The aim of this study was to select virulent strains of microfungi against Leucoagaricus gongylophorus, a symbiotic fungus cultivated by leaf-cutting ants. The results from in vitro assays showed that microfungal strains had a variable and significant impact on the colony development of L. gongylophorus. Specifically, Trichoderma harzianum, Escovopsis weberi CBS 810.71 and E. weberi A088 were more effective, inhibiting the L. gongylophorus colonies by 75, 68 and 67%, respectively (P < 0.05) after 15 days. Strain E. weberi A086 and Acremonium kiliense were less effective: 43 and 26%, respectively (P < 0.05). In spite of the current negative perspective of a microbiological control approach for these ants, the present work
discusses the possibility of using mycopathogenic fungi for the control of these insects, and points out the importance of

During early stages of brain development, neuroepithelial stem cells undergo intense proliferation as neurogenesis begins. Fibroblast growth factor 2 (FGF2) has been involved in the regulation of these processes, and although it has been suggested that they work in an autocrine-paracrine mode, there is no general agreement on this because the behavior of neuroepithelial cells is not self-sufficient in explants cultured in vitro. In this work, we show that during early stages of development in chick embryos there is another source of FGF2, besides that of the neuroepithelium, which affects the brain primordium, since the cerebrospinal fluid (E-CSF) contains several isoforms of this factor. We also demonstrate, both in vitro and in vivo, that the FGF2 from the E-CSF has an effect on the regulation of neuroepithelial cell behavior, including cell proliferation and neurogenesis. In order to clarify putative sources of FGF2 in embryonic tissues, we detected by in situ hybridization high levels of mRNA expression in notochord, mesonephros and hepatic primordia, and low levels in brain neuroectoderm, corroborated by semiquantitative PCR analysis. Furthermore, we show that the notochord segregates several FGF2 isoforms which modify the behavior of the neuroepithelial cells in vitro. In addition, we show that the FGF2 ligand is present in the embryonic serum; and, by means of labeled FGF2, we prove that this factor passes via the neuroepithelium from the embryonic serum to the E-CSF in vivo. Considering all these results, we propose that, in chick embryos, the behavior of brain neuroepithelial stem cells at the earliest stages of development is influenced by the action of the FGF2 contained within the E-CSF which could have an extraneural origin, thus suggesting a new and complementary way of regulating brain development. 相似文献

Wolbachia are intracellular bacteria that commonly infect arthropods. Its prevalence among ants of the genus Solenopsis is high. In the present study, the presence and distribution of these endosymbionts was examined among populations of Solenopsis spp. from Brazil. A phylogenetic analysis based on the wsp gene was conducted to infer the evolutionary history of Wolbachia infections within the populations surveyed. A high frequency of Wolbachia bacteria was observed among the genus Solenopsis, 51% of the colonies examined were infected. Incidence was higher in populations from southern Brazil. However, little genetic variability was found among different Wolbachia strains within supergroups A and B. Our findings also suggest that horizontal transmission events can occur through the social parasite S. daguerrei. 相似文献

Zebrafish are currently used at various stages of the drug discovery process and can be a useful and cost-effective alternative to some mammalian models. Nitric oxide (NO) plays an important role in physiology of zebrafish. The availability of appropriate analytical techniques to quantify the NO is crucial for studying its role in physiological and pathological conditions. This work aimed at establishing a high-performance liquid chromatography method for determination of NO levels in zebrafish larvae. Attempts were also made to assess the normal levels of NO at the first days postfertilization and the possible changes under pathological conditions. The method validation was quantitatively evaluated in terms of sensitivity, specificity, precision, accuracy, linearity, and recovery. NO levels from zebrafish larvae at the first days postfertilization and larvae challenged to N(G)-nitro-L-arginine methyl ester, sodium nitroprusside, Escherichia coli lipopolysaccharide, and copper sulfate were analyzed. The samples were derivatized with 2,3-diaminonaphthalene, and fluorescence detection was used for the indirect determination of NO. The method showed a good performance for all validation parameters evaluated and was efficient to monitor changes in NO concentration under physiological and pathophysiological conditions. Pleiotropy has played an important role in understanding quantitative traits. However, the extensiveness of this effect in the genome and its consequences for plant improvement have not been fully elucidated. The aim of this study was to identify pleiotropic quantitative trait loci (QTLs) in maize using Bayesian multiple interval mapping. Additionally, we sought to obtain a better understanding of the inheritance, extent and distribution of pleiotropic effects of several components in maize production. The design III procedure was used from a population derived from the cross of the inbred lines L-14-04B and L-08-05F. Two hundred and fifty plants were genotyped with 177 microsatellite markers and backcrossed to both parents giving rise to 500 backcrossed progenies, which were evaluated in six environments for grain yield and its components. The results of this study suggest that mapping isolated traits limits our understanding of the genetic architecture of quantitative traits. This architecture can be better understood by using pleiotropic networks that facilitate the visualization of the complexity of quantitative inheritance, and this characterization will help to develop new selection strategies. It was also possible to confront the idea that it is feasible to identify QTLs for complex traits such as grain yield, as pleiotropy acts prominently on its subtraits and as this "trait" can be broken down and predicted almost completely by the QTLs of its components. Additionally, pleiotropic QTLs do not necessarily signify pleiotropy of allelic interactions, and this indicates that the pervasive pleiotropy does not limit the genetic adaptability of plants. 相似文献

The purification of IgG from human plasma was studied by comparing two affinity membranes complexed with Ni(II), prepared by coupling iminodiacetic acid (IDA) and Tris(2-aminoethyl)amine (TREN) to poly(ethylenevinyl alcohol), PEVA, hollow fiber membranes. The Ni(II)-TREN-PEVA hollow fiber membrane had lower capacity for human IgG than the complex Ni(II)-IDA-PEVA, but with similar selectivity. The IgG in peak fractions eluted from the Ni(II)-IDA-PEVA with a stepwise concentration gradient of Tris-HCl pH 7.0 (100-700 mM) reached a purity of 98% (based on IgG, IgM, IgA, albumin, and transferrin nephelometric analysis). Adsorption IgG data at different temperatures (4-37 degrees C) were analyzed using Langmuir model resulting in a calculated maximum capacity at 25 degrees C of 204.6 mg of IgG/g of dry membrane. Decrease in Kd with increasing temperature (1.7x10(-5) to 5.3x10(-6) M) indicated an increase in affinity with increased temperature. The positive value of enthalpy change (26.2 kJ/mol) indicated that the adsorption of IgG in affinity membrane is endothermic. Developmental genes are silenced in embryonic stem cells by a bivalent histone-based chromatin mark. It has been proposed that this mark also confers a predisposition to aberrant DNA promoter hypermethylation of tumor suppressor genes (TSGs) in cancer. We report here that silencing of a significant proportion of these TSGs in human embryonic and adult stem cells is associated with promoter DNA hypermethylation. Our results indicate a role for DNA methylation in the control of gene expression in human stem cells and suggest that, for genes repressed by promoter hypermethylation in stem cells in vivo, the aberrant process in cancer could be understood as a defect in establishing an unmethylated promoter during differentiation, rather than as an anomalous process of de novo hypermethylation. 相似文献

Cork oak (Quercus suber) somatic embryos were coated with alginate for the production of synthetic seeds and their storability for commercialization
was investigated. Also, the automatic monitoring of somatic embryo growth with a digital system of image capture was tested.
A power regression model was fitted between size and fresh weight (Adjusted R-squared = 0.96). This method permitted growth
assessment without contamination risk and opens the possibility of an automated control of culture growth for the future up
scaling of plant production. Conversion rate of synthetic seeds was higher on medium supplemented with mineral nutrients than
on medium without nutrients. Also, when the somatic embryos were coated without mineral nutrients added to the capsule, conversion
rate was significantly lower. The addition of sucrose to the capsule had no significant effect on the conversion rate. No
differences were recorded between 50 and 100 mM CaCl2 for capsule complexation. Synthetic seeds were cold stored at 4°C for two months without significant loss of conversion capacity.
The present study reports the first attempts to determine optimal storage time and conditions for conversion of encapsulated

In vertebrates, early brain development takes place at the expanded anterior end of the neural tube, which is filled with embryonic cerebrospinal fluid (E-CSF). Most of the proteins contained within the E-CSF, which play crucial roles in CNS development, are transferred from the blood serum. Two important questions are how E-CSF is manufactured and how its homeostasis is controlled. In this respect, the timing of the blood-CSF barrier formation is controversial. Recently, the concept of a functional dynamic barrier has been introduced. This type of barrier is different from that found in adults and is adapted to the specific requirements and environment of the developing nervous system. In this study, we injected a number of proteins into the outflow of the heart and into the cephalic cavities and examined their transport rate between these two embryo compartments. The results indicated that a functional blood-CSF barrier dynamically controls E-CSF protein composition and homeostasis in chick embryos before the formation of functional choroid plexuses. We also showed that proteins are transferred through transcellular routes in a specific area of the brain stem, close to the ventral mesencephalic and prosencephalic neuroectoderm, lateral to the ventral midline, in particular blood vessels. This study contributes to our understanding of the mechanisms involved in CNS development, as this blood-CSF interface regulates the composition of E-CSF by regulating its specific composition. 相似文献

A central composite experimental design was used to investigate the influence of environmental composting parameters (moisture, aeration, particle size and time) for legume trimming residues, used on soil restoration, on the properties of products obtained (organic matter, Kjeldahl-N, C/N ratio and nitrogen losses (N-losses)) in order to determine the best composting conditions. A second-order polynomial model consisting of four independent process variables was found to accurately describe (the differences between the experimental values and those estimated by using the equations never exceeded 10% of the former) the composting process. Results of the experiment showed that compost with acceptably chemical properties (OM, 85%; Kjeldahl-N, 3.2%), high degradation and minimum N-losses entails operating at high operation time (78 days), low particle size (1cm), medium moisture content (40%) and medium to low aeration level (0.2-0.4 l air/min kg). 相似文献
