its Canadian range. We found unique mitochondrial haplotypes in Lyons Creek, a tributary of the Niagara River that faces a strong potential for habitat deterioration. Lyons Creek may therefore serve as a reservoir of unique genetic diversity. The sensitivity of Lyons Creek, combined with the genetic uniqueness of its E. sucetta population, call for further investigation into whether this population should be considered a separate designatable unit for conservation purposes. 相似文献

Phenacoccus solenopsis has been recognized as an aggressively invasive species on cotton plants in different countries. This study was conducted to investigate the effect of green synthesized Cu/Zn-nanoparticles using aqueous leaf extract of Prosopis juliflora (mezquite) against P. solenopsis. The scanning electron microscope (SEM) showed bimetallic nanoparticles of Cu/Zn-NPs with spherical shape with varying size of 74.33 nm to 59.46 nm. More than 30% mortality of P. solenopsis was observed with Cu/Zn-nanoparticles (100 ppm) at 96 hours after treatment. Negligible mortality of P. solenopsis was recorded with Cu/Zn solution (100 ppm) and aqueous P. juliflora extracts. The results of the viability test for Cu/Zn-nanoparticles of P. juliflora showed a significant reduction of the cell viability by 50% in insect exposed to Cu/Zn-nanoparticles-P.juliflora. Therefore studies about nanotoxicity of Cu/Zn-NPs of P.juliflora are needed to reveal the mechanism of toxicity this phytonanoparticles in P.solenopsis. 相似文献
