A previously unrecognized passive transport for pyridine dinucleotides has been described recently in the plasmamembrane of several mammalian cells. Despite elucidation of some functional and kinetic properties of this transport system, it is still undefined at the molecular level. Therefore, we have addressed the molecular characterization of the NAD+ transporter and identified it as connexin 43 (Cx43). This is a structural component of hexameric hemichannels that, when juxtaposed on adjacent cells, builds up intercellular gap junctions and mediates exchange of molecules between cells. However, the role of connexin hemichannels as potential pores in individual, noncoupled cells remains elusive. Bidirectional NAD+ transport in isolated Cx43-expressing mur ine 3T3 fibroblasts was affected by known modulators of connexin-mediated intercellular coupling and was completely inhibited by treatment of the cells with a Cx43-antisense oligonucleotide. NAD+ transport in proteoliposomes reconstituted with 3T3 membrane proteins was inhibited in the presence of a monoclonal anti-Cx43 antibody. Finally, Cx43 immunopurified to homogeneity was reconstituted in unilamellar proteoliposomes, which displayed full NAD+-transporting activity. This finding is the first evidence that connexin hemichannels can mediate transmembrane fluxes of a nucleotide in whole cells: The pleiotropy of NAD+-dependent cellular events, including redox reactions, signaling, and DNA repair, implicates Cx43 hemichannels in intercellular NAD+ trafficking, which suggests new paracrine functions of NAD. 相似文献

By combining in silico and bench molecular biology methods we have identified a novel human gap junction gene that encodes a protein designated HCx31.9. We have determined its human chromosomal location and gene structure, and we have identified a putative mouse ortholog, mCx30.2. We have observed the presence of HCx31.9 in human cerebral cortex, liver, heart, spleen, lung, and kidney and the presence of mCx30.2 in mouse cerebral cortex, liver and lung. The hypothesis that mature egg availability during a parasitoid's lifetime affects the functional response, as well as a parasitoid's handling time and searching efficiency, was tested. Campoletis grioti Blanchard (Hym: Ichneumonidae) female parasitoids and Spodoptera frugiperda Smith (Lep: Noctuidae) host larvae were used as an example. Each female of C. grioti was confined with different host densities (6, 10, 30, 50, 80, 120 and 160 larvae/day) of first and second S. frugiperda instars until the female died. We found that C. grioti showed a type II functional response, with a reduction in the number of parasitized hosts in relation to female ageing. Handling time and searching efficiency increased over time. When we standardized the data simulating unlimited egg supply, the functional response, handling time and searching efficiency became constant throughout the parasitoid's life. We discuss the relationship between egg availability and functional response and its associated parameters, and its consequence on biological control programs. 相似文献

The contents of three major digestive enzymes (amylase, lipase and chymotrypsinogen) were measured in the obese Zucker rat. Only minimal changes were found in 7-week-old rats, but in adult obese rats (14-16 weeks) the amylase content was decreased by 50%, whereas the lipase and chymotrypsinogen contents were increased by 45% and 20%, respectively, compared with lean controls. Abnormalities of enzyme secretion were also found. Since the changes observed in enzyme proportions in adult obese Zucker rats are qualitatively similar to those observed in insulinopenic diabetes and other states associated with decreased glucose metabolism, it is speculated that the abnormalities found in the obese Zucker rat may be due to decreased glucose metabolism in the exocrine tissue consequent to insulin resistance. 相似文献

The incubation of human bone marrow fibroblasts with hydrocortisone (10(-7) M) produces a time-dependent depletion of hydrocortisone-binding sites. This effect is glucocorticoid-specific, since glucocorticoid agonists cause depletion to the same extent as hydrocortisone. After withdrawal of the hormone from the incubation medium, cells are able to replenish their complement of receptor. The existence of this down-regulation process may represent a protective mechanism against the partial proliferative inhibitory action of glucocorticoids for bone marrow fibroblasts and also a stimulus for differentiation to adipocytes. 相似文献
