Complement regulator C4b-binding protein (C4BP) and the anticoagulant vitamin K-dependent protein S form a high affinity complex in human plasma. C4BP is composed of seven alpha-chains and a unique beta-chain, each chain comprising repeating complement control protein (CCP) modules. The binding site for protein S mainly involves the first of the three beta-chain CCPs (CCP1). However, recently it has been suggested that CCP2 of the beta-chain also contributes to the binding of protein S. To elucidate the structural background for the involvement of CCP2 in the protein S binding, several recombinant beta-chain CCP1-2 variants having mutations in CCP2 were expressed and tested for protein S binding. Mutations were chosen based on analysis of a homology model of the beta-chain and included R60A/R101A, D66A, L105A, F114A/I116A and H108A. All mutant proteins bound equally well as recombinant wild type to protein S. Several monoclonal antibodies against the beta-chain CCP2 were raised and their influence on protein S binding characterized. Taken together, the results suggest that the role of CCP2 in protein S binding is to orient and stabilize CCP1 rather than to be directly part of the binding site.  相似文献

The interest for lanthanide circularly polarized luminescence (CPL) has been quickly growing for 10 years. However, very few of these studies have involved correlation between the dissymmetry factor (glum) and the chemical modifications in a series of chiral ligands. Four polymeric compounds of Eu(III) were prepared by using a series of binaphtyl derivatives for which the size of the π system as well as the number of stereogenic elements (i.e., the binaphtyl moiety) are modulated. The resulting {[Eu(hfac)3((S)/(R)-Lx)]}n (x = 1 and 3) and {[Eu(hfac)3((S,S,S)/(R,R,R)-Lx)]}n (x = 2 and 4) have been characterized by powder X-ray diffraction by comparison with the X-ray structures on single crystal of the Dy(III) analogs. In solution, the structure of the complexes is deeply modified and becomes monomeric. The nature of the ligand induces change in the shape of the CPL spectra in CH2Cl2 solution. Furthermore, a large |glum| = 0.12 of the magnetic-dipole transition for the [Eu(hfac)3((S,S,S)/(R,R,R)-L2)] complex involving the ligand with three stereogenic elements and an extended ?? system has been measured. This report also shows CPL measurements in solid state for the series of {[Eu(hfac)3((S)/(R)-Lx)]}n (x = 1 and 3) and {[Eu(hfac)3((S,S,S)/(R,R,R)-Lx)]}n (x = 2 and 4) polymers.  相似文献

In unseren Untersuchungen prüften wir den Einfluß einer Saatgutbehandlung des Maises mit Imidacloprid (Gaucho) sowohl auf die Übertragung des persistenten barley yellow dwarf virus (BYDV) als auch auf die nichtpersistenten Potyviren maize dwarf mosaic virus (MDMV) und sugarcane mosaic virus (SCMV). Dazu wurden Freiland‐ und Gewächshausversuche sowie Experimente in einer Klimakammer angestellt. Für die Virusübertragungen nutzten wir die Blattlausarten Rhopalosiphum padi L. und weniger häufig Sitobion avenae F. Im Vergleich zu S. avenae wanderte R. padi schneller von den behandelten Pflanzen ab. In Gewächshausversuchen zeigte sich, daß das SCMV nach einer Saatgutbehandlung in deutlich geringerem Umfang übertragen wurde, wenn die Inokulation bis 3 Wochen nach dem Auslegen erfolgte. Inokulationen von älteren Pflanzen ließen keine insektizide Wirkung erkennen. Ebenso ergaben Freilandversuche keinen eindeutigen Zusammenhang zwischen Saatgutbehandlung und Virusbefall. Im Falle des BYDV konnte bei einer Inokulation bis 9 Tage nach dem Auslegen durch einen Imidacloprid‐Einsatz eine signifikante Reduzierung der Übertragung erreicht werden. Spätere Inokulationen verliefen in der Regel negativ. Die Ergebnisse des Feldversuches 1992 bestätigten die im Gewächshaus gewonnenen Resultate. Im Freiland war die natürliche Besiedlung des Maises durch Blattläuse in den Parzellen mit behandeltem Saatgut deutlich vermindert. Hinzu kam eine zeitliche Verzögerung des Massenwechselgeschehens um ca. 2 Wochen.

Für eine abschließende Beurteilung von Imidacloprid sind weiterführende Untersuchungen sowohl zum Saugverhalten der Blattläuse als auch zu seiner Wirkung auf die Virusübertragung unter Einbeziehung verschiedener Sorten erforderlich.  相似文献

Pelagia noctiluca is the most venomous jellyfish in the Mediterranean Sea where it forms dense blooms. Although there is several published research on this species, until now none of the works has been focused on a complete protein profile of the all body constituents of this organism. Here, we have performed a detailed proteomics characterization of the major protein components expressed by P. noctiluca. With that aim, we have considered the study of jellyfish proteins involved in defense, body constituents and metabolism, and furthered explore the significance and potential application of such bioactive molecules. P. noctiluca body proteins were separated by1D SDS–PAGE and 2DE followed by characterization by nanoLC-MS/MS and MALDI-TOF/TOF techniques. Altogether, both methods revealed 68 different proteins, including a Zinc Metalloproteinase, a Red Fluorescent Protein (RFP) and a Peroxiredoxin. These three proteins were identified for the first time in P. noctiluca. Zinc Metalloproteinase was previously reported in the venom of other jellyfish species. Besides the proteins described above, the other 65 proteins found in P. noctiluca body content were identified and associated with its clinical significance. Among all the proteins identified in this work we highlight: Zinc metalloproteinase, which has a ShK toxin domain and therefore should be implicated in the sting toxicity of P. noctiluca.; the RFP which are a very important family of proteins due to its possible application as molecular markers; and last but not least the discovery of a Peroxiredoxin in this organism makes it a new natural resource of antioxidant and anti-UV radiation agents.  相似文献

Purpose

Despite a mature debate on the importance of a time-dependent account of carbon fluxes in life cycle assessments (LCA) of forestry products, static accounts of fluxes are still common. Time-explicit inventory of carbon fluxes is not available to LCA practitioners, since the most commonly used life cycle inventory (LCI) databases use a static approach. Existing forest models are typically applied to specific study fields for which the detailed input parameters required are available. This paper presents a simplified parametric model to obtain a time-explicit balanced account of the carbon fluxes in a forest for use in LCA. The model was applied to the case of spruce as an example.

Methods

The model calculated endogenous and exogenous carbon fluxes in tons of carbon per hectare. It was designed to allow users to choose (a) the carbon pools to be included in the analysis (aboveground and belowground carbon pools, only aboveground carbon or only carbon in stem); (b) a linear or sigmoidal dynamic function describing biomass growth; (c) a sigmoidal, negative exponential or linear dynamic function describing independently the decomposition of aboveground and belowground biomass; and (d) the forest management features such as stand type, rotation time, thinning frequency and intensity.

Results and discussion

The parametric model provides a time-dependent LCI of forest carbon fluxes per unit of product, taking into account the typically limited data available to LCA practitioners, while providing consistent and robust outcomes. The results obtained for the case study were validated with the more complex CO2FIX. The model ensures carbon balance within spatial and time delimitation defined by the user by accounting for the annual biomass degradation and production in each carbon pool. The inventory can be used in LCA studies and coupled with classic indicators (e.g. global warming potential) to accurately determine the climate impacts over time. The model is applicable globally and to any forest management practice.

Conclusions

This paper proposes a simplified and flexible forest model, which facilitates the implementation in LCA of time-dependent assessments of bio-based products.

Olive is one of the most important tree crops in the Mediterranean region, because of its ability to grow and produce acceptable yields under limited water availability. In this study, the drought tolerance of an olive cultivar Canino was compared to the performance of its derived transgenic line expressing osmotin gene from tobacco, obtained by Agrobacterium-mediated transformation of Canino cultivar. Shoot cultures of both wild-type (wt) and transgenic lines were exposed to drought stress over a 28-day period, and their differential responses to in vitro-drought stress were investigated. After exposure to PEG, most of the shoots from wt plants resulted in damage and exhibited decreased levels of chlorophyll, while those of transgenic line did not show injuries and showed a normal growth even when exposed to the highest PEG concentration (4%). After preliminary evaluation we characterized Canino AT17-1, by measuring several physiological parameters, including the activities of the antioxidant enzymes (POD and CAT), and the content of malondialdehyde (MDA). Both the activity of catalase and the proline content were higher in the leaves of the transgenic shoots compared to wt plants. Consequently, it was observed that the transgenic line accumulated less MDA indicating that the presence of the osmotin gene protected the cell membrane from damage by lipid peroxidation. Together, these results could suggest that the transgenic line Canino AT17-1 was more efficient in the activation of defense responses against oxidative stress with respect to the Canino wt. The further finding that the transgenic shoots also showed higher proline accumulation supported the hypothesis that the osmotin gene conferred to transgenic shoots increased tolerance to drought stress compared with the wt.  相似文献

Background

Vitamin D (VitD) and parathyroid hormone (PTH) play important roles in calcium metabolism and skeletal homeostasis. Estimates of the VitD status in several European countries show large variations between them. In addition, no national population-based estimate has been published. VitD and PTH may also play important roles in cardiovascular risk, which has been suggested to be associated with metabolic syndrome (MetS) and is very prevalent in Portugal.The goal of our study was to evaluate the prevalence of hypovitaminosis D and its determinants as well as PTH serum level determinants and associations of the 25-hydroxyvitamin D and PTH serum levels with MetS and its individual components in a sample of the Portuguese mainland population.

Methods

PORMETS is a national cross-sectional study that includes a total sample of 4095 adults. A subsample, including 500 participants, was randomly selected for the present study. A structured questionnaire was administered to collect information on personal medical histories and socio-demographic and behavioral characteristics. Blood pressure and anthropometrics measurements were performed. Fasting venous samples were collected and PTH and 25-hydroxyvitamin D were measured. VitD adequacy was classified according to the Institute of Medicine, and MetS was classified according to the Joint Interim Statement recommendations. Multiple linear regression and unconditional logistic regression models were used to estimate the associations between the levels of PTH and 25-hydroxyvitamin D and with MetS and its individual components.

Results

The prevalence of VitD deficiency was 37.7%, and MetS was present in 191 participants (38.4%). The serum PTH levels showed a positive association (OR: 1.014; 95%CI: 1.002, 1.026) with the waist circumference component of MetS. The serum 25-hydroxyvitamin D levels were negatively associated with MetS (OR: 0.957; 95%CI: 0.922, 0.993) as well as with its blood pressure (OR: 0.949; 95%CI: 0.912, 0.987) and triglycerides (OR: 0.930; 95%CI: 0.892, 0.969) components.

Conclusion

This study showed a high national prevalence of hypovitaminosis D. The PTH levels showed a significant positive association with the WC component of MetS, and the VitD levels were negatively associated with the BP and triglycerides components as well as with the MetS.

Phosphodiesterase 4 (PDE4) inhibitors have attractive therapeutic potential in respiratory, inflammatory, metabolic and CNS disorders. The present work details the design, chemical exploration and biological profile of a novel PDE4 inhibitor chemotype. A diazepinone ring was identified as an under-represented heterocyclic system fulfilling a set of PDE4 structure-based design hypotheses. Rapid exploration of the structure activity relationships for the series was enabled by robust and scalable two/three-steps parallel chemistry protocols. The resulting compounds demonstrated PDE4 inhibitory activity in cell free and cell-based assays comparable to the Zardaverine control used, suggesting potential avenues for their further development.  相似文献

Objective To test the hypothesis that the identification of mutation in the carboxypeptidase E (CPE) gene which leads to marked hyperproinsulinaemia is consistent with a possible role for mutations in CPE in the development of coronary heart disease. Methods The study subjects consisted of 51 consecutive patients (34 males and 17 females) who will undergo coronary angiography for suspected or known coronary atherosclerosis. Coronary heart disease (CHD) was defined as having a luminal diameter stenosis ≥50% in at least one of three major coronary arteries by coronary angiography or based on the Rose Questionnaire. The insulin and proinsulin level were measured using highly sensitive two-site sandwich ELISA methods. Screening for mutations of the eight exons of the CPE gene was performed by polymerase chain reaction followed by bidirectional sequencing. Results We scanned eight exons and exon–intron junctional region. Overall, we found 12 distinct variants in the intron region and three variants in the exon region. Among the 15 variants, 10 mutations were rare. The further explored study reveal that the above five non-rare variants would not affect the level of glucose, insulin, and proinsulin. However, the results suggest that the prevalence of the coronary heart disease was significant difference between the wild type group and mutant type group according to the A4545G (P = 0.020). The results from the logistic regression reveal that the subjects with the CPE mutation of A4545G, the odds ratio for the coronary heart disease was 0.196 (95% CI: 0.046 to 0.830, P = 0.027). Conclusions In the present study, the mutation of CPE gene would not affect the level of glucose, insulin, and proinsulin. The hypothesis of a possible role for mutations in CPE in the development of coronary heart disease needs further study.  相似文献

Heme carrier HasA has a unique type of histidine/tyrosine heme iron ligation in which the iron ion is in a thermally driven two spin states equilibrium. We recently suggested that the H-bonding between Tyr75 and the invariantly conserved residue His83 modulates the strength of the iron-Tyr75 bond. To unravel the role of His83, we characterize the iron ligation and the electronic properties of both wild type and H83A mutant by a variety of spectroscopic techniques. Although His83 in wild type modulates the strength of the Tyr-iron bond, its removal causes detachment of the tyrosine ligand, thus giving rise to a series of pH-dependent equilibria among species with different axial ligation. The five coordinated species detected at physiological pH may represent a possible intermediate of the heme transfer mechanism to the receptor.  相似文献
