During the period of 1988–2000, three alien gammarid species, i.e. Gammarus tigrinus Sexton, 1939, Pontogammarus robustoides G. O. Sars, 1894, and Dikerogammarus haemobaphes (Eichwald, 1841) were recorded in the Odra estuary. This study, the first record of G. tigrinus and P. robustoides in Poland, focuses on the distribution, abundance, and biomass as well as on population structure of the three new species found in the coastal zone of the estuary. G. tigrinus and P. robustoides were first sampled in the Szczecin Lagoon (mid-part of the estuary) in 1988. The densities of both species were high from the time they were first recorded and exceeded 10 000 ind. m–2, while the biomass was 25 and 140 g m–2, respectively. G.tigrinus occurred at higher densities in northern and eastern parts of the Lagoon, while P. robustoides was particularly abundant in the south-eastern part, most heavily affected by River Odra discharge. In 1999, the third gammarid, D. haemobaphes, was recorded in the southern part of the estuary. Both in 1999 and 2000, the species was found in qualitative samples (dredge collections of Dreissena polymorpha) taken from the western branch of the river. As estimated from quantitative samples, densities and biomass in the eastern branch were rather low and did not exceed 150 ind. m–2 and 2.7 g m–2, respectively. The Szczecin Lagoon population of G. tigrinus produces at least two generations within a year: a summer one and an overwintering one. The latter is dominated by older and larger specimens. The species may reproduce in the Lagoon from April until November.  相似文献

Nerve growth factor (NGF) exerts anti-apoptotic, trophic and differentiating actions on sympathetic neurons and cholinergic cells of the basal forebrain and activates the expression of genes regulating the synthesis and storage of the neurotransmitter acetylcholine (ACh). We have been studying the intracellular signaling pathways involved in this process. Although, in the rat pheochromocytoma cell line PC12, NGF strongly activates the mitogen-activated protein kinase (MAPK) pathway, prolonged inhibition of MAPK kinase (MEK) activity by PD98059 or U0126 did not affect the ability of NGF to up-regulate choline acetyltransferase (ChAT) or to increase intracellular ACh levels. In contrast, the treatment with the phosphatidylinositol 3'-kinase (PI3K) inhibitor LY294002, but not with its inactive analogue LY303511, completely abolished the NGF-induced production of ACh. Inhibition of PI3K also eliminated the NGF effect on the intracellular ACh level in primary cultures of septal neurons from E18 mouse embryos. Blocking the PI3K pathway prevented the activation of cholinergic gene expression, as demonstrated in RT/PCR assays and in transient transfections of PC12 cells with cholinergic locus promoter-luciferase reporter constructs. These results indicate that the PI3K pathway, but not the MEK/MAPK pathway, is the mediator of NGF-induced cholinergic differentiation.  相似文献

In recent years, characterization of a copy number variation (CNV) of the genomic DNA has provided evidence for the relationship of this type of genetic variation with the occurrence of a broad spectrum of diseases, including cancer lesions. Copy number variants (CNVs) also occur in the genomes of healthy individuals as a result of abnormal recombination processes in germ cells and have a hereditary character contributing to the natural genetic diversity. Recent image analysis methods and advanced computational techniques allow for identification of CNVs using SNPs genotyping microarrays based on the analysis of signal intensity observed for markers located in the specific genomic regions. In this study we used CanineHD BeadChip assay (Illumina) to identify both natural and cancer-induced CNVs in the genomes of different dog breeds and in different cancer types occurring in this species. The obtained results showed that structural aberrations are a common phenomenon arising during a tumor progression and are more complex and widespread in tumors of mesenchymal tissue origin than in epithelial tissue originating tumors. The tumor derived CNVs, in comparison to healthy samples, were characterized by larger sizes of regions, higher number of amplifications, and in some cases encompassed genes with potential effect on tumor progression.  相似文献

Background

The human genome contains about 8% of endogenous retroviral sequences originated from germ cell infections by exogenous retroviruses during evolution. Most of those sequences are inactive because of accumulation of mutations but some of them are still capable to be transcribed and translated. The latter are insertionally polymorphic HERV-K113 and HERV-K115. It has been suggested that their presence and expression was connected with several human diseases. It is also believed that they could interfere with the replication cycle of exogenous retroviruses, including HIV.

Results

Prevalence of endogenous retroviral sequences HERV-K113 and HERV-K115 was determined in the Polish population. The frequencies were found as 11.8% for HERV-K113 and 7.92% for HERV-K115. To verify the hypothesis that the presence of these HERVs sequences could affect susceptibility to HIV infection, comparison of a control group (HIV-negative, not exposed to HIV; n = 303) with HIV-positive patients (n = 470) and exposed but uninfected (EU) individuals (n = 121) was performed. Prevalence of HERV-K113 and HERV-K115 in the EU group was 8.26% and 5.71%, respectively. In the HIV(+) group we detected HERV-K113 sequences in 12.98% of the individuals and HERV-K115 sequences in 7.23% of the individuals. There were no statistically significant differences between groups studied.

Conclusion

The frequency of HERV-K113 and HERV-K115 sequences in Poland were found to be higher than usually shown for European populations. No relation between presence of the HERVs and HIV infection was detected.  相似文献

The aim of the study was a quantitative and qualitative analysis of microflora, presentation of current data about prevalence of the microflora on the skin and mucous membranes, and determination of its possible effect on reproduction of English full blood horses bred in Poland. The material for analyses was sampled from the skin and mucous membranes (385 samples) of 55 English full blood mares. Taking into account reproduction traits, the mares were classified into three groups. Six yeast-like species, including five species from the genus Candida (C. albicans, C. guiliermondii, C. lusitaniae, C. sp., and C. tropicalis) and Trichomonascus ciferrii, were detected on the skin and mucous membranes in the English full blood mares. Growth of yeasts was observed in more than half of the samples taken from mares that had foaled and approximately 46 % of non-conceiving and barren mares. The high prevalence of various yeast strains in the mouth, nostrils, and collateral groove may suggest widespread occurrence of the microflora in the breeding environment. The results obtained indicate that the yeasts isolated in this study may be components of the normal microflora of the skin and mucous membranes in horses. The analysis results do not indicate unambiguously that the isolated microflora affects reproduction in mares, although this cannot be excluded.  相似文献

INTRODUCTION: Patients with differentiated thyroid carcinoma have to undergo radical surgical treatment, which includes total thyreoidectomy, radioiodine therapy and a life-time suppressive therapy with L-thyroxine. The aim of this study was a prospective evaluation of left ventricular hypertrophy during L suppressive-thyroxine treatment in patients treated for differentiated thyroid carcinoma. MATERIAL AND METHODS: The examined group comprised 50 patients with differentiated thyroid carcinoma, treated by total thyroidectomy and 131I therapy. Echocardiographic measurements were needed for estimation of left ventricular mass and its index, according to recommendations of American Echocardiography Society. RESULTS: During two-years long suppressive therapy we observed a significant rise in left ventricular mass. In woman group left ventricular mass was increased from 168+/-39 g to 204+/-45 g (p<0.001) and in men from 205+/-60 to 320+/-21 g. Likewise, left ventricular mass index was increased in women group from 96+/-18 g/m(2) to 116+/-25 g/m(2) (p<0.001) and in men group from 107+/-37 g/m(2) to 158+/-28 g/m(2). Simultaneous treatment with bisoprolol caused a regression of left myocardial hypertrophy. Already after 6 months of simultaneous treatment with L-thyroxin and bisoprolol, for left ventricular mass was reduced to normal: in woman 165+/-35 g, and in men to 178+/-38 g. Analogous results were obtained left ventricular mass index. After 6 months it was reduced to 94+/-12 g/m(2) in woman and in men to 132+/-32 g/m(2). CONCLUSIONS: 1. In differentiated thyroid cancer patients, treated postoperatively with L-thyroxine suppressive therapy, left ventricular hypertrophy is observed already during the first year of suppressive therapy and progresses during the next year of treatment. 2 Addition of a beta-adrenergic antagonist to suppressive doses of L-thyroxine causes a regression of left ventricular hypertrophy, thus, beta-adrenergic antagonists should be administered in this group of patients.  相似文献

We analyzed the transport of KCl solutions through the bacterial cellulose membrane and concentration boundary layers (CBLs) near membrane with pressure differences on the membrane. The membrane was located in horizontal-plane between two chambers with different KCL solutions. The membrane was located in horizontal-plane between two chambers with different KCL solutions. As results from the elaborated model, gradient of KCL concentration in CBLs is maximal at membrane surfaces in the case when pressure difference on the membrane equals zero. The amplitude of this maximum decreases with time of CBLs buildup. Application of mechanical pressure gradient in the direction of gradient of osmotic pressure on the membrane causes a shift of this maximum into the chamber with lower concentration. In turn, application of mechanical pressure gradient directed opposite to the gradient of osmotic pressure causes the appearance of maximum of concentration gradient in chamber with higher concentration. Besides, the increase of time of CBLs buildup entails a decrease of peak height and shift of this peak further from the membrane. Similar behavior is observed for distribution of energy dissipation in CBLs but for pressure difference on the membrane equal to zero the maximum of energy dissipation is observed in the chamber with lower concentration. We also measured time characteristics of voltage in the membrane system with greater KCl concentrations over the membrane. We can state that mechanical pressure difference on the membrane can suppress or strengthen hydrodynamic instabilities visible as pulsations of measured voltage. Additionally, time of appearance of voltage pulsations, its amplitude, and frequency depend on mechanical pressure differences on the membrane and initial quotient of KCl concentrations in chambers.  相似文献

Recently, there have been indications of participation of mitochondria in the carcinogenic process and the role of polymorphisms in increasing the risk of cancer. The aim of the study was to detect possible changes in the sequence of 22 genes encoding mitochondrial tRNA in breast cancer carcinoma, which take part in protein synthesis in the translation process. The analysis of tumour tissue and blood material sampled from 50 patients revealed that few mutations have been found. It cannot be excluded that, through their impact on the secondary and tertiary tRNA structure, polymorphisms may cause mitochondrial dysfunction and contribute to appearance of other changes in mtDNA. Mutations in tRNA genes in breast cancer may affect the cell and cause its dysfunction, as in mitochondrial diseases.  相似文献

Raccoon dogs' mitochondrial DNA genes (MT-CYTB, MT-CO1 and MT-CO2) containing a total of 1.5?kbp exhibited 11 synonymous substitutions in the polymorphism described, with almost 73% found in the gene MT-CO2. The polymorphism was observed in 27.3% of the loci in wild-living animals, and in over 90% of the loci in farm raccoon dogs. Seven mitochondrial haplogroups have been determined, among which three (Np1, Np2 and Np4) were found in the wild-living raccoon dogs and the other four (Np3, Np5, Np6 and Np7) in the farm animals. The occurrence of new haplogroups in the farm animals indicates the appearance of adaptive mutations. Differences were observed between the sequence under study and the reference sequence in MT-CYTB; they involved two non-synonymous substitutions (T304I and F332L). Analyses carried out to determine the deleterious effect of mutations indicated an almost 50% probability that a single amino acid substitution (T304I) in the protein has a negative impact on its function.  相似文献
