Restriction mapping and sequencing have shown that humans have substantially lower levels of mitochondrial genome diversity (d) than chimpanzees. In contrast, humans have substantially higher levels of heterozygosity (H) at protein-coding loci, suggesting a higher level of diversity in the nuclear genome. To investigate the discrepancy further, we sequenced a segment of the mitochondrial genome control region (CR) from 49 chimpanzees. The majority of these were from the Pan troglodytes versus subspecies, which was underrepresented in previous studies. We also estimated the average heterozygosity at 60 short tandem repeat (STR) loci in both species. For a total sample of 115 chimpanzees, d = 0.075 +/0 0.037, compared to 0.020 +/- 0.011 for a sample of 1,554 humans. The heterozygosity of human STR loci is significantly higher than that of chimpanzees. Thus, the higher level of nuclear genome diversity relative to mitochondrial genome diversity in humans is not restricted to protein-coding loci. It seems that humans, not chimpanzees, have an unusual d/H ratio, since the ratio in chimpanzees is similar to that in other catarrhines. This discrepancy in the relative levels of nuclear and mitochondrial genome diversity in the two species cannot be explained by differences in mutation rate. However, it may result from a combination of factors such as a difference in the extent of sex ratio disparity, the greater effect of population subdivision on mitochondrial than on nuclear genome diversity, a difference in the relative levels of male and female migration among subpopulations, diversifying selection acting to increase variation in the nuclear genome, and/or directional selection acting to reduce variation in the mitochondrial genome.   相似文献

Many species show large variation in lifetime reproductive success (LRS), with a few individuals producing the majority of offspring. This variation can be explained by factors related to individuals (fixed heterogeneity) and stochastic differences in survival and reproduction (dynamic heterogeneity). In this study, we study the relative effects of these processes on the LRS of a Dutch Kestrel population, using three different methods. First, we extended neutral simulations by simulating LRS distributions of populations consisting of groups with increasingly different population parameters. Decomposition of total LRS variance into contributions from fixed and dynamic heterogeneity revealed that the proportion of fixed heterogeneity is probably lower than 10% of the total variance. Secondly, we used sensitivities of the mean and variance in LRS to each parameter to analytically show that it is impossible to get equal contributions of fixed and dynamic heterogeneity when only one parameter differs between groups. Finally, we computed the LRS probability distribution to show that even when all individuals have identical survival and reproduction rates, the variance in LRS is large (females: 27.52, males: 12.99). Although each method has its limitations, they all lead to the conclusion that the majority of the variation in kestrel LRS is caused by dynamic heterogeneity. This large effect of dynamic heterogeneity on LRS is similar to results for other species and contributes to the evidence that in most species the majority of individual variation in LRS is due to dynamic heterogeneity.  相似文献

Summary We report on 50 couples with reproductive loss who did not have any detectable chromosome abnormality. A history of a previous child with multiple congenital abnormalities may be significant in identifying couples with a structural rearrangement. Only by studying more families can this hypothesis be tested. Studies of abortus tissue reveal a high percentage of chromosome abnormalities but a very low incidence of unbalanced translocations. Cytogenetic studies are indicated in a couple which has a past history of spontaneous abortions and a previous child with multiple congenital anomalies.  相似文献

Enzyme technology is explored on wool fibers to prevent shrinkage and consolidation behavior during washing of woolen bed coverings using normal household machine conditions. Enzyme modification of wool fibers after two different pretreatments has been realized on industrial scale. Enlarged proteolytic enzyme by chemical modification was applied successfully to prevent substantial fiber strength loss. Felt-ball analysis of the fibers as obtained from this industrial process showed substantial improvement in felting resistance. Further processing of these enzyme-modified fibers and finally integration in bed covering quilts have been executed successfully on industrial production lines. The observed fiber losses during processing were in the range of 4.5–6% which is comparable with that of nonmodified fibers. The machine-washability of these produced bed covering quilts was tested in a household washing machine using both wool and normal wash programs applied at different temperatures. It appeared that, contrary to the good washing results in terms of shrinkage and consolidation resistance using the wool program at moderate temperatures, this resistance is marginal when washed with the normal washing program with higher mechanical agitation level or with the wool program at elevated temperature. This result was different from that obtained with woolen fabrics and explained by the less-structured organisation of fibers within a fleece.  相似文献

Partial anomalous pulmonary venous connection (PAPVC) is an extremely rare congenital condition where one or more of the pulmonary veins are connected to the venous circulation. Although initially suspected with unexplained right ventricular enlargement on transthoracic echocardiography (TTE), cardiac MRI is able to delineate the anatomical variant. Human neutrophils aggregate and release mediators of inflammation, such as active oxygen species and lysosomal enzymes, when exposed to the chemoattractant, fMet-Leu-Phe, or the tumor promotor, phorbol myristate acetate. In order to 'stage' events which may lead to such neutrophil responses, we determined the temporal relationship between stimulus-induced changes in the endogenous phospholipids phosphatidylinositol (PI) and phosphatidic acid, the mobilization of calcium, and the onset of aggregation and generation of superoxide anion during the initial 2 min of cell activation. Within 5 s after addition of fMet-Leu-Phe (10(-7) M) neutrophils accumulated phosphatidic acid and the levels of PI decreased, as determined by two-dimensional thin-layer chromatography and phosphorus determinations. By 5 s, phosphatidic acid levels rose approximately 3.5-fold and at 15 s the loss of PI exceeded the quantity of phosphatidic acid generated. In response to phorbol myristate acetate (1 microgram/ml), however, changes in PI or phosphatidic acid were not observed until after 60 s. Accumulation of phosphatidic acid in fMet-Leu-Phe-stimulated cells was not inhibited by chelation of extracellular calcium. Neutrophils exposed to either fMet-Leu-Phe or phorbol myristate acetate also showed rapid decrements in fluorescence of cell-associated chlorotetracycline (used as an indirect probe of mobilization of intracellular membrane-associated calcium) and took up 45Ca2+ from the extracellular medium (under 60 s). The results indicate that changes in calcium mobilization, together with the alterations in phospholipid metabolism (under 5 s) anteceded aggregation and the generation of O2-. (10-15 s) induced by fMet-Leu-Phe. In contrast, when neutrophils were exposed to phorbol myristate acetate, changes in PI and phosphatidic acid (over 60 s) were observed after the mobilization of calcium (under 5 s) and the onset of O2-. generation and aggregation (30-35 s).  相似文献
