The serum prostaglandin (PG) F2alpha levels of women in the last trimester of pregnancy have been measured and compared with the levels found in samples taken during labor, during the 1st week postpartum, and from nonpregnant women. The laboratory procedures for drawing the samples and making the radioimmunoassay measurements are explained. 40 samples were taken during late pregnancy, 46 during labor, 7 during the 1st week postpartum, and 8 from nonpregnant women. The results are graphed. PGF2alpha levels during late pregnancy resemble those found in nonpregnant women. A significant rise (p.05) occurs during labor, most markedly during the late 1st stage. Levels during the 1st week postpartum were found to be very low. There was no difference between serum levels during the 33rd-36th weeks and those found during weeks 34-40. Serum levels did not differ according to whether labor was spontaneous, induced by artificial rupture of fetal membranes and infusion of Syntocinon, or merely accelerated by syntocinon infusion. This indicates that PG release occurs as a result of labor rather than as a triggering mechanism for the onset of labor. The highest levels of serum PGF2alpha seem to be associated with the time of greatest uterine activity and the period of greatest cervical stretching.  相似文献

The shanny Lipophrys pholis is an intertidal fish commonly found in Portuguese coastal waters. Spawning takes place from early autumn to mid spring, after which demersal eggs hatch and larvae disperse along the coast. Two to three months later, young juveniles return to the tide pools to settle. However, information on fish movement, habitat connectivity and population structure is scarce for this species. One hundred and twenty early juveniles (16–35?mm) were collected in April 2014 from six rocky beaches along the western and south Portuguese coasts (Agudela, Cabo do Mundo, Boa Nova, Peniche, Sines and Olhos de Água). δ18O and δ13C were determined by isotope-ratio mass spectrometry. Data were analysed to determine whether isotopic signatures could be used to assess the degree of separation between individuals collected from different locations. Mean δ13C and δ18O values ranged from ?0.02‰ to 1.14‰ and ?7.77‰ to ?6.62‰, respectively. Both seawater temperature and salinity caused differences in otolith δ18O among the four main sampling areas. The variation among areas in δ13C was most likely related to slight differences in the diet, growth and metabolism of fish. The distinct isotopic signatures, at least for the northern and central areas, suggested low levels of connectivity across large spatial scales during the juvenile stage. Furthermore, similar isotopic signatures within the same area indicated some degree of larval oceanic retention at short spatial scales. This study suggests that stable isotope records in otoliths could provide information about the home residency, movements and habitat connectivity of intertidal fishes.  相似文献

Caffeine is suspected to affect gastrointestinal function. We therefore investigated whether supplementation of a carbohydrate-electrolyte solution (CES) sports drink with 150 mg/l caffeine leads to alterations in gastrointestinal variables compared with a normal CES and water using a standardized rest-exercise-rest protocol. Ten well-trained subjects underwent a rest-cycling-rest protocol three times. Esophageal motility, gastroesophageal reflux, and intragastric pH were measured by use of a transnasal catheter. Orocecal transit time was measured using breath-H(2) measurements. A sugar absorption test was applied to determine intestinal permeability and glucose absorption. Gastric emptying was measured via the (13)C-acetate breath test. In the postexercise episode, midesophageal pressure was significantly lower in the CES + caffeine trial compared with the water trial (P = 0.017). There were no significant differences between the three drinks for gastric pH and reflux during the preexercise, the cycling, and the postexercise episode, respectively. Gastric emptying, orocecal transit time, and intestinal permeability showed no significant differences between the three trials. However, glucose absorption was significantly increased in the CES + caffeine trial compared with the CES trial (P = 0.017). No significant differences in gastroesophageal reflux, gastric pH, or gastrointestinal transit could be observed between the CES, the CES + caffeine, and the water trials. However, intestinal glucose uptake was increased in the CES + caffeine trial.  相似文献

It has been reported that a muramyl dipeptide (MDP) treatment regimen (200 micrograms MDP per mouse, Days -4, -3, -2, and -1) that, given prophylactically, affords protection against several infectious agents also induces lymph node hyperplasia, lymph node cell (LNC) hyperresponsiveness to mitogens, and spleen cell hyporesponsiveness to mitogens. The purpose of the present work was to extend those studies and delineate cellular mechanisms involved in these phenomena. It has been found that hyperresponsiveness of LNC was prolonged (7 days) posttreatment; in contrast, hyporesponsiveness of spleen cells was transient and rebounded by Day 4 posttreatment. Hyperresponsiveness of LNC and hyporesponsiveness of spleen cells actively enhanced and depressed normal lymphoid cell responses, respectively, in cell mixing experiments. Hyporesponsiveness of spleen cells was associated with the plastic-nonadherent, non-B-cell fraction and nylon wool-nonadherent subpopulations. Indomethacin (10(-6) M) did not abrogate hyporesponsiveness of spleen cells. These data suggest that splenic suppressor T cells result from MDP treatment and were responsible for spleen cell hyporesponsiveness. On the other hand, hyperresponsiveness of LNC was associated with the nylon wool-adherent cell subpopulations and a higher percentage of nonspecific esterase-positive cells. Hyporesponsiveness of spleen cells was associated with deficient production of interleukin 2 (IL-2), but not of interleukin 1 (IL-1). In contrast, hyperresponsiveness of LNC was not explained by enhanced IL-1 or IL-2 production.  相似文献

It is known thatHistoplasma capsulatum can resist the intraphagolysosomal environment and multiply inside macrophages. This resistance can be closly related to its pathogenicity. The mechanism of this resistance has been investigated, but it has not been clarified as yet. To learn about the metabolic condition of the yeast-form ofH. capsulatum (isolates G217B and CDC 105) when ingested by macrophages, we investigated protein synthesis by ingestedH. capsulatum with [35S]-methionine labeling. Cycloheximide at 5 to 10 µg/ml was used to preferentially inhibit macrophage uptake of [35S]-methionine without affectingH. capsulatum uptake. Protein synthesis byH. capsulatum in medium alone served as a positive control. The negative control consisted of macrophages with ingested heat-killedH. capsulatum. Analysis of cytosols with SDS-PAGE and fluorography disclosed that, respectively for G217B and CDC 105, ingestedH. capsulatum synthesized 4 and 5 novel proteins, increased the synthesis of 9 and 17 proteins and decreased the synthesis of 9 and 10 constitutive proteins. Ten of these novel or increased proteins were apparently common to both strains. These metabolic changes in ingestedH. capsulatum could reflect its adaptation to the intraphagolysosomal environment of macrophages and its ability to multiply there.  相似文献

Genotyping-by-sequencing (GBS) is a rapid and cost-effective genome-wide genotyping technique applicable whether a reference genome is available or not. Due to the cost-coverage trade-off, however, GBS typically produces large amounts of missing marker genotypes, whose imputation becomes therefore both challenging and critical for later analyses. In this work, the performance of four general imputation methods (K-nearest neighbors, Random Forest, singular value decomposition, and mean value) and two genotype-specific methods (“Beagle” and FILLIN) was measured on GBS data from alfalfa (Medicago sativa L., autotetraploid, heterozygous, without reference genome) and rice (Oryza sativa L., diploid, 100 % homozygous, with reference genome). Alfalfa SNP were aligned on the genome of the closely related species Medicago truncatula L.. Benchmarks consisted in progressive data filtering for marker call rate (up to 70 %) and increasing proportions (up to 20 %) of known genotypes masked for imputation. The relative performance was measured as the total proportion of correctly imputed genotypes, globally and within each genotype class (two homozygotes in rice, two homozygotes and one heterozygote in alfalfa). We found that imputation accuracy was robust to increasing missing rates, and consistently higher in rice than in alfalfa. Accuracy was as high as 90–100 % for the major (most frequent) homozygous genotype, but dropped to 80–90 % (rice) and below 30 % (alfalfa) in the minor homozygous genotype. Beagle was the best performing method, both accuracy- and time-wise, in rice. In alfalfa, KNNI and RFI gave the highest accuracies, but KNNI was much faster.  相似文献

Despite being mutated in cancer and RASopathies, the role of the activation segment (AS) has not been addressed for B‐Raf signaling in vivo. Here, we generated a conditional knock‐in mouse allowing the expression of the B‐RafAVKA mutant in which the AS phosphoacceptor sites T599 and S602 are replaced by alanine residues. Surprisingly, despite producing a kinase‐impaired protein, the BrafAVKA allele does not phenocopy the lethality of Braf‐knockout or paradoxically acting knock‐in alleles. However, BrafAVKA mice display abnormalities in the hematopoietic system, a distinct facial morphology, reduced ERK pathway activity in the brain, and an abnormal gait. This phenotype suggests that maximum B‐Raf activity is required for the proper development, function, and maintenance of certain cell populations. By establishing conditional murine embryonic fibroblast cultures, we further show that MEK/ERK phosphorylation and the immediate early gene response toward growth factors are impaired in the presence of B‐RafAVKA. Importantly, alanine substitution of T599/S602 impairs the transformation potential of oncogenic non‐V600E B‐Raf mutants and a fusion protein, suggesting that blocking their phosphorylation could represent an alternative strategy to ATP‐competitive inhibitors.  相似文献

The carboxylic ionophore, monensin, rapidly induced cell-wall acidification and a decrease in cytosolic pH when added to maize coleoptiles at low external pH and Na+ concentration. Elongation growth at rates equivalent to those obtained with indole-3-acetic acid was induced for about 1 h. Stimulation of the outwardly directed proton pump apparently occurred, since under the same conditions monensin induced membrane hyperpolarization of maize root rhizodermis cells. When the external pH was high (>8) and Na+ present, monensin treatment caused only minimal changes in membrane potential and cytosolic pH. Although the ionophore transported protons out of the cell, resulting in cell-wall acidification, no elongation growth occurred. However, under identical conditions, indole-3-acetic acid dit induce growth.
