The great challenges for researchers working in the field of vaccinology are optimizing DNA vaccines for use in humans or large animals and creating effective single-dose vaccines using appropriated controlled delivery systems. Plasmid DNA encoding the heat-shock protein 65 (hsp65) (DNAhsp65) has been shown to induce protective and therapeutic immune responses in a murine model of tuberculosis (TB). Despite the success of naked DNAhsp65-based vaccine to protect mice against TB, it requires multiple doses of high amounts of DNA for effective immunization. In order to optimize this DNA vaccine and simplify the vaccination schedule, we coencapsulated DNAhsp65 and the adjuvant trehalose dimycolate (TDM) into biodegradable poly (DL-lactide-co-glycolide) (PLGA) microspheres for a single dose administration. Moreover, a single-shot prime-boost vaccine formulation based on a mixture of two different PLGA microspheres, presenting faster and slower release of, respectively, DNAhsp65 and the recombinant hsp65 protein was also developed. These formulations were tested in mice as well as in guinea pigs by comparison with the efficacy and toxicity induced by the naked DNA preparation or BCG. Many species show large variation in lifetime reproductive success (LRS), with a few individuals producing the majority of offspring. This variation can be explained by factors related to individuals (fixed heterogeneity) and stochastic differences in survival and reproduction (dynamic heterogeneity). In this study, we study the relative effects of these processes on the LRS of a Dutch Kestrel population, using three different methods. First, we extended neutral simulations by simulating LRS distributions of populations consisting of groups with increasingly different population parameters. Decomposition of total LRS variance into contributions from fixed and dynamic heterogeneity revealed that the proportion of fixed heterogeneity is probably lower than 10% of the total variance. Secondly, we used sensitivities of the mean and variance in LRS to each parameter to analytically show that it is impossible to get equal contributions of fixed and dynamic heterogeneity when only one parameter differs between groups. Finally, we computed the LRS probability distribution to show that even when all individuals have identical survival and reproduction rates, the variance in LRS is large (females: 27.52, males: 12.99). Although each method has its limitations, they all lead to the conclusion that the majority of the variation in kestrel LRS is caused by dynamic heterogeneity. This large effect of dynamic heterogeneity on LRS is similar to results for other species and contributes to the evidence that in most species the majority of individual variation in LRS is due to dynamic heterogeneity. 相似文献

As a reference for ongoing studies reconstructing past vegetation, climate and environment, pollen spectra in tundra peat profiles from Svalbard, were investigated. The base of tundra peat cores collected from Ny Ålesund, Stuphallet, Blomstrand and Isdammen has been 14C dated to 350–490 BP, 5710 BP, 4670 BP and 700–900 BP, respectively. The Stuphallet and Blomstrand (Brøggerhalvøya) peat profiles were composed of a peat developed in a nutrient enriched and wet tundra environment of steep birdcliffs. Pollen concentrations were low, Brassicaceae pollen dominated the whole profile. In contrast, the Ny Ålesund and Isdammen profiles contained high pollen concentrations and suggest a nutrient-poor, dry tundra environment. Pollen of the polar willow, Salix polaris, occurred commonly throughout all four peat profiles. In the relatively high resolution (10 years per peat core sample) analysis of the Ny Ålesund core, starting before or at the beginning of the Little Ice Age (LIA, 16th-mid 19th century), dominance of Saxifraga oppositifolia indicates a cold and dry climate, followed by a decline of Saxifraga oppositifolia and gradual increase of Salix polaris after the LIA, which indicates a moist and milder climate. 相似文献

Cathelicidins are among the best characterized antimicrobial peptides and have been shown to have an important role in mammalian innate immunity. We recently isolated a novel mature cathelicidin peptide (codCath) from Atlantic cod and in the present study we functionally characterized codCath. The peptide demonstrated salt sensitivity with abrogation of activity at physiological salt concentrations. In low ionic strength medium we found activity against marine and non-marine Gram-negative bacteria with an average MIC of 10 μM, weak activity against a Gram-positive bacterium (MIC 80 μM), and pronounced antifungal activity (MIC 2.5 μM). The results suggest the kinetics and mode of action of codCath to be fast killing accompanied by pronounced cell lysis. Extracellular products (ECPs) of three marine bacteria caused breakdown of the peptide into smaller fragments and the cleaved peptide lost its antibacterial activity. Proteolysis of the peptide on the other hand was abolished by prior heat-treatment of the ECPs, suggesting a protease involvement. We observed no cytotoxicity of the peptide in fish cells up to a concentration of 40 μM and the selectivity of activity was confirmed with bacterial and mammalian membrane mimetics. We conclude that the potent broad-spectrum activity of codCath hints at a role of the peptide in cod immune defense. 相似文献

UV-B radiation currently represents c. 1.5% of incoming solar radiation. However, significant changes are known to have occurred in the amount of incoming radiation both on recent and on geological timescales. Until now it has not been possible to reconstruct a detailed measure of UV-B radiation beyond c. 150 yr ago. Here, we studied the suitability of fossil Pinus spp. pollen to record variations in UV-B flux through time. In view of the large size of the grain and its long fossil history, we hypothesized that this grain could provide a good proxy for recording past variations in UV-B flux. Two key objectives were addressed: to determine whether there was, similar to other studied species, a clear relationship between UV-B-absorbing compounds in the sporopollenin of extant pollen and the magnitude of UV-B radiation to which it had been exposed; and to determine whether these compounds could be extracted from a small enough sample size of fossil pollen to make reconstruction of a continuous record through time a realistic prospect. Preliminary results indicate the excellent potential of this species for providing a quantitative record of UV-B through time. Extracellular vesicles (EVs), including ‘microvesicles’ and ‘exosomes’, are highly abundant in bodily fluids. Recent years have witnessed a tremendous increase in interest in EVs. EVs have been shown to play important roles in various physiological and pathological processes, including coagulation, immune responses, and cancer. In addition, EVs have potential as therapeutic agents, for instance as drug delivery vehicles or as regenerative medicine. Because of their small size (50 to 1,000 nm) accurate quantification and size profiling of EVs is technically challenging.This protocol describes how tunable resistive pulse sensing (tRPS) technology, using the qNano system, can be used to determine the concentration and size of EVs. The method, which relies on the detection of EVs upon their transfer through a nano sized pore, is relatively fast, suffices the use of small sample volumes and does not require the purification and concentration of EVs. Next to the regular operation protocol an alternative approach is described using samples spiked with polystyrene beads of known size and concentration. This real-time calibration technique can be used to overcome technical hurdles encountered when measuring EVs directly in biological fluids. 相似文献

Summary We report on 50 couples with reproductive loss who did not have any detectable chromosome abnormality. A history of a previous child with multiple congenital abnormalities may be significant in identifying couples with a structural rearrangement. Only by studying more families can this hypothesis be tested. Studies of abortus tissue reveal a high percentage of chromosome abnormalities but a very low incidence of unbalanced translocations. Cytogenetic studies are indicated in a couple which has a past history of spontaneous abortions and a previous child with multiple congenital anomalies. 相似文献
