Background
Emerging evidence suggests that high density lipoprotein (HDL) may modulate glucose metabolism through multiple mechanisms including pancreatic insulin secretion as well as insulin-independent glucose uptake into muscle. We hypothesized that HDL may also increase skeletal muscle insulin sensitivity via cholesterol removal and anti-inflammatory actions in macrophages associated with excess adiposity and ectopic lipid deposition.Methods
Human primary and THP-1 macrophages were treated with vehicle (PBS) or acetylated low density lipoprotein (acLDL) with or without HDL for 18 hours. Treatments were then removed, and macrophages were incubated with fresh media for 4 hours. This conditioned media was then applied to primary human skeletal myotubes derived from vastus lateralis biopsies taken from patients with type 2 diabetes to examine insulin-stimulated glucose uptake.Results
Conditioned media from acLDL-treated primary and THP-1 macrophages reduced insulin-stimulated glucose uptake in primary human skeletal myotubes compared with vehicle (primary macrophages, 168±21% of basal uptake to 104±19%; THP-1 macrophages, 142±8% of basal uptake to 108±6%; P<0.05). This was restored by co-treatment of macrophages with HDL. While acLDL increased total intracellular cholesterol content, phosphorylation of c-jun N-terminal kinase and secretion of pro- and anti-inflammatory cytokines from macrophages, none were altered by co-incubation with HDL. Insulin-stimulated Akt phosphorylation in human skeletal myotubes exposed to conditioned media was unaltered by either treatment condition.Conclusion
Inhibition of insulin-stimulated glucose uptake in primary human skeletal myotubes by conditioned media from macrophages pre-incubated with acLDL was restored by co-treatment with HDL. Cox17, a copper chaperone for cytochrome-c oxidase, is an essential and highly conserved protein in eukaryotic organisms. Yeast and mammalian Cox17 share six conserved cysteine residues, which are involved in complex redox reactions as well as in metal binding and transfer. Mammalian Cox17 exists in three oxidative states, each characterized by distinct metal-binding properties: fully reduced mammalian Cox17(0S-S) binds co-operatively to four Cu+; Cox17(2S-S), with two disulfide bridges, binds to one of either Cu+ or Zn2+; and Cox17(3S-S), with three disulfide bridges, does not bind to any metal ions. The E(m) (midpoint redox potential) values for two redox couples of Cox17, Cox17(3S-S)<-->Cox17(2S-S) (E(m1)) and Cox17(2S-S)<-->Cox17(0S-S) (E(m2)), were determined to be -197 mV and -340 mV respectively. The data indicate that an equilibrium exists in the cytosol between Cox17(0S-S) and Cox17(2S-S), which is slightly shifted towards Cox17(0S-S). In the IMS (mitochondrial intermembrane space), the equilibrium is shifted towards Cox17(2S-S), enabling retention of Cox17(2S-S) in the IMS and leading to the formation of a biologically competent form of the Cox17 protein, Cox17(2S-S), capable of copper transfer to the copper chaperone Sco1. XAS (X-ray absorption spectroscopy) determined that Cu4Cox17 contains a Cu4S6-type copper-thiolate cluster, which may provide safe storage of an excess of copper ions. 相似文献

Resource availability is a limiting factor influencing the distribution and composition of faunal communities. Globally, hollow bearing trees are a resource required by wildlife at all trophic levels, and are used for a diverse range of ecological functions. In the northern hemisphere avian species act as primary hollow excavators, whereas the southern hemisphere must rely on complex interactions between stochastic events, and eventual decay. Hollow formation is therefore a slow process in the southern hemisphere. In contrast, hollow loss is quite rapid and influenced greatly by anthropogenic impacts. To identify the ecological characteristics driving hollows over an urban to forest gradient as a resource for the powerful owl (Ninox strenua) and its prey we used presence‐only modelling. The potential for an area to support tree hollows suitable for powerful owls and their prey was linked to the density of ephemeral rivers, land cover, tree cover and distance from riparian vegetation. The potential for large hollows throughout the landscape, suitable for the powerful owl, was also influenced by density of permanent rivers. Potential habitat for tree hollows, capable of supporting powerful owls and their prey was greatest in forested environments, declining with increased urbanization. However the urban region still supported some smaller tree hollows suitable for arboreal marsupials. Managing for urban dwelling species, is not as simple as retaining old hollow producing trees or providing alternate nesting structures. A simple and inexpensive system for the generation of fertile, transgenic maize plants has been developed. Cells from embryogenic maize suspension cultures were transformed using silicon carbide whiskers to deliver plasmid DNA carrying the bacterial bar and uidA (gus) genes. Transformed cells were selected on medium containing the herbicide bialaphos. Integration of the bar gene and activity of the enzyme phosphinothricin acetyl transferase (PAT) were confirmed in all bialaphos-resistant callus lines analysed. Fertile transgenic maize plants were regenerated. Herbicide spraying of progeny plants revealed that the bar gene was transmitted in a Mendelian fashion. 相似文献

Companion animals are increasingly being recognized by society as beneficial to our health and considered by many owners as authentic and affectional family members. Human relationship theories help us to understand the emotional and supportive aspect of the human– companion animal bond. This study uses attachment theory, social support theory, and the concept of the hierarchical nature of attachment relationships to further understand and measure human–animal attachment. In study 1,161 university-student pet owners completed a modified multidimensional scale of perceived social support (MSPSS) that included pets as a source of support, and we pre-tested a 60-item pet attachment measure. Results showed that students perceived their pets as distinctive sources of social support, at similar levels to their significant others, family, and friends. Principal components analysis of the 60-item measure reduced it to 31 items, and revealed four pet attachment components: (a) Proximity maintenance and interaction, (b) Emotional attachment behaviors, (c) Emotional support given and received, and (d) Emotional and monetary value. The scale was named the Emotional and Supportive Attachment to Companion Animals Scale (ESACA) (Cronbach’s α = 0.96). In study 2, 83 university students completed an attachment hierarchy scale and the ESACA. Companion animals were included in pet owners’ attachment hierarchies and ranked higher than siblings but lower than romantic partners, parents, and close friends. Those who indicated higher attachment to their companion animals ranked them higher in their attachment hierarchy than those less attached. This study supports and extends previous research that has used aspects of attachment theory and social support theory when exploring the human–animal bond. Many companion animal owners perceive their pets as additional sources of emotional support, fulfilling the four features of an attachment relationship and including them in their hierarchy of important attachment relationships. 相似文献

Relationships between key phytoplankton attributes including Chl a-specific light absorption, pigment composition and concentration, photosynthesis, primary production and community structure
were studied in two open shallow nutrient-poor coastal systems receiving similar amounts of sewage water. Both systems were
significantly nitrogen limited. However, differences in wastewater treatment (primary vs secondary) and sewage dilution (50%)
between the two systems caused a greater difference between systems than locally around the outflows. For both systems, water
at the outlet had significantly lower water transparency caused by a 20% higher absorption by coloured dissolved organic matter.
Nutrient concentrations were also elevated, gradually decreasing with distance north (governing current) of the outflows,
causing higher abundance of nano-sized phytoplankton, higher content of carotenoid pigments, 20–50% higher Chl a-specific absorption coefficients and higher photosynthetic capacity. Although maximum rates of Chl a-normalised photosynthesis were strongly related to nitrate availability, no effects were found on the derived areal primary
production or algal biomass suggesting that photosynthetic and optical parameters are more sensitive indicators of nutrient
enrichment than biomass or productivity.

From data collected via a large international collaborative study, we have constructed a growth chart for patients with molecularly confirmed congenital spondylo-epiphyseal dysplasia (SEDC) and other COL2A1 related dysplasias. The growth chart is based on longitudinal height measurements of 79 patients with glycine substitutions in the triple-helical domain of COL2A1. In addition, measurements of 27 patients with other molecular defects, such as arginine to cysteine substitutions, splice mutations, and mutations in the C-terminal propeptide have been plotted on the chart. Height of the patients progressively deviate from that of normal children: compared to normal WHO charts, the mean length/height is -2.6 SD at birth, -4.2 SD at 5 years, and -5.8 SD in adulthood. The mean adult height (male and female combined) of patients with glycine substitutions in the triple-helical region is 138.2?cm but there is a large variation. Patients with glycine to cysteine substitutions tend to cluster within the upper part of the chart, while patients with glycine to serine or valine substitutions are situated between +1 SD and -1 SD. Patients with carboxy-terminal glycine substitutions tend to be shorter than patients with amino-terminal substitutions, while patients with splice mutations are relatively tall. However, there are exceptions and specific mutations can have a strong or a relatively mild negative effect on growth. The observation of significant difference in adult height between affected members of the same family indicates that height remains a multifactorial trait even in the presence of a mutation with a strong dominant effect. 相似文献

The diversity in the Plasmodium falciparum genome can be used to explore parasite population dynamics, with practical applications to malaria control. The ability to identify the geographic origin and trace the migratory patterns of parasites with clinically important phenotypes such as drug resistance is particularly relevant. With increasing single-nucleotide polymorphism (SNP) discovery from ongoing Plasmodium genome sequencing projects, a demand for high SNP and sample throughput genotyping platforms for large-scale population genetic studies is required. Low parasitaemias and multiple clone infections present a number of challenges to genotyping P. falciparum. We addressed some of these issues using a custom 384-SNP Illumina GoldenGate assay on P. falciparum DNA from laboratory clones (long-term cultured adapted parasite clones), short-term cultured parasite isolates and clinical (non-cultured isolates) samples from East and West Africa, Southeast Asia and Oceania. Eighty percent of the SNPs (n = 306) produced reliable genotype calls on samples containing as little as 2 ng of total genomic DNA and on whole genome amplified DNA. Analysis of artificial mixtures of laboratory clones demonstrated high genotype calling specificity and moderate sensitivity to call minor frequency alleles. Clear resolution of geographically distinct populations was demonstrated using Principal Components Analysis (PCA), and global patterns of population genetic diversity were consistent with previous reports.
