Background

Genetic engineering offers the opportunity to generate unique genetic variation that is either absent in the sexually compatible gene pool or has very low heritability. The generation of transgenic plants, coupled with breeding, has led to the production of widely used transgenic cultivars in several major cash crops, such as maize, soybean, cotton and canola. The process for regulatory approval of genetically engineered crops is slow and subject to extensive political interference. The situation in forage grasses and legumes is more complicated.

Scope

Most widely grown forage, turf and bioenergy species (e.g. tall fescue, perennial ryegrass, switchgrass, alfalfa, white clover) are highly self-incompatible and outcrossing. Compared with inbreeding species, they have a high potential to pass their genes to adjacent plants. A major biosafety concern in these species is pollen-mediated transgene flow. Because human consumption is indirect, risk assessment of transgenic forage, turf and bioenergy species has focused on their environmental or ecological impacts. Although significant progress has been made in genetic modification of these species, commercialization of transgenic cultivars is very limited because of the stringent and costly regulatory requirements. To date, the only transgenic forage crop deregulated in the US is ‘Roundup Ready’ (RR) alfalfa. The approval process for RR alfalfa was complicated, involving several rounds of regulation, deregulation and re-regulation. Nevertheless, commercialization of RR alfalfa is an important step forward in regulatory approval of a perennial outcrossing forage crop. As additional transgenic forage, turf and bioenergy crops are generated and tested, different strategies have been developed to meet regulatory requirements. Recent progress in risk assessment and deregulation of transgenic forage and turf species is summarized and discussed.  相似文献

The dimerisation of Raf kinases involves a central cluster within the kinase domain, the dimer interface (DIF). Yet, the importance of the DIF for the signalling potential of wild-type B-Raf (B-Raf(wt)) and its oncogenic counterparts remains unknown. Here, we show that the DIF plays a pivotal role for the activity of B-Raf(wt) and several of its gain-of-function (g-o-f) mutants. In contrast, the B-Raf(V600E), B-Raf(insT) and B-Raf(G469A) oncoproteins are remarkably resistant to mutations in the DIF. However, compared with B-Raf(wt), B-Raf(V600E) displays extended protomer contacts, increased homodimerisation and incorporation into larger protein complexes. In contrast, B-Raf(wt) and Raf-1(wt) mediated signalling triggered by oncogenic Ras as well as the paradoxical activation of Raf-1 by kinase-inactivated B-Raf require an intact DIF. Surprisingly, the B-Raf DIF is not required for dimerisation between Raf-1 and B-Raf, which was inactivated by the D594A mutation, sorafenib or PLX4720. This suggests that paradoxical MEK/ERK activation represents a two-step mechanism consisting of dimerisation and DIF-dependent transactivation. Our data further implicate the Raf DIF as a potential target against Ras-driven Raf-mediated (paradoxical) ERK activation.  相似文献

Several hummock formation theories exist in the scientific literature, but are rarely mentioned in management publications where only one or two other theories are stressed. Our objectives were to identify and describe the edaphic, climatic, topographic and vegetation characteristics of riparian areas and wetlands in Colorado that do and do not support hummocks, and to classify hummocked sites according to hummock characteristics. 25 natural resource professionals throughout Colorado identified ten sites each that were a mix of hummocked and non-hummocked in proportion to their local abundance, from which 40 hummocked and 40 non-hummocked sites were randomly selected for sampling. Plant functional group canopy cover was determined using Daubenmire cover classes. Soil samples were collected and numerous site characteristics were recorded. Hummock size, shape and density were measured at hummocked sites. Forward model selection and multiple logistic regression were used to determine relationships between site characteristics and odds of hummock occurrence. Mean winter precipitation, mean annual temperature and forb cover were negatively related to odds of hummock occurrence while soil silt content and plant species richness were positively related to odds of hummock occurrence. Cluster analysis resulted in three groups of hummocked sites with differing hummock morphology, vegetation and climatic characteristics. Differential frost heave and plant biomass accumulation were likely involved in hummock formation at sites examined for this study, but conditions expected to occur following cryoexpulsion of clasts were not observed.  相似文献

The persistent problem of worldwide soil erosion has rekindled interest in perennial grain crops. All of our current grain crops are annuals; therefore, developing an array of new perennial grains - grasses, legumes, and others – will require a long-term commitment. Fortunately, many perennial species can be hybridized with related annual crops, allowing us to incorporate genes of domestication much more quickly than did our ancestors who first selected the genes. Some grain crops — including rye, rice, and sorghum — can be hybridized with close perennial relatives to establish new gene pools. Others, such as wheat, oat, maize, soybean, and sunflower, must be hybridized with more distantly related perennial species and genera. Finally, some perennial species with relatively high grain yields — intermediate wheatgrass, wildrye, lymegrass, eastern gamagrass, Indian ricegrass, Illinois bundleflower, Maximilian sunflower, and probably others — are candidates for direct domestication without interspecific hybridization. To ensure diversity in the field and foster further genetic improvement, breeders will need to develop deep gene pools for each crop. Discussions of breeding strategies for perennial grains have concentrated on allocation of photosyn-thetic resources between seeds and vegetative structures. However, perennials will likely be grown in more diverse agro-ecosystems and require arrays of traits very different from those usually addressed by breeders of annuals. The only way to address concerns about the feasibility of perennial grains is to carry out breeding programs with adequate resources on a sufficient time scale. A massive program for breeding perennial grains could be funded by diversion of a relatively small fraction of the world's agricultural research budget.  相似文献

Plasma oxytocin levels were measured serially in 22 women receiving prostaglandin E2 or F intravenously for the induction of labour. Oxytocin was detected in the plasma of 19 of the 22 women; positive levels were found in 60 (43%) of 139 plasma samples, an incidence similar to that in the late first stage of spontaneous labour. Oxytocin was found in the maternal plasma even when the fetus was dead, and in the plasma of two men receiving prostaglandin infusions. This indicates that prostaglandins stimulate the pituitary directly and suggests that this mechanism may play a part in the oxytocic action of infused prostaglandins.  相似文献

Human serum at 5 to 10% (v/v) in tissue culture medium RPMI-1640, inhibits the growth of Cryptococcus neoformans by 80 to 93%. Serum fractionated on molecular sieve columns (Sephadex G-200) yielded an active protein fraction. This fraction at 100 μg protein/ml inhibited the growth of C. neoformans by 54%. When an active G-200 fraction was applied to a dye affinity column (Affi-Gel Blue) the fraction with inhibitory activity was bound by the column and was eluted with 1.4 M NaCl in 0.1 M phosphate buffer (pH 7.4). The bound fraction at 62.5 μg protein/ml inhibited C. neoformans growth by 82%. On native polyacrylamide gel electrophoresis (Nu-PAGE) the bound fraction migrated as a major and a minor band. Under the reducing conditions of sodium dodecyl sulfate (SDS)-PAGE the bound fraction yielded 4 prominent bands with MW ranging from 175 kDa to 45 kDa. Purification of the active Sephadex G-200 peak was achieved using an anion exchange column (DEAE-Sephacel). Protein eluted with 0.1 M NaCl had strong anticryptococcal activity (12.5 μg/ml, 79% inhibition), which in SDS-PAGE migrated as a single band with an approximate MW of 85 kDA. This protein appears important in natural host resistance to C. neoformans and polymorphisms or deficiencies may have epidemiologic and diagnostic relevance. A region between Chelyabinsk and Ekaterinburg in the Southern Urals has been heavily contaminated due to operational and accidental releases from the first Soviet plutonium production facility Mayak. In 1992 and 1993, the German Federal Office for Radiation Protection organized a measuring campaign involving two Russian institutes to assist with the validation of former Soviet measurement data. The results of this measuring campaign are reported here. Environmental samples were collected from areas affected by significant radioactive releases into the Techa river, which started in 1948, and by fallout from the explosion of a fission product storage tank in 1957. Soil, sediment, water, milk and food samples were independently analysed for 90Sr, 137Cs and plutonium by the three institutes involved. This paper presents data on the present levels of environmental radioactivity. The highest contamination of areas accessible to the local population was found in the vicinity of the Techa river around Muslumovo. Activity concentration of floodplain samples reached up to 37 000 Bq ⋅ kg–1 137Cs, 5 600 Bq ⋅ kg–1 90Sr and 9.9 Bq ⋅ kg–1 Pu. Milk and potatoes from private farms in Muslumovo showed low activity in the range from 0.7 Bq ⋅ kg–1 to 25 Bq ⋅ kg–1 90Sr. The results of the three independent measurement teams showed sufficient agreement. One Russian laboratory obtained plutonium activities that exceeded the results of the other laboratories by about 20%. Contrary to the International Chernobyl Project, there was no overestimation of 90Sr activities in the Russian analyses. Therefore, the validity of earlier data sets acquired with same methodology and quality control can be considered a valuable basis for further assessments and for dose reconstruction in epidemiological projects. Cryptococcus neoformans var. neoformans is an opportunistic fungal pathogen, especially in AIDS patients, and is found world-wide. On the other hand, Cryptococcus neoformans var. gatti (CN-g) is restricted to an association with two species of Eucalyptus trees. Alveolar macrophages (AM) constitute the first line of defense to Cryptococcus neoformans and offers some resistance. The inflammatory response to Cryptococcus neoformans with an influx of neutrophils and monocytes affords a second line of defense. Secretion of proinflammatory monokines by human AM is now being defined. The inflammatory phagocytes are efficient in killing Cryptococcus neoformans and offer strong resistance. T and B cell responses to infection, a third line of defense, results in production of lymphokines (IFNg, etc.) and specific antibodies. Enhancement of lymphocyte responses by IL-12 and IL-18 to Cryptococcus neoformans infection appears to be critical. Susceptibility of AIDS patients to Cryptococcus neoformans is associated with low CD4+ T cell counts and likely reduced efficacy of the second line of defense.
