Summary The hexokinase (HK) activity in human fibroblasts was close to that expected for a gene dosage effect in a mosaic cell line with about 86% trisomy 10 cells (64% greater than four control lines with normal karyotypes). There was no dosage effect for HK in the cell line that was trisomic for 10q24qter, nor in the cell line monosomic for 10pterp12. The data suggest an assignment of the HK1 locus (the only hexokinase in fibroblasts) to 10p11q23 by exclusion. 相似文献

The association of three pairs of mother-yearling Siberian ibex was recorded by noting the distance between the mothers and daughters at 5-min intervals for 1–2-h observation periods for 1 yr at the Brookfield Zoo. Monthly averages of climatological data were obtained. All three pairs showed a seasonal fluctuation of association, with a higher level during spring and fall-winter and a low degree of sociality in midsummer and late winter. Within the high social periods, short 1–3-wk periods of low association occurred, which were correlated to actual rut and calving times in individual females. The mother-daughter associations were most highly correlated to day length and sunrise of the climatological measures. These data seem to support the original hypothesis that these social periods, which began as developmental associations in the infant, aid in keeping the matrifocal herd structure together despite aggression at the times of rut and calving. It seems also that the social cycling, although linked to reproductive cycling, has its own physiological mechanism. 相似文献

Topical application of juvenile hormone I and III or the hormone analogue methoprene to parasitized Manduca sexta larvae inhibited subsequent emergence of the endoparasitic wasp Apanteles congregatus. Methoprene treatment inhibited wasp emergence in a dose-dependent manner, causing either a delay or total inhibition of emergence. These results were interpreted as reflecting inhibitory effects of juvenile hormone on the second-larval ecdysis of the parasitoid that normally occurs during emergence from the host larva. Parasitoid ecdysis was disrupted even when methoprene was applied to host larvae a few hours prior to the normal expected time of emergence. A correlation between the number of emerging parasitoids and the timing of emergence was seen in methoprene-treated hosts, and few parasitoids emerged after day 9 of the host's fifth-instar. Our findings suggest that the suppression of emergence by juvenile hormone analogues noted in previous studies may be due to a similar inhibitory effect on parasitoid ecdysis. We also observed that parasitoids emerging from hosts treated with a low dose of methoprene (1 μg) later pupated normally but then formed nonviable pupal-adult intermediates. Thus use of this insect growth regulator must be undertaken carefully to prevent possible adverse effects on natural parasitoid populations. 相似文献

E. coli chromosomal DNA wastreated with various Pt co-ordiantion compounds and then used as donor DNA in E. coli transformation. Genetic analysis of transformants obtained with Pt-treated DNA showed effects of cis-diamminedichloroplatinum(II) (cis-Pt(II)) and cis-dimethyl-1,3-diaminopropane CL4 (cis-Pt(IV) (DMDAP) on the processing of DNA. With trans-diamminedichloroplatinum(II) (trans-Pt(II)) appllied in similar concentrations no effects were found.The effects of cis-Pt(II) and cis-Pt(IV) (DMDAP) on the genetic processing were different. Antisera from some hamsters bearing adenovirus-induced tumors contain antibodies to an 11,000 Mr adenovirus-induced protein. In adenovirus-infected HeLa cells, this early viral protein was specifically associated with the nuclear matrix fraction. After two-dimensional gel electrophoresis, two forms of the 11,000 Mr protein at pI 5.6 and pI 5.4 were found. Only the pI 5.4 form of this protein was associated with the nuclear matrix fraction. Adenoviruses from groups A, B, C, D and E all produced an early viral protein (10,000 to 12,000 Mr) that reacted with group C antibody to the 11,000 Mr protein. To date, this is the only known early viral protein that is immunologically conserved in all of the human adenovirus groups.The positions of two methionine and seven leucine residues were determined by sequencing the first 35 amino acids from the N terminus of the adenovirus serotype 2 group C 11,000 Mr protein. The positions of these amino acid residues were compared to the adenovirus serotype 2 nucleotide sequence, which uniquely localized the structural gene of the 11,000 Mr protein to region E4, subregion 3 in type 2 adenovirus. A frameshift mutant, which contained a deletion of one base-pair in the structural gene of the 11,000 Mr protein, was isolated and mapped by marker rescue and nucleotide sequence analysis. This mutant failed to produce immunologically detectable 11,000 Mr protein. The mutant had a viable phenotype, producing normal levels of infectious virus in both HeLa cells and WI38 cells in culture. Abstract: The effect of an inhibitor of N -glycosylation of glycoproteins, tunicamycin, on synthesis of PNS myelin proteins was investigated in vitro by using chopped sciatic nerves or spinal roots of 21-day-old Wistar rats. Tunicamycin when incubated with these nerves in the presence of 3 H-labeled fucose, mannose, or glucosamine inhibited the uptake of radioactivity into myelin proteins including some high-molecular-weight proteins, P0 , 23K protein, and 19K protein by amounts ranging from 42 to 79%. Uptake of 14 Camino acid mixture was inhibited much less by tunicamycin, but a new radioactive protein peak appeared when the protein mixtures had been separated by electrophoresis on polyacrylamide gels in the presence of sodium dodecyl sulfate. This protein ran directly in front of the P0 peak, did not correspond to any bands stained by Fast green, and was not labeled by fucose. This peak appeared in increasing larger proportions with progressive time of incubation of nerves with 3 H amino acids in the presence of tunicamycin. The new protein, which cross-reacts with P0 antiserum, was tentatively identified as a nonglycosylated P0 protein that appears to be almost as well incorporated as P0 into the subcellular fraction containing myelin. The levels of cAMP and cGMP were determined throughout the mitotic cycle of two independent strains of the naturally synchronous slime mold, Physarum polycephalum. The normal range of values was approx. 0.5–2.5 pmoles cAMP/ mg protein and 0.01–0.08 pmoles cGMP/mg protein. In our standard laboratory strain, there was no systematic pattern to the variations in the values within the normal range and no unique time in the cycle when values significantly above normal were noted. In the other strain recently cultured from spherules, elevated levels of cGMP, but not cAMP, were observed in late G2 and in the S phase. The data suggest that elevated levels of these cyclic nucleotides are not required for normal progression of the Physarum mitotic cycle which is unperturbed by artificial synchronizing procedures. 相似文献

Individuals of Arbacia punctulata (Lamarck) from Woods Hole, Massachusetts, and the northeastern Gulf of Mexico were reared from fertilized eggs through metamorphosis under comparable laboratory conditions. Interpopulation differences in spine length development were significant between pure-bred offsprings of these two widely separated geographic areas. Spine lengths of hybrid urchins were intermediate to pure-bred animals. Interpopulation differentiation of specific portions of the genome is proposed to account for the observed phenotypic variation in spine length. 相似文献
