Given the observed heterogeneity in fire severity produced within wildfires, we asked to what extent this heterogeneity might affect post-fire regeneration. For this purpose, we studied the post-fire dynamics of Pinus halepensis (Aleppo pine) in the eastern Iberian Peninsula. Sampling was stratified on the basis of fire severity. We defined three fire severity classes based on the degree of consumption of the pine canopy. The results suggested that there is no clear relationship between seedling density and fire severity; however, mortality was lower and growth (height, shoot biomass and root biomass) was higher in the high severity class. These results can be explained by soil processes: Sites in the high fire severity class may have sustained higher fire intensities, resulting in higher soil organic matter mineralisation and higher ash deposition, and thus in higher post-fire soil fertility. This higher fertility would produce faster growth in pine seedlings. Independent of the severity class, seedling mortality was higher in quadrats (50 × 50 cm) with higher cover of the perennial grass Brachypodium retusum (Poaceae), suggesting a possible competitive effect. For all plots in all 3 severity classes, spatial analysis suggests an aggregate seedlings pattern, but with independence from the position of the adult (source) trees. Scytonemin, the yellow-brown pigment of cyanobacterial (blue-green algal) extracellular sheaths, was found in species thriving in habitats exposed to intense solar radiation. Scytonemin occurred predominantly in sheaths of the outermost parts or top layers of cyanobacterial mats, crusts, or colonies. Scytonemin appears to be a single compound identified in more than 30 species of cyanobacteria from cultures and natural populations. It is lipid soluble and has a prominent absorption maximum in the near-ultraviolet region of the spectrum (384 nm in acetone; ca. 370 nm in vivo) with a long tail extending to the infrared region. Microspectrophotometric measurements of the transmittance of pigmented sheaths and the quenching of ultraviolet excitation of phycocyanin fluorescence demonstrate that the pigment was effective in shielding the cells from incoming near-ultraviolet-blue radiation, but not from green or red light. High light intensity (between 99 and 250 μmol photon · m?2· S?1, depending on species) promoted the synthesis of scytonemin in cultures of cyanobacteria. In cultures, high light intensity caused reduction in the specific content of Chl a and phycobilins, increase in the ratio of total carotenoids to Chl a, and scytonemin increase. UV-A (320–400 nm) radiation was very effective in eliciting scytonemin synthesis. Scytonemin production was physiological and not due to a mere photochemical conversion. These results strongly suggest that scytonemin production constitutes an adaptive strategy of photoprotection against short-wavelength solar irradiance.  相似文献

G-protein coupled receptors, the largest family of proteins in the human genome, are involved in many complex signal transduction pathways, typically activated by orthosteric ligand binding and subject to allosteric modulation. Dopaminergic receptors, belonging to the class A family of G-protein coupled receptors, are known to be modulated by sodium ions from an allosteric binding site, although the details of sodium effects on the receptor have not yet been described. In an effort to understand these effects, we performed microsecond scale all-atom molecular dynamics simulations on the dopaminergic D2 receptor, finding that sodium ions enter the receptor from the extracellular side and bind at a deep allosteric site (Asp2.50). Remarkably, the presence of a sodium ion at this allosteric site induces a conformational change of the rotamer toggle switch Trp6.48 which locks in a conformation identical to the one found in the partially inactive state of the crystallized human β2 adrenergic receptor. This study provides detailed quantitative information about binding of sodium ions in the D2 receptor and reports a possibly important sodium-induced conformational change for modulation of D2 receptor function.  相似文献

The detection of genes that show similar profiles under different experimental conditions is often an initial step in inferring the biological significance of such genes. Visualization tools are used to identify genes with similar profiles in microarray studies. Given the large number of genes recorded in microarray experiments, gene expression data are generally displayed on a low dimensional plot, based on linear methods. However, microarray data show nonlinearity, due to high-order terms of interaction between genes, so alternative approaches, such as kernel methods, may be more appropriate. We introduce a technique that combines kernel principal component analysis (KPCA) and Biplot to visualize gene expression profiles. Our approach relies on the singular value decomposition of the input matrix and incorporates an additional step that involves KPCA. The main properties of our method are the extraction of nonlinear features and the preservation of the input variables (genes) in the output display. We apply this algorithm to colon tumor, leukemia and lymphoma datasets. Our approach reveals the underlying structure of the gene expression profiles and provides a more intuitive understanding of the gene and sample association.  相似文献

The shoulder complex has a larger range of motion (ROM) than any other joint complex in the human body, leaving it prone to numerous injuries. Objective kinematic analysis could yield useful functional insights that may assist clinical practice. Non-invasive optoelectronic motion analysis techniques have been used to assess the shoulders of five healthy subjects performing ROM tasks and 10 functional tasks of daily living. The four most demanding tasks - touching the side and back of the head, brushing the opposite side of the head, lifting an object to shoulder height and lifting an object to head height, required 78%, 60%, 61% and 71%, respectively, of the glenohumeral elevation necessary for full abduction in the scapular plane for the 10 shoulders. This has implications for clinical practice where maximum arm elevation is commonly used to determine a patient's ability to return to work and other everyday activities.  相似文献

The sucrose operon from pUR400, a 78-kbp conjugative Salmonella plasmid, was cloned in Escherichia coli K12. The operon was located in a 5.7-kbp SalI restriction fragment and was subcloned, in each of two possible orientations, using the expression vector pUC18. The insert DNA was restriction mapped and duplicate restriction sites in the insert and in the polylinker of the vector were used to create various deletions promoter distal in the operon sequence. Additional deletions were made with the restriction exonuclease Bal31. Cells containing hybrid plasmids with specified deletions lacked the ability to transport sucrose or were constitutive for hydrolase and/or uptake activities. The scrA (enzyme IIScr) and scrR (regulatory) genes resided within 2900-bp SmaI-SalI DNA fragment and were assigned the order scrB, scrA, scrR. An amplified sucrose-inducible gene product, Mr 68,000, was detected only in the membrane fraction from recombinant cells that contained plasmid with the intact operon sequence. This protein represented 11% of the total membrane protein and was resistant to extraction with 0.5 M sodium chloride, 2% Triton X-100, and 0.5% sodium deoxycholate. The protein did not appear to be the product of either the scrA, scrB, or scrR gene and may therefore represent a previously unidentified membrane-bound sucrose protein. A new gene, scrC, is proposed. In addition, the cloned 5.7-kbp SalI and 2.5-kbp SmaI-SalI DNA fragments failed to hybridize to chromosomal DNA from Bacillus subtilis, Streptococcus lactis, Streptococcus mutans, and Lactobacillus acidophilus as well as to DNA from a sucrose plasmid from Salmonella tennessee. However, the probes showed weak homology with a 20-kbp EcoRI restriction fragment from Klebsiella pneumoniae.  相似文献

Saissetia oleae (Olivier) (Hemiptera: Coccidae) populations were studied and compared in citrus (Citrus spp.) and olive (Olea europaea L.) groves to determine the number of generations, crawler emergence periods and changes in population density during the year. Ten citrus and four olive groves were sampled regularly between March 2003 and December 2005 in eastern Spain, covering an area of 10,000 km2. Each sample consisted of 16 branches and 64 leaves. Saissetia oleae populations presented a similar trend in both crops during the three years of study. Populations peaked in July, when crawlers emerged after the egg-laying period, and decreased during several months due to mortality of first instars in summer. A second crawler emergence period, with lower numbers and more variability from year to year, occurred between October and March. Populations did not increase during this period, probably because most eggs and crawlers perished during the winter and also because females that gave rise to this fall-winter generation were half as big and fecund as spring females. No differences were found between the size of mature females that had developed on citrus and on olives during the spring. Considering this population pattern, the best seasonal period to apply pesticides to control S. oleae would be at the end of July, when populations are synchronous, all crawlers have already emerged, and first instars predominate.  相似文献
