Human and rat peritoneal macrophages and rat Kupffer cells were labelled with [1-14C] arachidonic acid and stimulated with the calcium ionophore A23187. The metabolites formed were separated by high pressure liquid chromatography (HPLC). Human peritoneal macrophages formed especially leukotriene B4, 5-hydroxy-6,8,11,14 eicosatetraenoic acid and small amounts of leukotriene C4 and thromboxane B2, 12-hydroxy-5,8,10 heptadecatrienoic acid and 6-keto-prostaglandin F1 alpha, whereas rat peritoneal macrophages mainly produced cyclooxygenase products and in particular thromboxane B2 and 12-hydroxy-5,8,10 heptadecatrienoic acid. Rat Kupffer cells synthesized mainly cyclooxygenase products such as prostaglandin F2 alpha, prostaglandin D2 and prostaglandin E2. These results indicate that the profile of eicosanoids production by macrophages is dependent both on the species and on the tissue from which the macrophage is derived.  相似文献

A series of heavy chain isotype switch-variant anti-CD3 monoclonal antibodies (mAb) was used to study the proliferation requirements of purified T cells. None of the variant antibodies was able by itself to induce proliferation. In the presence of exogenous interleukin 2 (IL-2) strong mitogenesis was observed upon stimulation with epsilon-anti-CD3, whereas gamma 1, gamma 2b, gamma 2a, and alpha-anti-CD3 failed to induce T cell proliferation. All variant antibodies induced vigorous proliferation in combination with phorbol myristate acetate. Purified T cells, cultured in the presence of epsilon-anti-CD3, in the absence of IL-2, did not express detectable amounts of TAC-antigen (CD25). The binding of the variant antibodies to the CD3 antigen was evaluated in cross-blocking experiments. It was demonstrated that the epsilon-anti-CD3 antibody, in comparison with the other variant mAb, has a relatively low avidity for the CD3 antigen. In modulation experiments, the IgE variant antibody was unable to induce a substantial loss of CD3 antigen. T cell triggering was investigated at the level of Ca2+ mobilization by means of the dye Indo-1. In contrast to the gamma 1, gamma 2b, gamma 2a, and alpha mAb, which induced a rapid and high rise in the free intracellular calcium level, epsilon-anti-CD3 caused a slow and low rise. These studies indicate that the epsilon-anti-CD3 antibody has a low avidity for the CD3 antigen, compared with the other variant mAb, possibly as a result of monovalent binding. Apparently, the avidity and/or valency of CD3 antigen binding not only has a major influence on CD3 modulation and anti-CD3-induced Ca2+ mobilization, but also sets T cell requirements for IL-2 responsiveness.  相似文献

The Escherichia coli UvrABC endonuclease is capable of initiating the repair of a wide variety of DNA damages. To study the binding of the UvrAB complex to the DNA at the site of a lesion we have constructed a synthetic DNA fragment with a defined cis-diamminedichloroplatinum(II) (cis-Pt).GG adduct. The cis-Pt.GG is the major adduct after treatment of DNA with the antitumor agent cisplatin. Binding to the DNA at the site of the defined lesion was studied with DNase I and MPE.Fe(II) hydroxyl radical footprinting. The results indicate that the UvrAB complex binds to the convex side of the kink in the DNA caused by the cis-Pt.GG adduct. Concerted incisions of the damaged strand by the UvrABC endonuclease were at the 8th phosphodiester bond 5' to and at the 4th bond 3' of the adjacent guanines. An additional incision was found at the 15th phosphodiester bond 5' to the damaged site. This extra incision was stimulated by a high concentration of UvrC.  相似文献

Thiophenes are polyacetylene-related heterocyclic metabolites. Some of these compounds are phototoxic, but the bithiophenes occurring inTagetes mainly accumulate in the root where photo-activation is not likely to occur. A cell-free extract from the fungusFusarium oxysporum induced biosynthesis of hydrophilic thiophenes in root cultures and roots of seedlings ofTagetes patula. The thiophenes formed were partially excreted into the culture medium. The transport of glutamate across the membrane of rat-liver mitochondria has been studied.

1. 1. Glutamate can be transported across the mitochondrial membrane in exchange for OH (or together with H+).

2. 2. Intramitochondrial glutamate is not extruded from the mitochondria by addition of aspartate when the mitochondria are preloaded with glutamate.

3. 3. N-Ethylmaleimide is a specific inhibitor of the movement of glutamate across the mitochondrial membrane.

Dynamics in the production of extracellular polymeric substances (EPS) were investigated for the benthic diatoms Cylindrotheca closterium (Ehrenberg) and Nitzschia sp. The effect of growth phase and light:dark conditions were examined using axenic cultures. Two EPS fractions were distinguished. Soluble EPS was recovered from the culture supernatant and represented polysaccharides that were only loosely associated with the cells. Bound EPS was extracted from the cells using warm (30° C) water and was more closely associated with the diatom aggregates. Concentrations of EPS exceeded internal concentrations of sugar throughout growth, indicating that EPS production is important in these organisms. Soluble and bound EPS revealed distinct differences in daily dynamics during the course of growth. Soluble EPS was produced continuously once cultures entered the stationary phase. During the stationary phase, chl a‐normalized EPS production rates equaled 6.4 and 3.4 d ? 1 for C. closterium and Nitzschia sp., respectively. In contrast, production of bound EPS occurred only in the light and was highest during the exponential phase. Up to 90% of the attached EPS that was produced in the light was degraded during the subsequent dark period. The monosaccharide distribution of EPS was constant during the course of the experiment. The soluble EPS consisted of high amounts of galactose and glucuronic acid, relative to rhamnose, glucose, xylose/mannose, and galacturonic acid. In contrast, glucose was the dominant monosaccharide present in the bound EPS. These differences suggest that the production of the two distinct EPS fractions is under different metabolic controls and probably serves different cellular functions.  相似文献

In field studies conducted at the Kongsfjord (Spitsbergen) changes of the irradiance in the atmosphere and the sublittoral zone were monitored from the beginning of June until the end of August 1997, to register the minimum and maximum fluxes of ultraviolet and photosynthetically active radiation and to characterise the underwater light climate. Measurements of photosynthesis in three abundant brown algal species (Alaria esculenta, Laminaria saccharina, Saccorhiza dermatodea) were conducted to test whether their photosynthetic performance reflects changing light climate in accordance with depth. Plants sampled at various depths were exposed to controlled fluence rates of photosynthetically active radiation (400–700 nm), UV-A (320–400 nm) and UV-B (280–320 nm). Changes in photosynthetic performance during the treatments were monitored by measuring variable chlorophyll fluorescence of photosystem II. In each species, the degree of inhibition of photosynthesis was related to the original collection depth, i.e. shallow-water isolates were more resistant than plants from deeper waters. The results show that macroalgae acclimate effectively to increasing irradiance levels for both photosynthetically active and ultraviolet radiation. However, the kinetics of acclimation are different within the different species. It is shown that one important strategy to cope with higher irradiance levels in shallow waters is the capability for a faster recovery from high light stress compared to isolates from deeper waters. A. terreus isolates isolated from some bakery products, corn and rice were found to be able to produce territrems. 90% of theA. terreus isolated from bakery products were able to produce territrem A, with a mean of 0.09 ppm, while 80% ofA. terreus isolates produce territrem B with a mean of 0.24 ppm. On the other hand 31.8% of the isolates ofA. terreus from corn were able to produce territrem A with a mean of 0.44 ppm. This paper illustrates the use of a powerful language, called J, that is ideal for simulating neural networks. The use of J is demonstrated by its application to a gradient descent method for training a multilayer perceptron. It is also shown how the back-propagation algorithm can be easily generalized to multilayer networks without any increase in complexity and that the algorithm can be completely expressed in an array notation which is directly executable through J. J is a general purpose language, which means that its user is given a flexibility not available in neural network simulators or in software packages such as MATLAB. Yet, because of its numerous operators, J allows a very succinct code to be used, leading to a tremendous decrease in development time.  相似文献

The degranulation of mast cells in an allergic response is initiated by the aggregation of high-affinity IgE receptors (Fc epsilon RI) by IgE and antigen. Recently it has been shown that such degranulation can be inhibited by cross-linking Fc epsilon RI and low-affinity IgG receptors (Fc gamma RII) which are also expressed by mast cells. The ability of various monoclonal antibodies to block the degranulation of rat basophil leukaemia (RBL) cells sensitized with IgE antidinitrophenyl (DNP) antibodies has been investigated. Sensitized cells were challenged with immune complexes formed using varying concentrations of antigen, and of both high- and low-valency antigen. It is reported here that rat IgG1 antibodies, which are associated in the rat with a Th1-type response, act as highly effective blocking antibodies over a wide concentration range. Rat IgG2a antibodies, which are associated with a Th2-type response, were able only to inhibit degranulation when immune complexes were formed with very low concentrations of high-valency antigen (DNP32-HSA). Under these conditions, some inhibitory activity was seen with high-affinity murine IgA anti-DNP but not with low-affinity rat IgG2b anti-DNP antibody-containing immune complexes. In addition to this inhibitory activity, IgG2a antibodies were shown to be capable of inducing degranulation of cells via unoccupied Fc epsilon RI. These results demonstrate that blocking activity may arise via both inhibitory receptors and by masking of antigen.  相似文献
